3. LITHIUM- Monovalent cation.
Mechanism of action:
Li inhibits the hydolysis of Ionositol phosphate-1 by
Ionositol monophosphatase. Hence, Free ionositol or
IP3/DAG are reduced.
Li partly reduces body Na+2,ths effects ionic fluxes
across brain cells / modify property of cellular
membranes.
Li decreases NA & DA release in bran of treated animals
without affecting 5-HT release.
Li inhibits action of ADH on distal tubules & causes DI

5. P/K
• Li given by mouth as Carbon salt & excreted in
urine.Abputhalf of the dose s excreted in
12Hrs.
Sodium depletion reduces the rate of excretion
by increasing reabsorption of li+ by PCT.

6. Adverse Effects
Nausea ,vomiting, Diarrhoea, Renal effects (coz
of inhbition of ADH release),Serous renal
tubular damage,Thyroid enlargement,Weight
gain,
Acute Li+ toxcity: Neurological effects
progressing from confusion,& motor
impairment to coma,convulsions & death.

7. DI
Li+ +Diuretics Promotes PCT reabsorption.
NSAID’S ,ACE inhibitors,Tetracyclines 
Li retention.
Neroleptics + Li+ Tremor & Rigidity

8. uses
Acute Mania– Motor restlessness, Non-stop
talking, Progressive loss of contact with
reality, Inappropriate cheerfulness,
Irritability.
Recurrent unipolar depression.
Recurret neuropsychiatric illness.
