2. INTRODUCTION Several factors are responsible for the inflamatory responses,likemicrobiological,immunological & toxic agents can induce the inflamation by activating a variety of humoral &cellular mediators. These factors activates lipid synthesis pathways & signaling pathways which results organ dysfunction.
3. FORMATIONOF EICOSANOID Pro inflamatory eicosanoid is produced by the precursor Arachidonic acid(AA). Arachidonic acid(AA) is released from membrane phospholipids by the action of Phospholipase-A2 PLA2.
4. INFLAMATORY MEDIATORS Pro inflamatory eicosanoid is metabolised in to prostaglandins(PGS)& Leucotrienes(LTS) by action of Cycloxygenase(COX) &Lipoxygenase(LO) enzymes,respectively. Faulty activation of COX,LO,PLA2 have been implicated in many inflamatory diseases.
5. Conted…. PLA2 is activated by Phospholipase-A2 activating protien(PLAP)&LO by 5-Lipoxigenase activating protien(FLAP) The inducible form of COX-2 enzyme, s normally absent in basal condition,but get induced in inflamatory state.
