Definition
• A plasmid is a small DNA molecule within
a cell that is physically separated from
a chromosomal DNA and can replicate
independently.
• Mostly found in bacteria (presents to be as
a small, circular, double-stranded DNA
molecule—as the picture shows), also
found in archaea and eukaryotic
organisms.
Natural and Artificial Plasmids
In nature, plasmids often carry genes that is essential for the survival of
an organism. Therefore people generate artificial plasmids that are
widely used as vectors to drive the replication of recombinant
DNA sequences within host organisms. They can be used for:

• Gene therapy

1. They are small. If they are small, they are easier to
isolate (you get more), handle (less shearing), and
transform.
2. They have multiple restriction enzyme sites. More
sites give you greater flexibility in cloning, perhaps even
allowing for directional cloning.
• 3. They have multiple ORIs. It is important to note that
two genes must have different ORIs if they are going to
be inserted in the same plasmid.
