The Hershey-Chase experiment helped confirm that DNA is the genetic material by showing that the DNA, but not the proteins, of bacteriophages enters host bacterial cells during infection. They uniquely labeled the DNA and proteins of bacteriophages with different radioactive isotopes, then showed that the progeny of infected bacteria contained the radioactivity from the original phage DNA but not proteins. This provided strong evidence that DNA, rather than protein, carries the genetic information required for reproduction.

- DNA was first isolated and characterized in 1868, and in 1944 it was discovered that DNA carries genetic information. Experiments in 1952 confirmed that DNA, not protein, carries genetic material.
- Rosalind Franklin and Maurice Wilkins used X-ray diffraction in the early 1950s to show that DNA has a helical structure with periodicities of 3.4Å and 34Å.

SMART Team Research
The Pittsburg High School SMART Team studied the crystal structure of the Ebola virus glycoprotein (GP) bound to a human survivor antibody. GP consists of two subunits, GP1 and GP2, and mediates host cell attachment and entry. The crystal structure revealed that three GP1 subunits are cradled by GP2 helices in a bowl shape, with the receptor-binding site inside. Three antibody units bind tightly to GP2 via interactions that prevent GP2 from rearranging into its fusion form, neutralizing the virus. Understanding this structure may help develop effective vaccines against Ebola by inhibiting GP's role in the viral life cycle.

This document summarizes the history and development of CRISPR-Cas9 genome editing technology. It discusses how early approaches to targeted DNA cleavage relied on site-specific DNA recognition by oligonucleotides or proteins. Zinc finger nucleases and TAL effector nucleases were developed that allowed for genome editing but were difficult to design and validate. The discovery of CRISPR immune systems in bacteria provided the basis for a new approach using Cas9 guided by CRISPR RNA. Experiments showed Cas9 could be programmed to cleave DNA sequences using a simple RNA guide. A New molecular biology techniques for gene therapy
The CRISPR (clustered, regularly interspaced, short palindromic repeats)-Cas9 (CRISPR-associated protein 9) system is a targeted nuclease technology which allows precise genome editing. Since the discovery of this system there has been great interest in its potential for human gene therapy. The CRISPR-Cas9 system has many advantages in comparison to other targeted nucleases transcription activator-like effector nucleases and zinc finger nucleases. As a relatively new genome editing platform, safety issues such as off-target editing have yet to be fully investigated.This presentation addresses the challenges as well as the socio-ethical considerations that surround the use of human genome editing.
