2. Noggin, which was identified in Xenopus embryos, is
the first molecule shown to have the neural inducing effect
of the organizer center [3]. It is expressed in the organizer
at the gastrula stage and encodes a novel 32 kDa glycoprotein
that is secreted and functions as a homodimer.

3. • Soluble noggin protein induces anterior neural markers in
• animal cap (AC) ectoderm that would otherwise develop
• epidermis [4]. Xu et al. [5] and other researchers have
• shown that inhibition of the BMP signal by a dominant
• negative BMP receptor (DN-BR) elicits neuralization in the
• AC tissue, whereas noggin exerts its neuralizing effect by
• binding and inactivating BMP4 [5–10]. We have previously
• shown that C145S or 3CS mutation leads to loss of noggin’s
• inducing effect [11].

• Maternally encoded noggin may play a role in
dorsal fate specification during Xenopus
embryogenesis as part of the early-acting
Nieuwkoop center . The Nieuwkoop center is
inculcated by endodermal cells of the vegetal
half of the embryo, and it induces dorsal
mesoderm and generation of the Spemann
organizer; noggin can mimic these effects of the
Nieuwkoop center.

5. • Bone morphogenetic protein 4 (BMP-4) induces
• ventral mesoderm but represses dorsal
mesoderm formation
• in Xenopus embryos. We show that BMP-4
inhibits two
• signaling pathways regulating dorsal mesoderm
formation,
• the induction of dorsal mesoderm (Spemann
organizer) and
• the dorsalization of ventral mesoderm.

6. • Ectopic expression of
• BMP-4 RNA reduces goosecoid and forkhead-l transcription
• in whole embryos and in activin-treated animal cap explants.
• Embryos and animal caps overexpressing BMP-4 transcribe
• high levels of genes expressed in ventral mesoderm (Xbra,
• Xwnt-8, Xpo, Mix.1, XMyoD). The Spemann organizer is ventralized
• in these embryos; abnormally high levels of Xwnt-8
• mRNA and low levels of goosecoid mRNA are detected in the
• organizer. In addition, the organizer loses the ability to dorsalize
• neighboring ventral marginal zone to muscle. Overexpression of
• BMP-4 in ventral mesoderm inhibits its response to dorsalization
• signals.

7. • The BMP signaling pathway has also been
implicated in tumorigenesis, highlighting on the
role of the ligands and receptors. A concurrent
survey of gremlin 1 and noggin protein
expression in multiple normal and cancer
samples, availing immunohistochemistry on
tissue microarrays containing 96 samples from 34
different normal organs/tissue sites and 208
samples of 34 different tumor types. Both normal
and tumor samples, gremlin 1 and noggin
expression was negative or weak.
