WHAT IS ORLISTAT?
 drug used in the treatment of obesity.
 Its primary function is preventing the absorption of
fats from the human diet, thereby reducing caloric
intake.
How Does it Work?
 Orlistat works by inhibiting pancreatic lipase, an enzyme that
breaks down triglycerides in the intestine. Without this
enzyme, triglycerides from the diet are prevented from being
hydrolyzed into absorbable free fatty acids and are excreted
undigested
Mechanism of Actions
 Orlistat is a reversible inhibitor of gastrointestinal lipases.
 It exerts its therapeutic activity in the lumen of the stomach and small intestine by
forming a covalent bond with the active serine residue site of gastric and
pancreatic lipases. The inactivated enzymes are thus unavailable to hydrolyze
dietary fat in the form of triglycerides into absorbable free fatty acids and
monoglycerides. As undigested triglycerides are not absorbed, the resulting
caloric deficit may have a positive effect on weight control.
How To Take Orlistat?
 Taken by mouth; With liquid during meals that
contain fat or within 1 hour after a meal; usually
thrice a day

 Pregnancy Factor X
 weight loss offers no potential benefit to a pregnant woman and may result in
fetal harm. A minimum weight gain, and no weight loss, is currently
recommended for all pregnant women, including those who are
already overweight or obese, due to the obligatory weight gain that occurs in
maternal tissues during pregnancy.

Overdose
 Single doses of 800 mg ORLISTAT and multiple doses of up to 400 mg three
times a day for 15 days have been studied in normal weight and obese subjects
without significant adverse findings.
 Should a significant overdose of ORLISTAT occur, it is recommended that the
patient be observed for 24 hours. Based on human and animal studies, systemic
effects attributable to the lipase-inhibiting properties of ORLISTAT should be
rapidly reversible.
