Automated single cell tracking was performed on A549 non-small cell lung cancer cells using the Operetta High Content Imaging System. Live-cell imaging was conducted every 5-10 minutes for up to 6 hours, acquiring two brightfield images at different focal planes. Digital phase contrast imaging allowed non-toxic labeling-free observation of cell migration. Using segmentation and tracking software, individual cell movements were quantified over time, showing that stimulation with EGF increased A549 cell migration compared to unstimulated cells.
