5. The single-test concept
● The VIDAS® principle is based on the interaction of two elements: the coated SPR®
receptacle, containing antigens or antibodies, and the Strip, made up of a series of wells
containing the correct amount of reagent necessary for the test.
● All reactions occur within the SPR in two key phases:
immunological reaction
enzymatic reaction
●The entire operation is fully automated: from incubation, to washing and final reading.
● Incubation time and the number of washing cycles are optimized to ensure peak
performance. are performed automatically by the instrument.The reaction medium is
cycled in and out of the SPR several times.Each step is followed by a wash cycles
which eliminates unbound components
THE STRIP: The strip consist of 10 wells covered with a labelled foil sealThe label
comprises a bar code which mainly indicates the assay code, kit lot number and
expiration date.
6. QUALITY CONTROL :One positive and negative control are also
included.These controls must be performed immediately after opening a new
kit to ensure that reagent performance has not been altered.Each calibration
must also be checked using these controls.
The foil of the first well is perforated to facilitate the introduction of the sample.
● The last well of each strip is a cuvette in which the fluorometric reading is
performed.
The wells in the center section contain the various reagents required for the assay.
During the final detection step,the substrate( 4 methyl umbelliferyl phosphate) is
cycled in and out of the SPR. ● The conjugate enzyme catalyzes the hydrolysis of
this substrate into a fluorescent product( 4 methyl umbelliferone) ● The
fluorescence is measured at 450 nm. ● The intensity of the fluorescence is
proportional to concentration of Ab present in the sample.
PROCEDURE
Remove the required reagents from the refrigerator and allow them to come to RT for
30’.
Use the necessary test strip and test SPR for samples ,controls or standards to be tested
Load the strip, SPR and sample

PRINCIPLE
● Chemiluminescent microparticle immunoassay is the emission of light
when an electrons returns from an excited or higher energy level to
lower energy level.
● When used in combination with immunoassay techonology, the light
produced by the reaction indicates the amount of analyte in a sample
● The amount of signal is directly propotional to the amount of analyte
pesent in the sample.
