What is
protoplast?
Definition of protoplast
1.The protoplast is entire of the cell,
excluding the cell wall and also known
as a naked plant cell.
2.Protoplast is the biological active
and most significant material of the
plant cell.
History of Protoplast Culture
1.The term Protoplast was given by
Hanstein in 1880.
2.In 1892 klercker isolate the protoplast at
the first time by the using mechanical
method.
3.In 1960 E.C Cooking for the first time
isolated the protoplast of plant tissue by
the using enzymatic degradation.
Isolation of Protoplast
Protoplasts are isolated by two techniques:
1.Mechanical method.
2.Enzymatic method.
Mechanical method of protoplast isolation
•Protoplast isolation by mechanical method is a
imperfect and tedious procedure. This results in
the isolation of a very small number of
protoplasts. The technique involves the following
slage:
1. A small piece of epidermis from a plant is
selected.
2.The cells are subjected to plasmolysis. This
causes protoplasts to shrink away from the cell
walls.
3.The tissue is dissected to release the
protoplast.
Mechanical method
Limitations of mechanical
method.
1.Yielding of Protoplast is very low.
2.Visibility of protoplast is low.
3.This laboratory method is very
hard.
