armour. We will set off in 1190, July 4 on our long journey, from Vezelay to our Holy

Philippe thought that i was going to refuse to marry his half sister Alais. It is true,

Experiment 3: Characterization of nonsense mutation in E. coli
A.Statement of Purpose:
The purpose of this experiment was to determine whether a point mutation on the lac
strain, CSH61, from the previous experiment is suppressed by amber suppressors,
SuI, SuII, and SuIII. If the point mutation on the lac strain was a nonsense mutation,
a second mutation on the tRNA for that codon would be able to restore function of
the lac gene by inserting an appropriate amino acid at the stop codon instead of
terminating protein production. Therefore, the CSH61 cells could grow on lactose. If
the point mutation was missense, no growth would be able to occur as the lac gene
would not have its function restored.
This experiment was carried out by mating Hfr lac mutants (CSH61) to three F
strains (CSH54, CSH55, and CSH56) with deletions in the lac pro region, each with
a different suppressor and a type of antibiotic resistance. Mating would allow for

The presence of colonies on all of these plates indicated that conjugation was
successful and the F strains received lac /pro+ as they were able to grow on
antibiotic minimal media plates. However, none of the exconjugants were able to
grow on minimal media plates with lactose and grew as white colonies on
MacConkey s agar, indicating that they were still lac . This result indicated that the
point mutation in the CSH61 mutant strain was unable to be reverted by supD, sup
E, or supF. This meant that neither serine, glutamine, nor serine were appropriate
amino acid replacements to restore function. Had they been sufficient to suppress the
point mutation, growth on the min+lac plates and pink colonies on MacConkey s
agar would be expected like the lac+ control colony
