D-Veniz (Generic Desvenlafaxine Succinate Extended Release Tablets)

D-Veniz (Generic Desvenlafaxine Succinate
Extended Release Tablets) is indicated for the
relief of major depressive disorder.
Desvenlafaxine is used to treat depression. It may
improve your mood, feelings of well-being, and
energy level. Desvenlafaxine is known as a
serotonin-norepinephrine reuptake inhibitor
(SNRI). It works by helping to restore the balance
of certain natural substances (serotonin and
norepinephrine) in the brain.

Active Ingredient
The active ingredient present in D-Veniz tablets is
Desvenlafaxine Succinate. Each film coated extended release tablet
contains 76 mg or 152 mg of Desvenlafaxine Succinate equivalent
to Desvenlafaxine 50 mg or 100 mg.

Desvenlafaxine Succinate Tablets – Brand Names
Desvenlafaxine Succinate Tablets are
available under various Drug trade names
that include Pristiq, Desfax, Ellefore, D-
Veniz, D-Venlor, Newven OD, Venpower,
Venz-OD and Zyven-OD.

Uses of D-Veniz Tablets
D-Veniz (Generic Desvenlafaxine Succinate Extended Release Tablets) is a
serotonin and norepinephrine reuptake inhibitor (SNRI) indicated for the
treatment of adults with major depressive disorder (MDD).
D-Veniz is used in the treatment and prevention of relapse of depression.
Depression can affect your whole body and may cause emotional and
physical symptoms such as feeling low in spirit, being unable to enjoy life,
poor appetite or overeating, disturbed sleep, loss of sex drive, lack of
energy and feeling guilty over nothing.

D-Veniz Tablets Dosage
The dosage of D-Veniz tablets (Generic Desvenlafaxine Succinate
Extended Release Tablets) is given below:
The recommended adult dosage of D-Veniz (Desvenlafaxine
Succinate Tablets) for the initial treatment of Major Depressive
Disorder (MDD) is 50 mg once daily, with or without meals.
There was no evidence that doses greater than 50 mg per day
confer any additional benefit.
The 25 mg per day dose is intended for a gradual reduction in dose
when discontinuing treatment or dosing in severe renal and end-
stage renal disease patients.

D-Veniz Tablets Contraindications
D-Veniz is contraindicated in patients with a hypersensitivity to
Desvenlafaxine Succinate, Venlafaxine Hydrochloride or any other
component of this medication.
D-Veniz (Desvenlafaxine Succinate Tablets) must not be administered to patients
taking other medications for depression known as monoamine oxidase inhibitors,
even if they have stopped taking them, but have taken them within the last 14
days due to the risk of serious, sometimes fatal, drug interactions with SNRI or
SSRI treatment or with other serotonergic drugs.
These interactions have been associated with symptoms that include tremor,
myoclonus, diaphoresis, nausea, vomiting, flushing, dizziness, hyperthermia with
features resembling neuroleptic malignant syndrome, seizures, rigidity, autonomic
instability with possible rapid fluctuations of vital signs, and mental status changes
that include extreme agitation progressing to delirium and coma.

Storage Instructions
D-Veniz (Generic Desvenlafaxine Succinate
Extended Release Tablets) have to be stored
at controlled room temperature i.e. from 20°C
to 25°C (68°F to 77°F) with excursions
permitted between 15° to 30°C (59° to 86°F).
Keep this as well as all other medicines away
from children and pets.

Warnings
Patients must be monitored for clinical worsening and suicide risk.
Serotonin syndrome or NMS-like reactions have been reported with SSRIs
and SNRIs in some patients. Discontinue D-Veniz (Desvenlafaxine
Succinate Tablets) and start supportive treatment in case symptoms
occur.
High Blood Pressure has occurred in patients undergoing treatment with
with Desvenlafaxine Succinate Tablets. Hypertension must be controlled
before starting treatment. Blood pressure has to be monitored regularly
during treatment.
Desvenlafaxine Succinate tablets may increase the risk of abnormal
bleeding. Patients must also be cautioned about the risk of bleeding
associated with the concomitant use of D-Veniz and NSAIDs, aspirin, or
other drugs that affect coagulation.

Warnings
Activation of Mania/Hypomania has occurred. Use Desvenlafaxine Succinate
Tablets cautiously in patients with Bipolar Disorder. Caution patients about
the risk of activation of mania / hypomania. Mydriasis has occurred with
Desvenlafaxine Succinate Tablets. Patients with raised intraocular pressure or
those at risk of angle-closure glaucoma must be monitored.
Taper the D-Veniz dose whenever possible when discontinuing and monitor
for discontinuation symptoms since discontinuation symptoms have occurred.
D-Veniz has to be used with caution in patients with cardiovascular or
cerebrovascular disease.
Cholesterol and Triglyceride Elevation has has occurred in certain patients
and hence must be used cautiously in patients with lipid metabolism disorders.
Consider monitoring serum cholesterol and triglyceride.

Warnings
Since renal impairment reduces the clearance of D-Veniz, dosage
adjustment is necessary in severe and ESRD. In moderate renal
impairment, the dose should not exceed 50 mg/day.
Since seizure can occur in certain patients. Use cautiously in patients
with seizure disorder.
Hyponatremia can occur in association with SIADH..
Medicines that contain Desvenlafaxine or Venlafaxine should not be
used simultaneously with D-Veniz (Desvenlafaxine Succinate Tablets).
Interstitial Lung Disease and Eosinophilic Pneumonia can occur.

Desvenlafaxine Succinate (D-Veniz Tablets) Side Effects
Most common Desvenlafaxine Succinate (D-Veniz Tablets)
side effects with an incidence rate of 5% or greater and
twice the rate of placebo in the 50 or 100 mg dose groups
were: nausea, dizziness, insomnia, hyperhidrosis,
constipation, somnolence, decreased appetite, anxiety, and
specific male sexual function disorders.

Overdose
During pre-marketing clinical studies, no cases of fatal acute overdose of
Desvenlafaxine have been reported.
Among the patients included in the Major Depressive Disorder (MDD) pre-
marketing studies of Desvenlafaxine, there were four adults who took
Desvenlafaxine Succinate (4000 mg , 900, 1800 and 5200 mg ); all patients
recovered. In addition, one patient's 11-month-old child accidentally
consumed 600 mg of Desvenlafaxine Succinate, was treated, and recovered.
The adverse reactions reported within 5 days of an overdose more than 600
mg that were possibly related to Desvenlafaxine included headache,
vomiting, agitation, dizziness, nausea, constipation, diarrhoea, dry mouth,
paraesthesia and tachycardia.

Treatment of Overdose
No specific antidotes for Desvenlafaxine tablets are currently known.
Overdose treatment must consist of those general measures employed
in the management of over dosage with any SSRI/SNRI. Ensure an
adequate airway, oxygenation and ventilation. Monitor cardiac rhythm
and vital signs of the patient.
General supportive and symptomatic measures are also recommended.
Gastric lavage with a large-bore orogastric tube with appropriate
airway protection, if required, may be prescribed if performed soon
after ingestion or in symptomatic patients. Activated charcoal must be
administered. Induction of emesis is not recommended.

Desvenlafaxine Succinate (D-Veniz Tablets) During Pregnancy
Desvenlafaxine Succinate (D-Veniz Tablets) has been classified by
the US FDA as Pregnancy Category C.
When Desvenlafaxine Succinate was administered orally to pregnant rats
and rabbits during the period of organogenesis, there was no evidence of
teratogenicity in rats at any doses tested, up to 10 times a human dose in
rats, and up to 15 times a human dose in rabbits.
There are however no adequate and well-controlled studies of D-Veniz
conducted in pregnant women. Therefore, D-Veniz (Desvenlafaxine
Succinate Tablets) should be used during pregnancy only if the potential
benefits justify the potential risks.
