The present study was undertaken to investigate the protective effect and possible mechanism of
alcoholic (AlE) and aqueous extract (AqE) from Commelina benghalensis root (CB) on EtOH-induced hepatic
injury in Wistar rat. Hepatotoxic parameters studied in vivo include serum transaminases (AST, and ALT),
ALP, bilirubin, protein, lipid profile (Cholesterol, triglyceride, VLDL and HDL) and level of antioxidants
together with histopathological examination. Liv 52® was used as a reference hepatoprotective agent
(5ml/kg-1b.w.). AlE and AqE (200 mg/kg-1b.w.) on oral administration decreased the level of AST, ALP, ALT,
bilirubin, cholesterol, triglyceride, VLDL, MDA and increased the level of protein, HDL and antioxidants (SOD,
GSH and CAT) in rats being treated with ethanol (EtOH). Pentobarbitone -induced sleeping time study was

Nelumbo nucifera (family Nymphaeaceae) are free floating plants.The methanolic extract of Nelumbo nucifera leaves was obtained by soxhlet extraction apparatus. The extract was subjected to preliminary phytochemical screening by using standard procedures.The toxicity studies and dose fixation were carried out by using OECD 425 guideline. According to OECD 425 guideline toxicity study no toxic symptoms were observed up to dose 2000 mg/kg.The anti diabetic effect of Nelumbo nucifera leaf methanolic extract given in streptozotocin induced diabetic rats. Oral administration of methanolic extract for 15 days in diabetic mice exhibits highly significant (P < 0.01) antidiabetic activity and also alters the body weight significantly . The data were analyzed using analysis of variance followed by Dunnett's test.The observations confirm that methanolic extract of NELUMBO NUCIFERA leaf and stem has antidiabetic activity due to presence of alkaloids,aminoacids, saponins, glycosides, triterpenoid, vitamins etc There is a need of further investigation to isolate and identify the principle chemical constituents for its anti diabetic property.

This study evaluated the immunostimulatory and antioxidant properties of Phoenix dactylifera, commonly known as dates. Mice were injected with various concentrations of a Phoenix dactylifera extract. Results showed that the extract significantly increased phagocytic activity and reduced the half-life of carbon in the blood, indicating enhanced function of the reticuloendothelial system. The extract also significantly increased levels of the antioxidant glutathione in the liver. The concentration of 50 mg/kg produced the highest effects on phagocytosis and glutathione. The study aimed to examine the protective effects of taxifolin on cisplatin-induced kidney damage in rats. Rats were divided into three groups: a healthy control group, a cisplatin group, and a taxifolin-cisplatin group. The cisplatin group was given cisplatin only, while the taxifolin-cisplatin group was given both taxifolin and cisplatin. Low beneficial effects of short term antidiabetic diet treatment in streptozo...iosrphr_editor
Oxidative stress is currently suggested to play a role in the pathogenesis of Diabetes mellitus. The role of dietary management in diabetes mellitus is to provide a proper balance of total nutrients while meeting the special dietary needs of the patient. The present study was designated to evaluate the effect of special antidiabetic diet treatment upon oxidative stress parameters in the initial stages of the development of diabetes. Male Wistar strain rats were used as an experimental model, divided into five groups. A significant decrease in superoxide dismutase and total glutathione activities were observed in the liver of diabetic rats when compared with control animals. The plasma level of aminotransferases, creatine kinase, lactate dehydrogenase and urea were significantly increased after induction of diabetes, in all groups under treatment. In contrast, rats fed special diet food, have shown slight different, but not significant changes. The findings of the present study suggest that special diet formula useful for prevention of progressive hyperglycaemia in age induced diabetes in dogs, could not restore the imbalance of cellular defence mechanism provoked by streptozotocin.
This research article studied the effects of quercetin (QCT) on experimentally induced diabetes in rats. Rats were divided into three groups: a control group, a diabetic group induced with streptozotocin (STZ), and a QCT-treated group that received QCT before and after STZ induction. Blood glucose levels increased significantly in the diabetic group but decreased in the QCT-treated group. This study evaluated the safety of rambutan rind extract (RRE) in male rats. In the acute toxicity study, the lethal dose (LD50) of RRE was found to be greater than 5000 mg/kg. Significant decreases in body weight gain and food consumption were observed at doses above 1000 mg/kg. In the sub-chronic toxicity study, no mortality or signs of toxicity were observed at doses up to 1000 mg/kg/day for 30 days. At 2000 mg/kg/day, the mortality rate was 12.5%. was to investigate preliminary studies shows satisfactory result. The chromatographic studies like TLC, HPTLC and
HPLC show good spot. HPTLC shows maximum height and area of 18.83%.HPLC shows maximum peak at 1.867
minutes having area coverage of 87.4%.The free radical scavenging activity of chloroform fraction (CF) of a crude drug
shows 510μg/ml of scavenging activity. The IC50 value for MTT assay was found to be 84.7μg/ml. The GLUT4 study
shows significant uptake of glucose. PPAR gamma activity regulation of glucose disposal and insulin sensitivity in the
skeletal muscles shows concentration dependence response using standard Pioglitazone. The bud of Withania coagulants
will be a promising medicine for more ailments.

Hepatoprotective Effect of Cestrum parqui L. aerial parts and Phytochemical ...Jing Zang
This study deals with the investigation of hepatoprotective effect of 70% methanolic extract from Cestrum parqui aerial parts and determination of the bioactive components of the plant. The hepatoprotective effect of Cestrum parqui methanol extract (100, 500, 1000 mg/kg) was analysed on carbon tetrachloride (CCl4)-induced acute liver injury. The administration of a single dose of 40% CCl4 (1ml/kg b.w.) causes an increase in the activities of serum alanine aminotransferase (ALT) and aspirate aminotransferase (AST) enzymes and so pretreated orally of a dose from Cestrum parqui methanol extract (100, 500, 1000 mg/kg) and silymarin (200 mg/kg) for three consecutive days prior to The administration of a single dose of CCl4 significantly prevented the increase in the activities of these enzymes. Histological analysis showed that Cestrum parqui methanol extract at doses of 500 and 1000 mg/kg and silymarin reduced the incidence of liver lesions including vacuole formation, neutrophil infiltration and necrosis of hepatocytes induced by CCl4. The extract cause a negative result on the antioxidative enzymes, superoxide dismutase (SOD), glutathione peroxidase (GPx), glutathione reductase (GRd) and decreased malondialdehyde (MDA) level in liver, as compared to those in the CCl4-treated group and this suggests that the hepatoprotective activity of the extract is due to the antioxidant effect of the extract. Phytochemical analysis of the methanol extract from Cestrum parqui aerial parts showed that it contained different phytoconstituents, flavonoids, tannins, saponins, alkaloids, terpenes and carbohydrates.

Oxidative stress is currently suggested to play a role in the pathogenesis of Diabetes mellitus. The role of dietary management in diabetes mellitus is to provide a proper balance of total nutrients while meeting the special dietary needs of the patient. The present study was designated to evaluate the effect of special antidiabetic diet treatment upon oxidative stress parameters in the initial stages of the development of diabetes. Male Wistar strain rats were used as an experimental model, divided into five groups. A significant decrease in superoxide dismutase and total glutathione activities were observed in the liver of diabetic rats when compared with control animals. The plasma level of aminotransferases, creatine kinase, lactate dehydrogenase and urea were significantly increased after induction of diabetes, in all groups under treatment. In contrast, rats fed special diet food, have shown slight different, but not significant changes. The findings of the present study suggest that special diet formula useful for prevention of progressive hyperglycaemia in age induced diabetes in dogs, could not restore the imbalance of cellular defence mechanism provoked by streptozotocin.

This study deals with the investigation of hepatoprotective effect of 70% methanolic extract from Cestrum parqui aerial parts and determination of the bioactive components of the plant. The hepatoprotective effect of Cestrum parqui methanol extract (100, 500, 1000 mg/kg) was analysed on carbon tetrachloride (CCl4)-induced acute liver injury. The administration of a single dose of 40% CCl4 (1ml/kg b.w.) causes an increase in the activities of serum alanine aminotransferase (ALT) and aspirate aminotransferase (AST) enzymes and so pretreated orally of a dose from Cestrum parqui methanol extract (100, 500, 1000 mg/kg) and silymarin (200 mg/kg) for three consecutive days prior to The administration of a single dose of CCl4 significantly prevented the increase in the activities of these enzymes. Histological analysis showed that Cestrum parqui methanol extract at doses of 500 and 1000 mg/kg and silymarin reduced the incidence of liver lesions including vacuole formation, neutrophil infiltration and necrosis of hepatocytes induced by CCl4. The extract cause a negative result on the antioxidative enzymes, superoxide dismutase (SOD), glutathione peroxidase (GPx), glutathione reductase (GRd) and decreased malondialdehyde (MDA) level in liver, as compared to those in the CCl4-treated group and this suggests that the hepatoprotective activity of the extract is due to the antioxidant effect of the extract. Phytochemical analysis of the methanol extract from Cestrum parqui aerial parts showed that it contained different phytoconstituents, flavonoids, tannins, saponins, alkaloids, terpenes and carbohydrates.

The pygmy halfbeak Dermogenys colletei, is known for its viviparous nature, this presents an intriguing case of relatively low fecundity, raising questions about potential compensatory reproductive strategies employed by this species. Our study delves into the examination of fecundity and the Gonadosomatic Index (GSI) in the Pygmy Halfbeak, D. colletei (Meisner, 2001), an intriguing viviparous fish indigenous to Sarawak, Borneo. We hypothesize that the Pygmy halfbeak, D. colletei, may exhibit unique reproductive adaptations to offset its low fecundity, thus enhancing its survival and fitness. To address this, we conducted a comprehensive study utilizing 28 mature female specimens of D. colletei, carefully measuring fecundity and GSI to shed light on the reproductive adaptations of this species. Our findings reveal that D. colletei indeed exhibits low fecundity, with a mean of 16.76 ± 2.01, and a mean GSI of 12.83 ± 1.27, providing crucial insights into the reproductive mechanisms at play in this species. These results underscore the existence of unique reproductive strategies in D. colletei, enabling its adaptation and persistence in Borneo's diverse aquatic ecosystems, and call for further ecological research to elucidate these mechanisms. This study lends to a better understanding of viviparous fish in Borneo and contributes to the broader field of aquatic ecology, enhancing our knowledge of species adaptations to unique ecological challenges.

EWOCS-I: The catalog of X-ray sources in Westerlund 1 from the Extended Weste...Sérgio Sacani
Context. With a mass exceeding several 104 M⊙ and a rich and dense population of massive stars, supermassive young star clusters
represent the most massive star-forming environment that is dominated by the feedback from massive stars and gravitational interactions
among stars.
Aims. In this paper we present the Extended Westerlund 1 and 2 Open Clusters Survey (EWOCS) project, which aims to investigate
the influence of the starburst environment on the formation of stars and planets, and on the evolution of both low and high mass stars.
The primary targets of this project are Westerlund 1 and 2, the closest supermassive star clusters to the Sun.
Methods. The project is based primarily on recent observations conducted with the Chandra and JWST observatories. Specifically,
the Chandra survey of Westerlund 1 consists of 36 new ACIS-I observations, nearly co-pointed, for a total exposure time of 1 Msec.
Additionally, we included 8 archival Chandra/ACIS-S observations. This paper presents the resulting catalog of X-ray sources within
and around Westerlund 1. Sources were detected by combining various existing methods, and photon extraction and source validation
were carried out using the ACIS-Extract software.
Results. The EWOCS X-ray catalog comprises 5963 validated sources out of the 9420 initially provided to ACIS-Extract, reaching a
photon flux threshold of approximately 2 × 10−8 photons cm−2
s
−1
. The X-ray sources exhibit a highly concentrated spatial distribution,
with 1075 sources located within the central 1 arcmin. We have successfully detected X-ray emissions from 126 out of the 166 known
massive stars of the cluster, and we have collected over 71 000 photons from the magnetar CXO J164710.20-455217.
When I was asked to give a companion lecture in support of ‘The Philosophy of Science’ (https://shorturl.at/4pUXz) I decided not to walk through the detail of the many methodologies in order of use. Instead, I chose to employ a long standing, and ongoing, scientific development as an exemplar. And so, I chose the ever evolving story of Thermodynamics as a scientific investigation at its best.
Conducted over a period of >200 years, Thermodynamics R&D, and application, benefitted from the highest levels of professionalism, collaboration, and technical thoroughness. New layers of application, methodology, and practice were made possible by the progressive advance of technology. In turn, this has seen measurement and modelling accuracy continually improved at a micro and macro level.
Perhaps most importantly, Thermodynamics rapidly became a primary tool in the advance of applied science/engineering/technology, spanning micro-tech, to aerospace and cosmology. I can think of no better a story to illustrate the breadth of scientific methodologies and applications at their best.

The nonlinear activation functions in these networks result in interesting nonlinear equivariant maps between simple representations, and motivate the key player of this talk: piecewise linear representation theory.

6. 01 Introduction
Diabetes mellitus (DM) is a chronic metabolic disorder of insulin insufficiency due to a
gradual decrease in its production or tissue resistance to its action. The deficiency of insulin
leads to an increase in the concentration of blood glucose level, which gradually
compromises the physiology of vital organs.
The plant of Sorbaria tomentosa Lindl. Rehder belongs to the Rosaceae family. The plant is
a shrub and is 1.5–3 m tall. The leaves are short petiolate and 1–8 cm long and 2–4 cm
wide. The buds are 2 mm long and ovoid, obtuse, and brown.
Project was designed to assess the plant phytochemistry and evaluate it for efficacy
against type 2 DM using in vitro and in vivo approaches.

8. Extraction Fractionation
A Reddish Black Semisolid mass of methanolic
extract obtained
Filtrate evaporated at 40°C by Rotary
Evaporator
Filtered using muslin cloth, then through filter
paper
Soaked in 80% methanol for 14 days with
occasional shaking
Shade-Dried plant was cut into small pieces and
crushed to a powder
All extracts were collected in separate containers
and solvents removed by Rotary Evaporator
All n-hexane extracts were mixed.
n-hexane layer was separated and then again 500ml
n-hexane was added. Repeated thrice
Stirred vigorously in a separating funnel for some
time to form two layer
500ml of distilled water then 500ml n-hexane was
added
9. GC-MS Analysis Diabetes Induction
To find whether it contains a single compound or
a group of compounds
Interpretation of the mass spectrum was performed
using the National Institute of Standards and
Technology (NIST) database
Identification was done on basis of molecular mass,
molecular structure, and calculated fragments.
GC-MS analyzer (GC Clarius 500 Perkin Elmer),
Elite-1 Column(100% dimethyl polysiloxane) and
carrier gas used was Helium(99.9%)
2 µL methanolic plant extract
The blood glucose
level was recorded after 48 h of administration by a
glucometer.
Animals were kept on a fasting mode for 8–12 h but were
allowed freely to water before bioassay.
Causes
hyperglycemia in mice
Alloxan monohydrate (10%) was administered at a dose
of 160 mg kg−1 body weight
Alloxan monohydrate was obtained
from Sigma Aldrich
10. In-vitro Testing In-vivo Testing
Further Diluted using 10ml DW and absorbance
was checked at 540nm
Reaction was stopped by adding 1 mL of 3,5-
dinitro salicylic acid reagent. Then incubated for 5
min and cooled.
A solution of 1% starch (500 µL) prepared in 0.02
M sodium phosphate buffer was added to each
tube
Incubated for 10 min at 25°C
Test sample and standard drug were added to a
tubecontaining 0.20 mM PBS at pH 6.9 and and
a solution of α-amylase (conc 0.5 mg mL−1)
Group IV: Treatment group received i/p test
compounds (150 and 300 mg kg−1
Group III: Treatment group received
glibenclamide (5 mg kg−1)
Group II: Diabetic group received i/p
alloxan and
Tween 80.
Group I: Control (normal/non-diabetic)
group received
i/p only normal saline.
All animals were grouped into 4 groups
having 6 rats in each group.
0.1 M Na2CO3 soln. was
added to stop the reaction.
Absorption was observed at
405 nm.
3 mM pNPG solution (100 µL)
was added to it and was
incubated for
10 min at 37°C.
pH was adjusted to 6.8. The
solution mixture was mixed
and incubated for 15 min at
37°C.
Various concentrations of
plant samples were mixed
with 50 µL of enzyme soln.

Experimental animals
Animals used in this study
were rats weighing 145–180 g
with an average weight of
162.5 g. The animals were
placed in an animal house
under standard laboratory
conditions of 25°C temperature
and 12 h light and dark with the
supply of water ad libitum
during the whole experiment
period.

13. GC-MS analysis
35 various peaks were observed. The most abundant compounds were oleic acid (20.52%), α-sitosterol (10.44%),
8-octadecenoic acid, methyl ester, (E) (9.11%), trichothec9-en-8-one, 4-(acetyloxy)-12,13-epoxy-3,7,15-trihy droxy-
(3α,4α,7α), (7.57%), phthalic acid, 6-ethyloct-3-yl 2-ethylhexyl ester (7.55%), 9,12,15-octadecatrienoic acid, 2,3-
bis[(trimethylsilyl)oxy]propyl ester, (Z,Z,Z) (7.44%), lupeol (6.02%), α-D-galactopyranose, 6-O-(trimethylsilyl)-cyclic
1,2:3,4-bis (butylboronate) (5.15%), and α-sitosterol trimethylsilyl ether (5.06%)

14. In-vitro Study Inhibition of α-amylase and α- glucosidase
The most potent extract was chloroform, which
exhibited an inhibition of 84.4 ± 1.12% of the enzyme
at a concentration of 1,000 µg 𝑚𝑙−1
.
IC50 for the crude methanolic extract was 530 µg𝑚𝑙−1
thus it exhibited concentration-dependent moderate
enzyme inhibitory potentials.

15. In-vivo Studies
1. ACUTE TOXICITY STUDY
No morbidity or mortality was observed.
No aberrant behaviour was observed during the acute toxicity studies at the tested doses.
2. ANTI-HYPERGLYCEMIC EFFECT
Diabetic animals treated with the
standard drug exhibited a gradual
decrease in blood glucose during
therapy.
The treatment of diabetic animals
with 150 mg kg−1 dose of crude
methanolic extract also showed a
steady decrease in blood glucose
levels. Furthermore, the crude
methanolic extract at a dose of 300
mg kg−1 exhibited lowered blood
glucose levels.

16. In-vivo Studies
3. CHANGES IN THE WEIGHT OF VITAL ORGANS
Overall, no major changes were recorded during therapy in the weight of these organs

17. 04 CONCLUSION
The results of the phytochemical analysis of the study revealed that plants
contain various metabolites, which might be responsible for the anti-diabetic
potentials of the plant.
Various extracts exhibited concentration-dependent inhibition of key
enzymes implicated in postprandial hyperglycemia and type-2 diabetes.
The crude methanolic extract showed considerable in vivo efficacy against
alloxan-induced diabetes in rats.
Further studies include activity-guided isolation of bioactive metabolites, and
molecular studies are required for further results.
18. 05 OUTCOMES AND DELIVERIES
The anti-diabetic activity has been proved and the compounds have been
identified as a whole. But which compound is the prime phyto-component
that shows this anti-diabetic property is unknown.
In vivo studies needed to be performed with other solvents. Testing with
Methanolic extract was performed, Chloroform, Ethyl Acetate and n-Hexane
was not performed.
Acute toxicity study data is not shown.
HPTLC and Electrophoresis might be helpful in finding the active
metabolite.
Molecular Docking can be a resourceful tool in finding the active site.
