3. TITLE OF RESEARCH:- BIOCHEMICAL ANALYSIS FOR LODGING TOLERANCE IN BUCKWHEAT
PROBLEM ETERNAL :-
Lodging causes a drastic reduction in grain yield and quality of wheat,
Leads to lower profitability.
Can occur at any time from the booting stage to harvesting stage (i.e., Period of 60 days).
HYPOTHESIS:-
Buckwheat grows well under a wide range of conditions.
Tends to lodge when subjected to high winds or heavy rains and when grown on very fertile soils.
Lignin, cellulose and hemicellulose, provide stem rigidity and strength.
Lignin and cellulose are the dominant components in common buckwheat hull.
The content of lignin and cellulose reached 31.6% and 35.6% in research (2019).

TECHNICAL PROGRAMME
Location of study:-
The present study entitled “BIOCHEMICAL ANALYSIS FOR LODGING TOLERANCE IN BUCKWHEAT”
shall be caried at:
Sampling of selected portion of stem of shoots of 120 genotypes of diverse Buckwheat germplasm to be grown in
PVC pipes at Green house, Division of Genetics & Plant Breeding, FoA, Wadura, SKUAST-Kashmir.
Genomics and Biochemical analysis will be undertaken in the Proteomics Laboratory, Division of Plant
Biotechnology SKUAST-K, Shalimar.

Treatment details:
5. Experimental setup:-
Experiment 1: Column culture experiment in Green house:-
The plants shall be grown in PVC root columns of dimensions 1.3meter height and 20 cm internal diameter. Initially four
each genotype shall be sown after surface sterilization with 10%NaoCL for 5 minutes and subsequent rinsing by distilled
water. At the first trifoliate leaf stage only two competitive plants per column shall be maintained . drought will be imposed
at first fully expanded trifoliate leaf stage by withholding water in drought treatment while as irrigated treatment will be
regularly watered. The root and shoot will be harvested after forty-eight day of sowing.
Observations to be recorded:-
A. Shoot traits
*shoot length(cm) : the length of the longest shoot from the base of stem .
*Shoot volume(cm):volume of shoot mass shall be measured as the deviation of .
volume in a measuring cylinder and converted into a volume .
*Shoot Biomass(g): weight of total shoot biomass.
*No of leaves per plant: the total no of leaves in a plant and their dimension .
*Root shoot biomass ratio: the ratio of root biomass to shoot biomass
6. *Tissue mass density(g/cm): the ratio of shoot biomass to shoot volume.
*Stem dimension: The diameter of the stem.
B. Physiological parameter:
* canopy temperature depression(C)measured as the difference between surface temperature and then canopy
temperature. It shall be measured at two stages viz, second and fourth week after imposition of stress between 10 am to 2
pm using a thermal imager.
7. Experimental 2:In vitro study
In laboratory in-vitro studies shall be carried out to study the various shoot traits for 120 genotypes.
Following data shall be recorded from laboratory experiment
*Cellulose: Differential Grain Cellulose (%)estimated by Gravimetric method of Hasnen and Moller(1975).
Chemical composition :Acetic Acid, Nitric Acid.
*Lignin: Differential Grain Lignin content (mg/g) estimated by Acetyl Bromide method proposed by Hatfield el
al in 1999.
Chemical composition : AcBr, Phosphate buffer, Glacial Acetic acid, Sodium hydroxide, Hydroxylamine-
hydrochloric acid.
*Pectin: Differential Grain pectin content(mg/g) estimated by Gravimetric method given by Emmet and carre in
1926.
Chemical composition: Hydrochloric acid, sodium hydroxide, Acetic acid, Calcium chloride .
