3. Baltymų klasifikacija
Pagal cheminę sudėtį skirstomi į
paprastuosius (proteinus) ir
sudėtinius (proteidus).
▲ Paprastiems baltymams
priskiriami tik tie baltymai, kurie
sudaryti vien tik iš aminorūgščių.
▲ Sudėtiniams baltymams
priskiriami baltymai, turintys
nepeptidinę dalį, kaip pavyzdžiui:
lipoproteinai turi lipidinę grupę,
glikoproteinai – angliavandenio

4. Sudėtiniai baltymai
Sudėtinių baltymų struktūroje aptinkama
nebaltyminė dalimis.
▲ Jei ši dalis yra tvirtai prijungta prie
molekulės ir baltymui atliekant savo
funkcijas ji neatsiskiria, tai ši grupė
vadinama prostetine grupe. Pvz., hemo
prostetinė grupė hemoglobine.
▲Jei, baltymui atliekant savo funkcijas,
nebaltyminė dalis tai prisijungia, tai
atsiskiria nuo baltymo, ji vadinama
kofaktoriumi. Kofaktorius – į fermento
sudėtį įeinanti nebaltyminės kilmės
nedidelės molekulinės masės organinė
medžiaga ar jonas, būtina fermento
veikimui. Jei kofaktorius yra organinės
kilmės, jis vadinamas kofermentu
(coenzyme).
5. Sudėtiniai baltymai
Baltymas be savo prostetinės
grupės arba kofermento
vadinamas apoenzimu
(apofermentu), o baltymas su
prisijungusia grupe –
holoenzimu (holofermentu).

Pagal struktūrą, t.y.
polipeptidinių grandinių
skaičių skirstomi į:

nuo terpės pH. Tirpalo pH reikšmė,
kuriai esant dalelė netenka krūvio,
t.y., tampa neutrali, vadinama
izoelektriniu tašku (pI). Tai reiškia,
jog aminorūgštyje yra vienodas
teigiamų ir neigiamų jonų krūvių
skaičius.
▲ Paprastai gamtoje būna tik L-
konfigūracijos aminorūgštys.
20. Erdvinė struktūra
Baltymų erdvinė struktūra susideda
dažniausiai iš trijų lygmenų (nors gali būti ir
keturi):
▲ Pirminė struktūra - tai aminorūgščių seka,
kurią koduoja DNR ir kuri susijungusi
peptidiniais ryšiais.
▲ Antrinė baltymo struktūra susidaro tada,
kai polipeptidinė aminorūgščių grandinėlė
susiveja vandenilinių ryšių dėka ir erdvėje
sudaro dviejų tipų struktūras: α spirales ir β
klostes.
▲ Tretinė baltymo struktūra susidaro
susilanksčius antrinei baltymo struktūrai į
įvairios formos molekules, kurias stabilizuoja
vandeniliniai, joniniai, kovalentiniai ryšiai bei
hidrofobinė sąveika.
▲ Ketvirtinė baltymo struktūra susidaro
susijungus kelioms polipeptidinėms
grandinėms.
21. α spiralės ir β klostės
▲ α-spiralės susidaro taip:
vandeniliniais ryšiais kiekviena
karbonilo (>C=O) grupė
susijungia su kas ketvirta po jos
einančia amino (>N-H) grupe.

lygiagrečiai sugulus dviem ir

Šoninės aminorūgštys skirstomos į grupes:

• disocijuoti ir įgauti neigiamą krūvį (pvz., asparto rūgštis) -COOH virsta į -COO- ;
• prisijungti protoną ir įgauti teigiamą (pvz., lizinas) krūvį -NH2 virsta -NH3

▲ Nežymiai polinės (turinčios -OH ir -NH2 grupes, kurios turi dalinius teigiamus arba
dalinius neigiamus krūvius (pvz., treoninas).
▲ Nepolinės (hidrofobinės) – šoninėse grupėse turi aromatinę grupę (pvz.,
tirptofanas) arba yra sudarytos iš alifatinių (neturinčių ciklo) dalių (pvz., izoleucinas).
▲ Tio grupė (-SH) grupės – sudaro nepolines molekulių dalis, vadinamas disulfidiniais
tilteliais (-S-S-). Svarbiausia aminorūgštis, turinti -SH grupę, yra cisteinas.

jo atliekamas funkcijas. Toks
baltymas, kuris turi natūralią
konformaciją, t.y., atlieka jam
būdingas funkcijas, vadinamas
natyviu.

organiniais tirpikliais, įvyksta jo
dentaūracija. Denatūruotas
baltymas praranda funkcinį

28. Baltymų gryninimo metodai
1.Išsūdymas
Išsūdymas remiasi jų išskirtiniu tirpumu įvairios
koncentracijos druskų tirpaluose. Dažniausiai
išsūdyti naudojamos įvairios amonio sulfato
(NH4)2SO4 koncentracijos.
29. Dializė atliekama naudojant pusiau pralaidžias
membranas, kaip pavyzdžiui, celofaną. Tokios
membranos praleidžia tik tam tikro dydžio
molekules, o didesnės yra sulaikomos.

Elektroforezė
SDS-PAGE
SDS-PAGE – tai natrio
dodecilsulfato poliakrilamidinio
gelio elektroforezė, kuri atliekama
denatūruojančiomis sąlygomis.
NDS yra anijoninis detergentas,
kuris suteikia baltymams neigiamą
krūvį. Tad paveikus baltymą SDSu,
jis tampa neigiamas.
PAGE atskiria skirtingus baltymus
pagal jų molekulinę masę.
31. Norint atlikti baltymų išgryninimą elektroforezės metodu, būtina sukurti
denatūravimo terpę.
Kaip buvo minėta praeitoje skaidrėje, viena iš medžiagų, denatūruojančių
baltymus, yra SDS.
Šioje PAGE reakcijoje taip pat naudojami:
EDTA – medžiaga, prisijungianti divalenčius katijonus, todėl sumažina
proteazių, kurių kofaktoriai yra tokie divalenčiai metalai kaip magnis ir kalcis,
aktyvumą.
Tris buferis reikalingas pastovaus pH palaikymui.
Glicerolis – alkoholis, kuris neleidžia mėginiui iškilti į paviršių iš šulinėlio.
Bromfenolio mėlis yra dažas, kurio dėka galime sekti elektroforezės eigą.
Ditiotreitolis (DTT) yra reduktorius, kuris redukuoja disulfidines jungtis tarp
cisteino molekulių.

Jonų mainų chromatografija remiasi baltymų krūvių
skirtumu. Reakcijoje naudojami polimerai, turintys
skirtingo krūvio funkcines grupes. Skiriami teigiamą
krūvį turintys polimerai ir neigiamą krūvį turintys
polimerai.

4.Chromatografija
Afininėje (giminingumo) chromatografijoje panaudojama
baltymų savybė sąveikauti su skirtingais ligandais, kurie yra
imobilizuoti prie kieto nešiklio. Ligandai gali būti substratas arba
kofermentas. Prie ligandų specifiškai jungiasi tik baltymai. Visi kiti
baltymai patenka į eliuatą (skystį, išsiskiriantį reakcijos metu).
