This study aimed to induce apoptosis in a non-invasive retinoblastoma cell line through transfection of a suicide gene plasmid driven by a tissue-specific promoter. The researchers transfected the plasmid into the cell line and detected over 10-fold increased expression of the suicide gene. Protein expression was also confirmed through immunofluorescence. Apoptosis assays showed that the suicide gene induced apoptosis in over 60% of the transfected non-invasive retinoblastoma cells. The researchers concluded that the tissue-specific promoter-driven suicide gene in a non-viral vector could be used as a potential treatment for retinoblastoma.

This study characterized stem rust resistance genes in Nebraska wheat germplasm. 270 F6 lines and 60 F7 lines were phenotyped for stem rust resistance and genotyped using GBS and marker analysis. Single marker analysis identified Sr24 as significantly associated with resistance in the F6 population and Sr38 in the F7 population. Genome-wide association also identified additional resistance genes. The study provides information on stem rust resistance genes in these wheat lines to enable gene pyramids for improved resistance breeding. Further testing will identify additional genes such as Sr6, Sr30, Sr36, Sr9e and SrTmp.

Expression of BTK* can also relieve interleukin-5 dependence in a B cell line. The results demonstrate that the PH domain critically regulates BTK transformation and activation.

Adenoviruses can be used for virotherapy by arming them with genes to target cancer cells. Three pigeonpea recombinant inbred line mapping populations, PRIL_A, PRIL_B, and PRIL_C, have been developed that segregate for resistance to fusarium wilt and sterility mosaic disease. The populations range from 329-366 lines and were derived from crosses between parental lines that differ in disease resistance. Over 4,600 markers were screened on the parental lines, with 159-157 found to be polymorphic between each population's parents. The populations will be phenotyped for disease resistance and genotyped with the polymorphic markers to construct genetic maps and identify genomic regions associated with resistance. Several promising lines have already been selected from the populations with high yield and disease resistance.

The genome was sequenced and assembled into 23 contigs totaling 2.3 megabases. Annotation identified over 2,000 protein-coding genes, 10 rRNAs, and 50 tRNAs. Comparison to other P. multocida genomes confirmed the identification as P. multocida and determined it was sequence type 128, which has only been reported once before from a sheep in New Zealand. The genome provides new genomic resources that can improve understanding of P. multocida pathogenesis.

Three pigeonpea recombinant inbred line mapping populations, PRIL_A, PRIL_B, and PRIL_C, have been developed that segregate for resistance to fusarium wilt and sterility mosaic disease. The populations range from 329-366 lines and were derived from crosses between parental lines that differ in disease resistance. Over 4,600 markers were screened on the parental lines, with 159-157 found to be polymorphic between each population's parents. The populations will be phenotyped for disease resistance and genotyped with the polymorphic markers to construct genetic maps and identify genomic regions associated with resistance. Several promising lines have already been selected from the populations with high yield and disease resistance.

The genome was sequenced and assembled into 23 contigs totaling 2.3 megabases. Annotation identified over 2,000 protein-coding genes, 10 rRNAs, and 50 tRNAs. Comparison to other P. multocida genomes confirmed the identification as P. multocida and determined it was sequence type 128, which has only been reported once before from a sheep in New Zealand. The genome provides new genomic resources that can improve understanding of P. multocida pathogenesis.

Black rice bran has a number of health benefits and it contains a phytochemical that is associated with a decreased cancer risk. This research aimed to examine the ability of ethanolic extract fractions of ‘Woja Laka’ black rice bran to prevent the growth of liver carcinoma HepG2 cells. Ethanolic extracts of ‘Woja Laka’ and ‘IR 64’ (white rice) bran were fractionated by preparative thin layer chromatography. The cytotoxic activities of the fractions were conducted on HepG2 cell lines by using the MTT assay. When the IC50 value of the fraction showed lower than 500 μg/ml, it was tested for anti-proliferation using the doubling time method. Apoptotic induction and cell cycle arrest were measured by a flow cytometry method. The selectivity cells respon was performed on Vero cells. Result of this study showed that six fractions (F) of F1, F2, F3, F4, F5 and F6 from the ‘Woja Laka’ and three fractions of F1, F2 and F3 from the ‘IR 64’ were examined for their preventive activities on HepG2 cells. The selected fractions F3 and F6 from ‘Woja Laka’ showed highly cytotoxic activity (184.35 ± 16.81 and 283.64 ± 58.13 μg/ml respectively) on HepG2 cells. Furthermore, the fractions of the white rice bran extract showed IC50 > 500 μg/ml. We found that the fractions F3 and F6 from ‘Woja Laka’ were able to inhibit cell proliferation, induce apoptosis and cause G0/G1-phase arrest in the HepG2 cells, while the Vero cells were less responsive to these fractions (IC50 > 470 μg/ml). This study investigated the protective effects of Salacia oblanga and quercetin on cyclophosphamide-induced chromosome aberrations in rat bone marrow cells. Rats were treated with Salacia oblanga or quercetin for 15 days, then given cyclophosphamide on days 14 and 15. Cyclophosphamide is known to induce chromosome aberrations and oxidative stress. The study found that quercetin completely prevented cyclophosphamide-induced chromosome aberrations, while Salacia oblanga partially prevented them. Both treatments decreased oxidative stress caused by cyclophosphamide. Plant growth-promoting rhizobacteria (PGPR) facilitate the plant growth and enhance their
induced systemic resistance (ISR) against a variety of environmental stresses. In this study,
we carried out integrative analyses on the proteome, transcriptome, and metabolome to investigate
Arabidopsis root and shoot responses to the well-known PGPR strain Paenibacillus
polymyxa (P. polymyxa) E681. Shoot fresh and root dry weights were increased, whereas root
length was decreased by treatment with P. polymyxa E681. 2DE approach in conjunction
with MALDI-TOF/TOF analysis revealed a total of 41 (17 spots in root, 24 spots in shoot)
that were differentially expressed in response to P. polymyxa E681. Biological process- and
molecular function-based bioinformatics analysis resulted in their classification into seven different
protein groups. Of these, 36 proteins including amino acid metabolism, antioxidant,
defense and stress response, photosynthesis, and plant hormone-related proteins were upregulated,
whereas five proteins including three carbohydrate metabolism- and one amino
acid metabolism-related, and one unknown protein were down-regulated, respectively. A good
correlation was observed between protein and transcript abundances for the 12 differentially
expressed proteins during interactions as determined by qPCR analysis. Metabolite analysis
using LC-MS/MS revealed highly increased levels of tryptophan, indole-3-acetonitrile (IAN),
indole-3-acetic acid (IAA), and camalexin in the treated plants. Arabidopsis plant inoculated
P. polymyxa E681 also showed resistance to Botrytis cinerea infection. Taken together these
results suggest that P. polymyxa E681 may promote plant growth by induced metabolism and
activation of defense-related proteins against fungal pathogen.

There is increasing confidence that cell therapies will soon play a role in the treatment of autoimmune disorders, but the extent of this impact remains to be seen. Early readouts on autologous CAR-Ts in lupus are encouraging, but manufacturing and cost limitations are likely to restrict access to highly refractory patients. Allogeneic CAR-Ts have the potential to broaden access to earlier lines of treatment due to their inherent cost benefits, however they will need to demonstrate comparable or improved efficacy to established modalities.
In addition to infrastructure and capacity constraints, CAR-Ts face a very different risk-benefit dynamic in autoimmune compared to oncology, highlighting the need for tolerable therapies with low adverse event risk. CAR-NK and Treg-based therapies are also being developed in certain autoimmune disorders and may demonstrate favorable safety profiles. Several novel non-cell therapies such as bispecific antibodies, nanobodies, and RNAi drugs, may also offer future alternative competitive solutions with variable value propositions.
Widespread adoption of cell therapies will not only require strong efficacy and safety data, but also adapted pricing and access strategies. At oncology-based price points, CAR-Ts are unlikely to achieve broad market access in autoimmune disorders, with eligible patient populations that are potentially orders of magnitude greater than the number of currently addressable cancer patients. Developers have made strides towards reducing cell therapy COGS while improving manufacturing efficiency, but payors will inevitably restrict access until more sustainable pricing is achieved.
Despite these headwinds, industry leaders and investors remain confident that cell therapies are poised to address significant unmet need in patients suffering from autoimmune disorders. However, the extent of this impact on the treatment landscape remains to be seen, as the industry rapidly approaches an inflection point.
