게 생긴 당을 유산균이 발효를 통해 젖산으로 만드는 중간단계 ③이렇게
만들어진 김치를 잘 포장하거나 저장해 김치라는 최종 상품으로 만드는
후단계로 구분할 수 있다. 이 3단계는 대부분의 생물공정, 즉 플라스틱을
갈대로부터 만드는 바이오화학공정에도 적용된다.

6. 바이오매스는 크게 보면 사탕수수와 같이 설탕(sugar)이 주성분인
것, 옥수수, 고구마와 같이 전분(starch)이 주성분인 것, 나무, 억새 등과
같이 셀룰로오스(cellulose)가 주성분인 것으로 나눌 수 있다.
사탕수수로부터 쉽게 얻어지는 설탕은 직접 미생물이 섭취할 수 있
으므로 별도의 전처리 또는 당화공정이 없어도 된다.
그러나 옥수수처럼 전분이 주성분인 경우는 분자 크기가 커서 직접
미생물이 섭취할 수가 없다. 그렇기 때문에 작은 크기의 포도당으로 쪼
개 주어야 한다. 이 과정에 물이 들어가게 돼 이를 가수분해(hydrolysis)*
라고 한다. 전분의 가수분해는 주로 효소에 의해 이루어지는데, 이는 우
리가 밥을 먹고 소화시키는 것과 같다.

7. 우리가 쌀로 밥을 해서 먹으면 밥은 침샘에서 나오는 효소에 의해 위
에서 분해되어 포도당으로 변환되고, 이 포도당은 우리 몸의 세포에 전
달되어 대사작용*에 사용된다. 전분을 포도당으로 바꾸는 첫 번째 과정
은 우리가 밥을 하듯이 온도를 올려서 전분의 구조를 느슨하게 만드는
것이다. 이래야 효소가 전분에 접근해 전분을 포도당으로 쪼갤 수 있다.
효소에 의해 전분을 포도당으로 쪼개는 과정은 세부적으로 복잡한
메커니즘으로 구성되어 있다. 단순하게 생각하면 글루코아밀라아제
(glucoamylase)*라는 효소에 의해 전분이 포도당으로 변환되는 것이다.
따라서 이때 중요한 것은 글루코아밀라아제 효소의 성능이다. 전분으
로부터 포도당을 얻는 방법은 오래전부터 사용되어 오고 있기 때문에
효율이 좋다.
그러나 셀룰로오스를 주성분으로 하는 바이오매스로부터 포도당을
얻는 방법은 최근에 본격적으로 연구가 이루어지고 있어 앞으로 많은
기술 발전이 기대된다.
지금까지 알려진 내용을 정리하면 다음과 같다. 나무 종류, 억새와
같은 풀 종류, 볏짚과 같은 폐농산물의 성분을 분석하면 크게 셀룰로오
스(cellulose), 헤미셀룰로오스(hemicellulose)*, 그리고 리그닌(lignin)으
로 되어 있다.
셀룰로오스는 탄소가 6개인 포도당이, 헤미셀룰로오스라는 탄소가

8. 5개인 자이로스가 길게 연결된 것이다. 리그닌은 방향족 화합물*이 포
함된 바이오플라스틱 물질로서 셀룰로오스와 헤미셀룰로오스의 구조
를 받쳐 주는 접착제와 같은 역할을 한다. 일반적으로 리그닌은 별도로
분리해 계면활성제 등으로 사용하거나 연소시켜 에너지를 얻는 데 사
용한다.
어떻게 해야 셀룰로오스와 헤미셀룰로오스라는 커다란 분자의 화합
물을 미생물이 섭취할 수 있는 작은 분자로 분해시킬 수 있을까?
기본 원리는 전분을 분해하는 것과 같다. 목질계 바이오매스를 고온
에서 산 또는 알칼리 처리를 해 분자구조를 느슨하게 할 필요가 있다.
이렇게 전처리를 한 후에 효소를 넣어 주면 작은 분자의 포도당과 자이
로스로 분해된다.
목질계 바이오매스를 분해하는 것은 전분을 분해하는 것보다 복잡

9. 하다. 이것은 목질계 바이오매스 자원을 이용하는 것이 비용이 많이 소
요된다는 것을 뜻한다. 여기에 기술 개발의 필요성이 있다.
필요한 기술은 전처리는 어떻게 하는 것이 좋은가, 효소를 어떻게 싸
고 좋게 만들 수 있는가, 어떻게 전처리와 효소공정*을 결합하는 것이
좋은가와 관련된 것들이다.
이 중에서 제일 중요한 것은 셀룰로오스를 분해하는 셀룰라아제
(cellulase) 효소*를 값싸고 성능이 우수하게 개량하는 것이다. 현재 이
목표를 달성하기 위해 수많은 과학자들이 연구에 몰두하고 있다.
셀룰로오스는 포도당이 직선상으로 길게 연결되어 있다. 비유하면
셀룰라아제 효소는 셀룰로오스 표면에 부착하여 포도당과 포도당의 연
결부위를 잘라내는 것이다. 이 효소의 성능을 개선하기 위해서는 포도
당과 포도당의 연결부위를 빨리 잘라내도록 하는 기술, 효소를 값싸게
생산하는 기술 개발이 요구된다. 효소의 개량기술과 대량 생산기술, 즉
효소공학(enzyme engineering) 기술이 핵심인 것이다.
에탄올을 생산하는 미생물
효모(yeast)라는 미생물은 포도당을 에탄올로 변환시킬 수 있다. 효
모는 산소가 없는 상황에서도 에너지를 얻기 위해 포도당을 에탄올로

10. 변환시킨다.
전분(starch)으로부터 에탄올을 생산하려면 첫 단계는 전분을 포도당
으로 가수분해해야 한다. 전분은 크기가 크기 때문에 효모미생물 내로
들어갈 수 없다. 그래서 전분을 효모미생물이 먹을 수 있는 작은 크기의
포도당으로 분해해야 한다. 전분의 가수분해는 효소에 의해 일어난다.
이 과정을 당화라고 한다. 당화효소*로는 아밀라아제(amylase)* 효소가
이용된다.
이렇게 얻은 포도당에 효모미생물을 넣어 주면 성장하면서 에탄올
을 만들어낸다. 이 과정은 발효*라고 한다. 그런데 이 과정에서 생각할
점이 있다. 꼭 전분을 포도당으로 분해한 다음에 효모미생물*을 넣어 주
어야 하는가?
이러한 발상에서 그리고 생명공학기술의 발달에 의해 효모미생물에
아밀라아제 효소를 생산하는 유전자를 넣은 새로운 효모미생물이 탄생
하였다.

11. 아밀라아제 효소를 합성하는 유전자를 넣어준 새로운 효모미생물은
아밀라아제 효소를 합성해 미생물 밖으로 내보낸다. 그러면 이 효소에
의해 전분은 포도당이 되고, 효모미생물은 변환된 포도당을 먹고 에탄
올을 만들어 내는 것이다. 2단계의 전통적인 방법이 1단계의 단순화된
방법으로 바뀐 것이다.
생명공학기술의 진보는 목질계 바이오매스로부터 에탄올을 생산하
고자 하는 경우에도 적용할 수 있다. 목질계 자원은 크게 셀룰로오스
(cellulose), 헤미셀룰로오스(hemicellulose) 그리고 리그닌(lignin)으로
구성되어 있다. 철근 콘크리트 구조물에 비유하면 셀룰로오스는 구조
를 지탱해 주는 철근, 헤미셀룰로오스는 셀룰로오스를 연결해 주는 철
사, 그리고 리그닌은 서로를 하나로 붙게 해주는 콘크리트에 비유할 수
있다.
리그닌은 구조가 매우 복잡한 바이오플라스틱 화합물이므로 에탄올
생산에 활용할 수 없다. 그러나 셀룰로오스를 분해하면 탄소가 6개인
포도당이 얻어지고 헤미셀룰로오스를 분해하면 탄소가 5개인 자이로
스(xylose)가 얻어진다.

12. 그런데 효소미생물은 포도당만 먹을 수 있고 자이로스는 섭취할 수
없다. 관계되는 대사경로가 없기 때문이다. 역시 생명공학기술의 진보
는 포도당과 자이로스 두 가지 탄소원을 다 먹고 에탄올을 만들어 내는
새로운 효모미생물을 탄생시켰다.
기술의 진보는 지금도 계속되고 있다. 동시당화 발효기술*과 포도당
과 자이로스를 이용하는 새로운 효모미생물 기술 2개 등을 결합하면 한
차원 높은 기술이 탄생한다. 기술의 핵심은 미생물의 대사 작용을 조작
하는 기술, 즉 대사공학(metabolic engineering)기술이다.
미생물을 배양하거나 화학적으로 합성하여 얻을 수 있는 화합물은
매우 많다. 그중 12개의 화합물이 향후 플랫폼(platform)화합물로서 많
이 사용될 것으로 예측되었으나 최근 10개로 수정됐다.

효소는 세포 내의 단백질로 구성되어 있는 생체촉매 분자이며 효모는 살아 있는 미생
물이다.
박테리아, 곰팡이, 효모 등이 미생물에 속한다. 미생물은 우리 몸에 질병을 일으키기
도 하지만, 유용한 물질을 만들어 내는 유익한 미생물도 많다.
김치는 미생물의 발효를 이용한 발효식품이며 페니실린은 페니실리움이라는 곰팡이
가 만들어 내는 대사산물로서 항생제로 이용되고 있다.
