Additionally, pre-treating cells with testosterone before cytokine exposure continued to decrease CX3CR1 and CCR2 expression levels compared to cytokine treatment alone. Future work will utilize controls to ensure the testosterone effects are receptor-mediated and not due to conversion to estrogen.

The white box model
Standardized cells with completely predictable and controllable behavior is a key focus of synthetic biology. Recent advances such as the implantation of a synthesized genome into a DNA-free bacterial shell at the J. Craig Venter Institute marked the dawn of these whole-genome scale projects. A first step towards predictable synthetic minimal cells is made through further reduction of the cellular complexity by minimizing the bacterial genome. This has lead to a short list of essential genes for cells to still be considered living entities. Although these minimal cells increase our fundamental understanding of key cellular processes, there are other important criteria to consider for synthetic cells to become biotechnologically relevant.
In this colloquium I will address the current state of research concerning biotechnological useful cells that can be used as a platform for operating precisely designed and independent biological systems. Top-down methods for the design of these organisms will be reviewed, highlighting remarkable experiments.

This study examined how chronic physical restraint stress alters gene expression in mouse spleens. Mice were subjected to restraint stress for 12 hours per day for two days. Microarray analysis found that 48 out of 225 apoptosis- and p53 signaling-related genes showed significant expression changes in stressed mice compared to unstressed controls. Four genes (Fas, FADD, p53, p21) were validated with real-time PCR. The results provide evidence that apoptotic pathways contribute to stress-induced lymphopenia and identify potential stress response biomarkers.

The assay was tested on DNA samples from 200 Danes, 100 Somalis, and 100 Greenlanders. Being able to distinguish between individuals with different copy numbers of GSTM1 and GSTT1 allows for investigating whether these genes influence factors like cancer susceptibility or drug response in a dose-dependent manner.

New data from the NASA Wise telescope suggests that the fragments from the Baptistina asteroid that supposedly collided with another asteroid are too young to have killed the dinosaurs. The Baptistina family of asteroids formed only around 80 million years ago, leaving only 15 million years for one of the fragments to travel from the asteroid belt to Earth, making the timing unlikely according to the new evidence.

The assay was tested on DNA samples from 200 Danes, 100 Somalis, and 100 Greenlanders. Being able to distinguish between individuals with different copy numbers of GSTM1 and GSTT1 allows for investigating whether these genes influence factors like cancer susceptibility or drug response in a dose-dependent manner.

My laboratory focused on an enzyme KDM5b (aka PLU-1, JARID1b) that was widely expressed during development and played a key role in progression of breast cancer through HER-2.
My lab focused on understanding the key biochemical activity of the enzyme through dissecting the proteomic and genomic interactors.
Our results were confirmed through the use of ES cells, adult stem cells and mouse models.

This study identified a mosaic activating mutation in the AKT1 gene associated with Proteus syndrome. Researchers sequenced DNA from tissue samples of 29 patients with Proteus syndrome and identified a recurrent somatic mutation, c.49G>A, in the AKT1 gene in 26 patients. This mutation leads to increased phosphorylation and activation of the AKT1 protein. Western blot analysis showed increased phosphorylation of AKT1 in tissues affected by Proteus syndrome. The findings link activation of the AKT1 signaling pathway through somatic mutations to overgrowth observed in Proteus syndrome patients.
Niemann pick disease
Niemann-Pick disease is a rare genetic disorder caused by mutations in either the NPC1 or NPC2 gene that results in the abnormal accumulation of cholesterol and other lipids within cells. There are four variants of the disease (A, B, C1, C2) which are differentiated based on their genetic cause and clinical presentation. The majority of Type C1 cases are due to mutations in the NPC1 gene, while Type C2 results from mutations in the NPC2 gene. Both genes encode proteins that are involved in the intracellular transport of cholesterol, though their precise functions are still being investigated.

I develop the Ruby programming language, RubyGems, and Bundler, which are package managers for Ruby. Today, I will introduce how to enhance the security of your application using open-source software (OSS) examples from Ruby and RubyGems.
The first topic is CVE (Common Vulnerabilities and Exposures). I have published CVEs many times. But what exactly is a CVE? I'll provide a basic understanding of CVEs and explain how to detect and handle vulnerabilities in OSS.
Next, let's discuss package managers. Package managers play a critical role in the OSS ecosystem. I'll explain how to manage library dependencies in your application.
I'll share insights into how the Ruby and RubyGems core team works to keep our ecosystem safe. By the end of this talk, you'll have a better understanding of how to safeguard your code.
Artificial Intelligence for XMLDevelopment
In the rapidly evolving landscape of technologies, XML continues to play a vital role in structuring, storing, and transporting data across diverse systems. The recent advancements in artificial intelligence (AI) present new methodologies for enhancing XML development workflows, introducing efficiency, automation, and intelligent capabilities.
