This study evaluated the efficacy of Circovac® vaccination in piglets on an Austrian farm with a history of PCV2 infection. 1105 piglets were randomly allocated to a vaccinated or control group. Vaccinated piglets showed significantly higher average daily weight gain, lower rates of wasting and mortality compared to controls. Laboratory results found vaccination reduced PCV2 viremia and shedding over time, lowering viral pressure on the farm. The study demonstrated Circovac® vaccination effectively improved piglet performance and health.

This study compared the effects of vaccinating pigs at 3 weeks of age with Ingelvac CircoFLEX, Ingelvac PRRS MLV, or both vaccines concurrently, versus a non-vaccinated control group. Over 27,500 pigs were monitored through the nursery and finishing phases. Vaccination had no negative impacts in the nursery. In finishing, pigs vaccinated with both vaccines had significantly reduced cull/mortality rates and increased percentage of pigs marketed, compared to the non-vaccinated controls. Vaccination helped protect against PCV2 and PRRSv exposure and associated respiratory disease in finishing pigs.

Background. Respiratory syncytial virus (RSV) is a leading cause of infant morbidity and mortality. A recombinant RSV fusion protein nanoparticle vaccine (RSV F vaccine) candidate for maternal immunization was tested for safety and immunogenicity in women of childbearing age. Conclusions. The vaccine appeared safe, immunogenic, and reduced RSV infections. Further development as a vaccine for use in maternal immunization is warranted.

Researchers are looking for environmentally sustainable feeding strategies that will lower diet cost, agricultural use of land, water depletion, and climate change impact. We used linear models to formulate 4 single-objective diets including least-cost, least-land use, least-water depletion, and least-climate change impact diets. Preliminary results showed that the use of wheat and wheat middlings hold potential to reduce pig diet cost and the environmental footprint.

They administered 4 live hot IBD vaccinations but saw variable egg production. In 2010, they started vaccinating flocks with Vaxxitek HVT+IBD which improved overall bird health, achieved good uniform body weights, and allowed removal of the 4 on-farm live IBD vaccinations. Data from 2012 showed improved bodyweights, uniformity and laying performance since adopting the Vaxxitek vaccination.

A field study was conducted in France to compare the laying performance of commercial brown layers vaccinated with Vaxxitek HVT+IBD versus controls vaccinated with a live intermediate IBD vaccine. Over 250 birds per group were reared on a pullet and layer farm and housed in enriched cages. Results showed that the Vaxxitek group had a 3.88 egg/hen higher laying rate and higher average egg weight than the controls. Growth was also improved in the Vaxxitek group as they had a higher body weight at 5 weeks of age. In conclusion, Vaxxitek vaccination benefits included higher pullet weight at point of lay, improved laying performance and egg quality.

This highlights the importance of including both ORF1 and ORF2 in PCV2 vaccine design.

Cell Therapy Expansion and Challenges in Autoimmune Disease

There is increasing confidence that cell therapies will soon play a role in the treatment of autoimmune disorders, but the extent of this impact remains to be seen. Early readouts on autologous CAR-Ts in lupus are encouraging, but manufacturing and cost limitations are likely to restrict access to highly refractory patients. Allogeneic CAR-Ts have the potential to broaden access to earlier lines of treatment due to their inherent cost benefits, however they will need to demonstrate comparable or improved efficacy to established modalities. In addition to infrastructure and capacity constraints, CAR-Ts face a very different risk-benefit dynamic in autoimmune compared to oncology, highlighting the need for tolerable therapies with low adverse event risk. CAR-NK and Treg-based therapies are also being developed in certain autoimmune disorders and may demonstrate favorable safety profiles. Several novel non-cell therapies such as bispecific antibodies, nanobodies, and RNAi drugs, may also offer future alternative competitive solutions with variable value propositions. Widespread adoption of cell therapies will not only require strong efficacy and safety data, but also adapted pricing and access strategies. At oncology-based price points, CAR-Ts are unlikely to achieve broad market access in autoimmune disorders, with eligible patient populations that are potentially orders of magnitude greater than the number of currently addressable cancer patients. Developers have made strides towards reducing cell therapy COGS while improving manufacturing efficiency, but payors will inevitably restrict access until more sustainable pricing is achieved. Despite these headwinds, industry leaders and investors remain confident that cell therapies are poised to address significant unmet need in patients suffering from autoimmune disorders. However, the extent of this impact on the treatment landscape remains to be seen, as the industry rapidly approaches an inflection point.
