2. INTRODUCTION
What is DNA BARCODING?
DNA barcoding is a method of species identification using a short section of DNA from a
specific gene or genes. DNA barcodes for living organisms are used to provide data
that is critical in helping to understand, conserve, and utilize the world's biodiversity.
3. Objectives:
At the end of the course, we were able to:
● Understand the scientific method
● Plan an effective scientific experiment
● Demonstrate laboratory skills in DNA extraction, PCR,
electrophoresis and bioinformatics.
● Analyze protocol failures and solutions
● Effectively communicate the scientific results to an audience.
4. DNA BARCODING PROCESS
● Collection
● DNA Extraction
● PCR and Gel Electrophoresis
● Sequencing
● Bioinformatics
6. DNA Extraction
A small amount of the plant tissue was utilised for the extraction of the DNA. This
was done by the lysing and grinding of each plant sample to obtain their precipitate.
7. Amplification of DNA by PCR
The DNA was amplified using polymerase chain reaction
(PCR). This process increased the number of copies.
8. Analyze PCR Products by Gel
Electrophoresis
Gel electrophoresis was used to check the size of the copied DNA fragments
and to ensure that there was plenty of good quality DNA.
9. DNA Sequencing
The DNA sample was sent to a sequencing company then was
imported into DNA Subway. This sequence produced a DNA barcode
that was specific to the sample specimen.
12. ANNONA SQUAMOSA
ANNONA SQUAMOSA, also known as
custard apple; is a green, fruit with scaly
skin and sweet, creamy flesh. Custard
apple originates from the West Indies but is
now grown in all tropical regions.
USES:
It is rich in fiber, vitamins, and minerals,
it boosts immunity,
fights inflammation,
It contains magnesium, which helps in
relaxing muscles and protecting the heart
against diseases.
13. PLUMERIA RUBRA
PLUMERIA RUBRA,also known as red
frangipani; a deciduous plant species that
grows as a spreading shrub with flowers. It
is originally found in Colombia,Mexico and
other countries. It is widely cultivated in
other tropical regions.
USES:
Used to scent coconut oil
Used for leis (necklaces)
Remedy for bloating
Used in traditional
chinese medicine
