Type of staining done-gram staining
Stain used-methylene blue , safranin
Suspected microbe - propionibacterium and staphylococcus
Identifying features of staphylococcus – spherical structure,
attached colonies, gram +ve
Identifying features of propionibacterium -rod shaped
structure, non motile , gram +ve
Effect of staphylococcus and propionibacterium on scalp - these
bacteria’s live in human scalp ad when there population
increases this lead to formation of flaky scales in scalp called
“dandruff”
This condition is common in most of people and dosent require
medical care it can be treated by any anti dandruff shampoo .

Standing water from money plant pot-
Type of staining- gram staining
Stain used – safranin , methylene blue
Suspected microorganism - e.coli , bacillus sapp.:
Identifying features of e.coli – rod shaped , 1-2
micrometer in length , gram –ve
Identifying features of bacillus sapp.: - rod shaped ,
gram+ve , its size is greater than many gram –ve
bacteria’s
Effect of E.coli on plant and human – E.coli is not plant
pathogen . most of e.coli strains are non-pathogenic to
humans only virulent strains cause gut/ gastrointestinal
diseases .
Effect of bacillus sapp.: on plant and humans – most
bacillus species are beneficial for plants and animals
except some human pathogenic species like anthrax
bacillus

Bread mold
Staining technique- lactophenol cotton blue staining
Stain used – lactophenol solution
Suspected fungal species – rhizopus spp.
Identifying feature of rhizopus – aseptate , branched ,
has dark colored sporangium and sporangiospore
Effect of rhizopus to human – it is responsible for
mucormycosis which is rare but life threatening fungal
infection .

Staining used – gram staining
Stain used – methylene blue , safranin
Suspected microorganism- lactobacillus
Identifying feature of lactobacillus – rod shaped , gram
+ve
Effect of lactobacillus on humans – lactobacillus is a
beneficial bacteria to humans as it improves digestion ,
immune system ,oral health , lactose digestion etc.

Topical layer of human skin
Staining used – gram staining
Stain used – methylene blue , safranin
Suspected microorganism – streptococcus aureus
Identifying features of streptococcus aureus – spherical ,
appear in cluster , gram +ve , golden yellow colonies due
to production of caratenoid pigament
Effect of streptococcus aureus to humans –
It is common bacteria found on human skin and it
prevent growth of other microbial colonies on skin . it
sometimes cause skin and respiratory infections and it
has antibiotic resistance from a variety of antibiotic .
