ABSTRACT- Lung cancer is the most common cause of cancer related mortality worldwide. The epidermal-growth-factor receptor (EGFR) cascades the signaling pathway that regulates tumor-cell proliferation, invasion, angiogenesis, metastasis, and apoptosis. Since EGFR is often over-expressed in NSCLC and the level of EGFR expression correlates with poor prognosis. EGFR inhibitors have been developed as a novel therapy for non-small-cell lung cancer (NSCLC). Gefitinib is the first molecular targeted agent approved for the treatment of advanced NSCLC. It is a highly effective EGFR TK inhibitor (TKI) selectively blocks the signal transduction pathways implicated in cancer growth. Vegf expression in hrf
This study analyzed the expression of angiogenic factors in retinoblastoma tumors with high-risk features (HRF) compared to those without. The results showed that HRF tumors expressed significantly higher levels of VEGF and VEGFR2 than non-HRF tumors. Staining indicated the VEGF was secreted by tumor cells. Neovascularization was also higher in the iris of HRF tumors. Expression of VEGF correlated with the stem cell marker SOX2, suggesting HRF tumors are more stem-like. The conclusions were that HRF tumors may regulate invasiveness through a VEGF feedback loop, and anti-VEGF therapy may be a promising treatment for retinoblastoma due to these findings.

Vegf expression in hrf
This study analyzed the expression of angiogenic factors in retinoblastoma tumors with high-risk features (HRF) compared to those without. The results showed that HRF tumors expressed significantly higher levels of VEGF and VEGFR2 than non-HRF tumors. Staining indicated the VEGF was secreted by tumor cells. Neovascularization was also higher in the iris of HRF tumors. Expression of VEGF correlated with the stem cell marker SOX2, suggesting HRF tumors are more stem-like. The conclusions were that HRF tumors may regulate invasiveness through a VEGF feedback loop, and anti-VEGF therapy may be a promising treatment for retinoblastoma due to these findings.

ABSTRACT:
Anti-VEGF agents such as Bevacizumab (Avastin), Ranibizumab (Lucentis), and Aflibercept (Eylea) have revolutionized the care of a number of retinal diseases such as diabetic macula edema, proliferative diabetic retinopathy, retinal vein occlusions, and exudative age-related macular degeneration. In this talk, I focus on the practical steps and considerations of administering intravitreal injections, and I show a video of how I perform the procedure in my clinic.

This study examined the effects of topical vascular endothelial growth factor (VEGF) on post-surgical tracheal healing in rabbits. Rabbits underwent tracheal surgery using various reconstruction materials, including autograft patches, xenopericardium patches, intraluminal stents, and controls. Half of the reconstruction materials were pretreated with VEGF while the other halves received saline. The study found that autograft repair induced the least granulation tissue and fibrosis. Pretreating materials with VEGF reduced postoperative fibrosis and granulation tissue, except in stents which rapidly caused airway obstruction. VEGF staining suggested local VEGF production over time in addition to the applied topical VEGF.

VEGF-C and VEGF-D are the two central signaling molecules that stimulate the development and the growth of lymphatic system. Both belong to the VEGF protein family which plays important roles in the growth of blood vessels (angiogenesis) and lymphatic vessels (lymphangiogenesis). In mammals the VEGF family comprises five members: VEGF, PlGF, VEGF-B, VEGF-C and VEGF-D. The family was named after its first discovered member VEGF (“Vascular Endothelial Growth Factor”). VEGF-C and VEGF-D form functionally and structurally a subgroup within this family. They differ from the other VEGFs by their peculiar biosynthesis: they are produced as inactive precursors and need to be activated by proteolytic removal of their long N- and C-terminal propeptides. Unlike the other VEGFs, VEGF-C and VEGF-D are direct stimulators of lymphatic growth. They exert their lymphangiogenic function via VEGF receptor-3, which is expressed in the adult organism almost exclusively on lymphatic endothelial cells. In this review we give an overview of the VEGF protein family and their receptors with the emphasis on the lymphangiogenic VEGF-C and VEGF-D, and we discuss their biosynthesis and their role in embryonic lymphangiogenesis.
EWMA 2014 - EP425 COMPARATIVE STUDY OF HUMAN FULL THICKNESS SKIN GRAFT SURVIV...
This study compared the effect of applying human adipose-derived stem cells (ADSCs) by different methods on the survival of human full-thickness skin grafts in nude mice. ADSCs were applied either by intrafascial injection under the skin graft or by topical application on the skin graft. The results showed that intrafascial injection of ADSCs led to the highest skin graft survival rate of 100% and shortest time for engraftment of 7 days. Topical application of ADSCs also improved graft survival to 75% and time to engraftment compared to the control group. Histological and molecular analysis revealed that intrafascial injection of ADSCs promoted the most angiogenesis, collagen formation
NFAT/VEGF Pathway, Griselle, Jocelyn
Jocelyn Griselle is interning at Loughborough University researching heart development pathways. Her project focuses on modeling the NFAT/VEGF protein interaction pathway, which regulates heart development and disease. The pathway involves VEGF activating calcium influx and the Ca2+/Calcineurin/NFAT and DSCR1/NFAT interactions that control VEGF expression levels. Her model divides the pathway into three parts modeled with ODEs in Matlab and SBML in COPASI. The model will help understand how tightly controlling VEGF levels is important for valve development and disease like Tetralogy of Fallot. Further work includes integrating the submodels and performing parameter analysis and experiments to improve accuracy.

Nevertheless, renal side effects, such as proteinuria and thrombotic microangiopathy, have been reported. The underlying physiopathological mechanisms remain unclear, but animal models and clinical similarities with preeclampsia suggest that such therapies affect the function of the endothelial and epithelial layers of the glomerular basement membrane, with activation of the endothelin signaling system and loss of glomerular slit diaphragm integrity. Abstract—
Aim: The aim of the present work was to evaluate associations between the risk of endometriosis and -460C/T (rs833061) and +405G/C (rs2010963) polymorphisms in the VEGF gene.
Methodology: In the present study, we examined group of 100 patients with endometriosis and 100 controls. Genomic DNA was extracted from peripheral blood. Determination of genes polymorphic variants was made using polymerase chain reaction-restriction fragment length polymorphism technique (PCR-RFLP).
Results: Presented study showed statistically significant increase in the endometriosis development risk for the -460T/T genotype (OR 3.39; 95% CI, 1.60-7.13; p = 0.002) and for the -460T allele (OR 2.49; 95% CI, 1.64-3.78; p <.0001), as well as for the +405C/C genotype (OR 2.16; 95% CI, 1.047-4.48; p = 0.035) in patients with endometriosis in comparison with healthy control group. We also observed positive association of the +405C/C genotype (OR 0.26; 95% CI, 0.08-0.79; p = 0.019) as well as the +405C allele occurrence with an increased endometriosis development risk (OR 0.31; 95% CI, 0.19-0.71; p = 0.005), assessed by the degree of rASRM classification stages.
Conclusion: The results support the hypothesis that the -460C/T and +405G/C polymorphisms of the VEGF gene may be associated with endometriosis occurrence in Poland.

1) Preeclampsia is a hypertensive disorder of pregnancy defined as new onset high blood pressure and proteinuria after 20 weeks of gestation. It affects 5-8% of pregnancies and can range from mild to severe.
2) Women who develop preeclampsia have higher levels of soluble fms-like tyrosine kinase 1 (sFlt1), an anti-angiogenic factor, earlier in pregnancy which may contribute to reduced placenta perfusion.

OVEREXPRESSION OF VEGF AND VEGFR2 IN CHRONIC HEPATITIS AND LIVER CIRRHOSIS
VEGF (vascular endothelial growth factor) and the receptor for VEGF- Flk-1 (fetal liver kinase 1) are important players of normal and pathologic angiogenesis. Also, it was proved that they are involved in tumor progression and metastasis in many tumors types by overexpression in cancer cells. Liver malignances and premalignant lesions represent controversial issues concerning VEGF and VEGFR2 (vascular endothelial growth factor receptor 2) expression and their potential involvement in the progression of inflammatory and cirrhotic lesions and also in malignant transformation is virtually unknown. The aim of this work was to describe the differentiate expression and distribution of VEGF and VEGFR2 in chronic hepatitis and liver cirrhosis, and according to these findings to better characterize the molecular profiling of liver disease with malignant transformation potential. We investigated 20 cases with chronic hepatitis and cirrhosis on specimens taken during surgery. Immunohistochemistry was performed in all cases for VEGF, VEGFR2, and FVIII related antigen (Von Willebrand factor). We found significant correlation between HAI (histological activity index) value, VEGF and VEGFR2 expression and factor FVIII related antigen in central part of specimens with chronic hepatitis. Liver cirrhosis lacks this correlation. Our findings suggested that VEGF dependent angiogenesis is more active in chronic hepatitis in the center of the lesion compared with cirrhosis where MVD (microvessel density) is higher at the periphery of the nodules. We hypothesize that the involvement of VEGF and VEGFR2 complex in development of chronic hepatitis and liver cirrhosis could be considered for the use of anti VEGF antibodies as adjuvant therapy in early stages of these diseases.

Fibroblasts migrate in and produce collagen, forming scar tissue through angiogenesis, fibroblast migration and proliferation, extracellular matrix deposition, and remodeling. Vascular endothelial growth factor is important for angiogenesis and permeability while transforming growth factor promotes fibroblast migration.

Cox modeling displayed that 5YS of LCP significantly depended on: N0-N12, T3-4, blood cell circuit, cell ratio factors (ratio between cancer cells-CC and blood cells subpopulations), LC cell dynamics, recalcification time, heparin tolerance, prothrombin index, protein, AT, procedure type (P=0.000-0.031). Neural networks, genetic algorithm selection and bootstrap simulation revealed relationships between 5YS and N0-12 (rank=1), thrombocytes/CC (rank=2), segmented neutrophils/CC (3), eosinophils/CC (4), erythrocytes/CC (5), healthy cells/CC (6), lymphocytes/CC (7), stick neutrophils/CC (8), leucocytes/CC (9), monocytes/CC (10). Correct prediction of 5YS was 100% by neural networks computing (error=0.000; area under ROC curve=1.0).
