He described a "Three Peak Challenge" approach using NaCl/PEG size selection, bead beating, and MetaPolyzyme lysis to generate overlapping read length distributions up to 778kb. This method enabled assembly of genomes from a mock community at different abundance levels, including detection over 6 logs of abundance. Future work includes applying these methods to clinical samples like fecal microbiome transplants.
