Donecept Tablets (Generic Donepezil Hydrochloride Tablets)

Donecept (Generic Donepezil Hydrochloride Tablets) is a
cholinesterase inhibitor that reduces or prevents acetylcholine
breakdown in brain tissue. Donecept tablets are used to treat
the symptoms of dementia that are found in patients
diagnosed as having mild to moderately severe Alzheimer's
disease.
Donecept is not a cure; it reduces symptoms. Donepezil
hydrochloride tablets do not cure Alzheimer’s disease. All
patients with Alzheimer’s disease get worse over time, even if
they take Donecept tablets.

Active Ingredient
The active ingredient present in Donecept tablets is
Donepezil Hydrochloride. Each tablet contains Donepezil
Hydrochloride equivalent to either 5 mg or 10 mg of Donepezil.

under various drug trade names that include
Aricept, Donecept, Donaz, Alzidoc, Alzil, Donep,
Dopezil, Dorent.

Uses of Donecept Tablets
Donecept Tablets (Generic Donepezil Hydrochloride Tablets) are
used to treat dementia (memory loss and mental changes)
associated with mild, moderate, or severe Alzheimer's disease.
Donepezil will not cure Alzheimer's disease, and it will not stop
the disease from getting worse, but it may improve memory,
awareness, and the ability to function. Efficacy has been
demonstrated in patients with mild, moderate, and severe
Alzheimer’s Disease.
This medication is an enzyme blocker that works by restoring the
balance of natural substances (neurotransmitters) in the brain.

Donecept Tablets Dosage
The dosage of Donecept tablets (Generic Donepezil Hydrochloride
Tablets) is given below:
The recommended adult dosage of Donecept Tablets is orally once a day
just before bedtime, with or without meals . Treatment should be started
with Donecept Tablets 5 mg/day (once-a-day dosing).
Dosing in Mild to Moderate Alzheimer’s Disease: The recommended
starting dosage of Donecept Tablets is 5 mg administered once per day in
the evening, just prior to retiring. The maximum recommended dosage of
Donecept Tablets in patients with mild to moderate Alzheimer’s disease is
10 mg per day. A dose of 10 mg should not be administered until patients
have been on a daily dose of 5 mg for 4 to 6 weeks.

Donecept Tablets Dosage
The dosage of Donecept tablets (Generic Donepezil Hydrochloride
Tablets) is given below:
Dosing in Moderate to Severe Alzheimer’s Disease: The recommended
starting dosage of Donecept Tablets is 5 mg administered once per day in
the evening, just prior to retiring. The maximum recommended dosage of
Donecept Tablets in patients with moderate to severe Alzheimer’s disease is
10 mg per day. A dose of 10 mg should not be administered until patients
have been on a daily dose of 5 mg for 4 to 6 weeks.

Contraindications
Donecept tablets are contraindicated in persons with a
hypersensitivity (allergy) to Donepezil Hydrochloride,
Piperidine derivatives or any of the other ingredients of this
medicine.

Storage Instructions
Donecept Tablets (Generic Donepezil
Hydrochloride Tablets) have to be stored at
controlled room temperature i.e. from 20°C to
25°C (68°F to 77°F) with excursions
permitted between 15° to 30°C (59° to 86°F).
Keep this as well as all other medicines away
from children and pets.

Warnings
Donepezil Hydrochloride (Donecept Tablets), as a cholinesterase inhibitor,
is likely to exaggerate succinylcholine-type muscle relaxation during
anaesthesia.
Because of their pharmacological action, Cholinesterase inhibitors may
have vagotonic effects on the sinoatrial and atrioventricular nodes
manifesting as bradycardia or heart block. Through their primary action,
cholinesterase inhibitors may be expected to increase gastric acid secretion
due to increased cholinergic activity. Patients must be monitored closely
for symptoms of active or occult gastrointestinal (GI) bleeding, especially
those at increased risk for developing ulcers.
Donecept can cause vomiting. Patients should be observed closely at
initiation of treatment and after every dose increase.

Warnings
The use of Donecept in a dose of 23 mg once daily is associated with
weight loss.
Because of their cholinomimetic actions, cholinesterase inhibitors should
be prescribed with care to patients with a history of asthma or obstructive
pulmonary disease.
Cholinomimetics may cause bladder outflow obstructions.
Cholinomimetics are believed to have some potential to cause
generalized convulsions.
Cholinesterase inhibitors should be prescribed with care to patients with a
history of asthma or obstructive pulmonary disease.
You must avoid drinking alcohol while you are being treated with
Donepezil tablets, because it could reduce the effect of Donepezil tablets.

Effects on Ability to Drive and Use Machines
Donepezil Hydrochloride Film-coated tablets (Donecept tablets)
have minor or moderate influence on the ability to drive and use
machines.
Dementia may cause impairment of driving performance or
compromise the ability to use machinery.
Furthermore, Donepezil can induce fatigue, dizziness and muscle
cramps, mainly when initiating or increasing the dose. The treating
physician should routinely evaluate the ability of patients on
Donepezil to continue driving or operating complex machines.

Donecept Tablets Side Effects
Common side effects observed during trials with Donepezil
Hydrochloride tablets (Donecept tablets) are nausea,
diarrhea, insomnia, vomiting, muscle cramps, fatigue, and
anorexia.

Donecept Tablets Side Effects
Donepezil Hydrochloride tablets may cause the following serious side
effects:Slow heartbeat and fainting. This happens more often in people with
heart problems. Call the doctor right away if the patient faints while taking
Donepezil Hydrochloride tablets. More stomach acid. This raises the chance
of ulcers and bleeding. The risk is higher for patients who had ulcers, or take
aspirin or other NSAIDs. Worsening of lung problems in people with asthma or
other lung disease.Seizures. Difficulty passing urine.
Call the doctor immediately if the patient has: Fainting, Heartburn or stomach
pain that is new or won't go away, Nausea or vomiting, blood in the vomit,
dark vomit that looks like coffee grounds, Bowel movements or stools that
look like black tar, New or worse asthma or breathing problems, Seizures or
Difficulty passing urine.

Overdose
Overdosage with cholinesterase inhibitors like Donepezil
Hydrochloride (Donecept Tablets) can result in a cholinergic
crisis characterized by severe nausea, vomiting, salivation,
sweating, bradycardia, hypotension, respiratory depression,
collapse and convulsions. Increasing muscle weakness is a
possibility and may result in death if the respiratory muscles
are involved.

Overdose
Tertiary anticholinergics such as atropine may be used as an antidote for
Donepezil Hydrochloride (Donecept Tablets) overdosage. Intravenous
atropine sulphate titrated to effect is recommended, with an initial dose of
1.0–2.0 mg IV with subsequent doses based upon clinical response. Atypical
responses in blood pressure and heart rate have been reported with other
cholinomimetics when co-administered with quaternary anticholinergics
such as glycopyrrolate.
It is not known whether Donepezil and/or its metabolites can be removed
by dialysis (haemodialysis, peritoneal dialysis, or haemofiltration). Dose-
related signs of toxicity in animals included reduced spontaneous
movement, prone position, staggering gait, lacrimation, clonic convulsions,
depressed respiration, salivation, miosis, tremors, fasciculation and lower
body surface temperature.

Donecept Tablets During Pregnancy
Donepezil Hydrochloride (Donecept Tablets) is classified
by the US FDA as Pregnancy Category C.
There are no adequate or well-controlled studies in
pregnant women. Donecept (Donepezil Hydrochloride
Tablets) should be administered during pregnancy only
if the potential benefit justifies the potential risk to the
fetus.
