Differential Media
Assay Media
Media for Enumeration of bacteria
Media for Characterization of Bacteria
Maintenance Media
Solid and Semisolid Media
6. Selective Media
These media provide nutrients that
enhance the growth and predominance of a
particular type of bacterium but do not
enhance other types of organism that may
be present.
7. Types of Selective Media
There are some types of selective media-
Eosin Methylene Blue(EMB) agar based Selective Media
MacConkey agar based Selective Media
Mannitol salt agar based selective Media
Phenyl Ethyl Alcohol (PEA) agar based Selective Media
8. EMB agar based Selective Media
Eosin Methylene Blue agar (EMB agar) contains the dyes
eosin and methylene blue. They inhibit Gram-positive
organisms. Such a medium is selective for Gram-negative
species. Lactose-fermenting organisms such as S.
enterica produce a black precipitate on EMB.

9. MacConkey agar based Selective Media
MacConkey agar is similar to EMB agar in that it is also
selective for Gram-negative species and differential with
respect to lactose fermentation. MacConkey agar is used
for the detection of coliforms and enteric pathogens based
on their ability to ferment lactose.

10. Mannitol agar based Selective Media
Mannitol salt agar (7.5% NaCl) medium is a selective for
staphylococci and differential with respect to mannitol
fermentation. Growth of most bacteria without
staphylococci, is inhibited by the high concentration of salt
in the medium.

11. PEA agar based Selective Media
Phenyl ethyl Alcohol (PEA) agar is a selective medium
which inhibits the growth of most Gram negative
organisms. Phenyl ethyl alcohol acts to disrupt lipid
structure in the membrane of Gram negative organisms as
well as inhibiting protein synthesis.
