Ethidium homodimer is responsible for the red color observed but cannot cross intact plasma membranes. Calcein is a green dye that can cross membranes but is then trapped inside cells by esterase, which makes it negatively charged. Ethidium homodimer, a positively charged stain, also becomes trapped inside cells with calcein.
