This document describes an experiment involving the preparation and analysis of elastic protease from pancreas. It includes three parts: (1) the preparation of elastic protease through a multi-step process involving pancreas activation, plasma separation, adsorption, and elution; (2) the definition of enzyme activity as the amount hydrolyzing 1.0 mg of Congo red elastin within 20 minutes at pH 8.8 and 37°C; and (3) the analysis of enzyme activity through preparation of a reference substance, standard curve, and measurement of sample absorption to calculate relative enzyme activity and total enzyme recovery yield.
