3. PRINCIPLE
Acid treatment denatures fragmented DNA
The lysing solution removes most of the nuclear
proteins
In the absence of massive DNA breakage produces
nucleoids with large halos of spreading DNA loops,
emerging from a central core.
The nucleoids from spermatozoa with fragmented
DNA either do not show a dispersion halo or the halo
is minimal.

5. The Sperm Chromatin Dispersion Test: A Simple
Method for the Determination of Sperm DNA
Fragmentation
Presence of extensive DNA breaks,
High amounts of ssDNA
Suppressing the generation of DNA dispersion
The results suggest that ssDNA interacts within the
sperm head in such a way that the removal of most
nuclear proteins by lysing solutions does not result in
dispersion of the DNA fragments.
6. SPERM CHROMATIN STRUCTURAL ASSAY
Treatment with an acid solution
That opens up the DNA strand at the breaks or
fragmented regions.
The dye Acridine Orange binds
Double stranded DNA (dsDNA) and single stranded
DNA (ssDNA) and fluoresces green or red
