Vibrio Species Isolated from Farmed Fish in Basra City in IraqDrNajimRKhamees
This study was carried out to investigate the occurrence of potentially pathogenic species of Vibrio in seven types of fish sampled from fish farms located in different districts in Basra governorate, Iraq. A total of 153 live fishes was collected from fish farms during the period January till May 2016. Bacteria were isolated using selective medium thiosulfate citrate bile sucrose salt agar. Presumptive Vibrio colonies were identified using the VITEK 2 system and selected biochemical tests. In the present study V. alginolyticus (24 of 60) was the predominant species, followed by V. cholerae (10 of 60), V. furnisii (10 of 60), V. diazotrophicus (7 of 60), V. gazogenes (5 of 60) and V. costicola (4 of 60). The signs of vibriosis appeared in three
types of fish, including Cyprinus carpio, Coptodon zillii and Planiliza subviridis in spite of the using Oxytetracycline in most fish farms. The results of the present study demonstrated the presence of pathogenic Vibrio species nearly in all fish farms. So the farm owners should be concerned about the presence of these pathogenic bacteria which also contributes to human health risk and should adopt best management practices for responsible aquaculture to ensure the quality of fish.

Pathogenic Bacteria in Corals from Veracruz Reef System National Park IJMER
The Veracruz Reef System National Park (PNSAV) is situated in front of Veracruz-Boca del
Río and Antón Lizardo conurbation with approximately 65,516 hectares. In 1992 this reef system was
declared a Natural Protected Area and in 2006 it became Biosphere reserve. Despite this recognition,
the PNSAV is exposed to anthropogenic effects such as river discharge, port traffic, tourism, and waste
discharges; all this causes sediment and bacteria, which affect corals. Studies conducted by PNSAV
show the presence of diseases and syndromes in scleractinian corals but do not denote the causal agent
of the disease. For this reason, the present study aimed to make the diagnosis of bacteria causing
damages to the scleractinian coral and its relation to discharges from areas of influence of PNSAV. 12
fixed 80 meter transects were established and coral samples were collected from the genera:
Colpophyllia, Montastraea, Porites and Siderastrea. The lesions identified as criteria for the collection
were bands, marks and spots on the body of the coral. The sample was run in duplicate, both the affected
and the visually healthy. A microbiological analysis of wastewater from point sources of pollution was
performed. The coral and water samples were analyzed by simple PCR in laboratory of the Instituto
Tecnológico de Boca del Río in Veracruz (ITBOCA). Primers of pathogenic bacteria include species of
the genera Oscillatoria, Vibrio, Serratia y Aurantimonas. There was a positive presence of Vibrio
bacteria with an 87% in the water samples and a 27.1% in coral tissue. The bacteria Serratia
marcescens was only present in water samples and the Aurantimona coralicida in diseased coral tissue.
It was also noted that there is a relationship in bacteria in the positive presence of 3-point sources of
pollution and coral tissue.

Johne's disease (JD) or Paratuberculosis (PTB) has gained a great attention by many industrial countries for its severing economic losses and possibly zoonotic concerns. In the current study conventional clinical and direct microscopic examination compared to real time polymerase chain reaction (RT-PCR) were used to diagnose JD in clinically suspected small ruminants. Clinical examination revealed 130 (8.7%) suspected cases that showed history of emaciation and diarrhea out of the total examined (1500) animals. Direct microscopy of Ziehl-Neelsen (ZN) stained smears (130) revealed 62 (47.7%) acid fast bacteria resembled Mycobacterium avium subsp. paratuberculosis (MAP). RT-PCR insertion sequence gene (IS900) detected MAP in 25 (65.8%) out of 38 fecal samples harbored acid fast bacilli. We concluded and recommended that RT-PCR considers the most rapid confirmatory method for screening and diagnosis of the MAP in comparison to low specific conventional phenotypic methods, which still remained valuable techniques in the diagnosis of JD in developing countries.

The purpose of this study was to investigate species specific inhibitory effects of esDNA isolated from two conspecific organisms: Vibrio parahaemolyticus (VP) and Vibrio harveyi (VH), and to assess the functional role of esDNA to enhance the survival rate of Artemia sp. In an in vitro study, nine doses of Extracellular self-DNA of Vibrio parahaemolyticus (esDNAVP) and Vibrio harveyi (esDNAVH) were used as the target for the challenge test with the conspecific bacteria. In an in vivo study, the protective effect of esDNA was then tested in nauplii of the brine shrimp Artemia at various priming times and concentrations of esDNA under gnotobiotic conditions prior to challenge with VP and VH at the concentration of 5 × 105 CFU mL-1. The results from in vitro study showed that the use of esDNAVP at levels of 24.02 and 48.05 ng µl-1 and esDNAVH at concentrations of 13.33 and 26.67 ng µl-1 were able to inhibit the growth of the conspecific species when added to the culture medium at the concentration level of 5 × 105 CFU mL-1. The results from in vivo study showed that the use of 24.02; 48.05 and 72.07 ng µl-1 of esDNAVP as well as the use of 13.33; 26.67 and 40.00 ng µl-1 of esDNAVH inhibited the growth of VP and VH and enhanced the survival rate of Artemia sp compared to the control treatment (P<0.05). Taken together, we confirmed that esDNA obtained from the extraction and random fragmentation from esDNAVP and esDNAVH, produces a species-specific inhibitory effect on the same species and can serve as a potential alternative strategy for disease control to deliver the functionality of esDNA to the fish and shrimp.
The early mortality syndrome (EMS) in shrimp has been ravaging production systems, spreading vertically in Asia and horizontally to countries as far away as Mexico since first reported in 2009.

Alternative strategies for minimizing the detrimental effects of bacterial infection and prevention of diseases in aquaculture are necessary since the ongoing efficacy of antibiotics is proving to be unsustainable. One of the most promising approach is the use of aqua herbal conditioners to stimulate the immune system of fish to allow them to fight off infections. In this study, the protective effect of aqua herbal conditioners produced from, mainly, mangrove and neem plant extracts in marine fish, was tested on Asian Seabass Lates calcarifer and Silver Pompano Trachinotus blochii at 8-10 g of weight size. Challenge tests were performed by immersion with two pathogenic bacteria: Vibrio harveyi and Vibrio parahaemolyticus, at a concentration of 105 cells ml-1 for 60 minutes after 12 h, 24 h and 36 h conditioning treatment. The experimental trial show that after 72 h, commercially available aqua herbal conditioners (AquaHerb) was able to significantly increase the percentage survival of L. calcarifer and T. blochii and reduces their susceptibilityto the V.harveyi and V.parahaemolyticus. Significantly higher leukocytesnumber, monocyte, neutrophil andphagocyticindexwere detected in all conditioning group for Silver Pompano and Asian Seabass. These results suggest that the combination of herbal extracts together with other trace elements contained in AquaHerb were able to act as immunostimulants and appear to improve the immune status and disease resistance of Asian Seabass and Silver Pompano.

The successful of pregnancy in humans and rodents occur between the interaction maternal and fetal
interface, specially involving the participation of uNK cells. This interaction involved neo angiogenesis,
placentation and presence of mediators like nitric oxide. During the pregnancy the administration of LPS
in the dams can results in necrosis, preterm birth, IUGR, miscarriage or neurological problem. Once the
uNK cells are activated, they can produce vasodilators, like NO. So, the main purpose of this study was
to evaluate if LPS cause alteration in the uNK cells in pregnant mice and if the same behaviour can be
detected by NO in the blood. Also we evaluated the effect of LPS to cause neurological injuries. To do that
we used pregnant mice on gd 10th and those was treated with LPS for different times. Uterine samples
were collected at 0.5,1,2 and 6hr after LPS treated and processed for paraffin embedding and tissue
homogenate. The samples designated for paraffin embedding was performed the Dolichos biflorus (DBA)
lectin cytochemistry and anti-iNOS immunocytochemistry. The samples designated to tissue homogenates
were processed for SDS-PAGE and Western-blot using anti-iNOS and evaluate of NO concentration. We
found after 2h LPS exposure the mice showed fever and low capacity to explore different environment.
At the same time, we found increase in the nitrate/nitrito ratio in a dose dependent manner in the uterus
after 2h LPS exposure.

New Directions in Targeted Therapeutic Approaches for Older Adults With Mantl...i3 Health
i3 Health is pleased to make the speaker slides from this activity available for use as a non-accredited self-study or teaching resource.
This slide deck presented by Dr. Kami Maddocks, Professor-Clinical in the Division of Hematology and
Associate Division Director for Ambulatory Operations
The Ohio State University Comprehensive Cancer Center, will provide insight into new directions in targeted therapeutic approaches for older adults with mantle cell lymphoma.
STATEMENT OF NEED
Mantle cell lymphoma (MCL) is a rare, aggressive B-cell non-Hodgkin lymphoma (NHL) accounting for 5% to 7% of all lymphomas. Its prognosis ranges from indolent disease that does not require treatment for years to very aggressive disease, which is associated with poor survival (Silkenstedt et al, 2021). Typically, MCL is diagnosed at advanced stage and in older patients who cannot tolerate intensive therapy (NCCN, 2022). Although recent advances have slightly increased remission rates, recurrence and relapse remain very common, leading to a median overall survival between 3 and 6 years (LLS, 2021). Though there are several effective options, progress is still needed towards establishing an accepted frontline approach for MCL (Castellino et al, 2022). Treatment selection and management of MCL are complicated by the heterogeneity of prognosis, advanced age and comorbidities of patients, and lack of an established standard approach for treatment, making it vital that clinicians be familiar with the latest research and advances in this area. In this activity chaired by Michael Wang, MD, Professor in the Department of Lymphoma & Myeloma at MD Anderson Cancer Center, expert faculty will discuss prognostic factors informing treatment, the promising results of recent trials in new therapeutic approaches, and the implications of treatment resistance in therapeutic selection for MCL.
Target Audience
Hematology/oncology fellows, attending faculty, and other health care professionals involved in the treatment of patients with mantle cell lymphoma (MCL).
Learning Objectives
1.) Identify clinical and biological prognostic factors that can guide treatment decision making for older adults with MCL
2.) Evaluate emerging data on targeted therapeutic approaches for treatment-naive and relapsed/refractory MCL and their applicability to older adults
3.) Assess mechanisms of resistance to targeted therapies for MCL and their implications for treatment selection

Primary taste sensations: Sweet, Sour, Salty, Bitter, Umami
Chemical basis and signal transduction mechanisms for each taste
Taste Threshold and Index:
Taste threshold values for Sweet (sucrose), Salty (NaCl), Sour (HCl), and Bitter (Quinine)
Taste index relationship: Inversely proportional to taste threshold
Taste Blindness:
Inability to taste certain substances, particularly thiourea compounds
Example: Phenylthiocarbamide
Structure and Function of Taste Buds:
Composition: Epithelial cells, Sustentacular/Supporting cells, Taste cells, Basal cells
Features: Taste pores, Taste hairs/microvilli, and Taste nerve fibers
Location of Taste Buds:
Found in papillae of the tongue (Fungiform, Circumvallate, Foliate)
Also present on the palate, tonsillar pillars, epiglottis, and proximal esophagus
Mechanism of Taste Stimulation:
Interaction of taste substances with receptors on microvilli
Signal transduction pathways for Umami, Sweet, Bitter, Sour, and Salty tastes
Taste Sensitivity and Adaptation:
Decrease in sensitivity with age
Rapid adaptation of taste sensation
Role of Saliva in Taste:
Dissolution of tastants to reach receptors
Washing away the stimulus

Mechanisms behind taste preference and aversion
Influence of receptors and neural pathways
Impact of Sensory Nerve Damage:
Degeneration of taste buds if the sensory nerve fiber is cut
Abnormalities of Taste Detection:
Conditions: Ageusia, Hypogeusia, Dysgeusia (parageusia)
Causes: Nerve damage, neurological disorders, infections, poor oral hygiene, adverse drug effects, deficiencies, aging, tobacco use, altered neurotransmitter levels
Neurotransmitters and Taste Threshold:
Effects of serotonin (5-HT) and norepinephrine (NE) on taste sensitivity
Supertasters:
25% of the population with heightened sensitivity to taste, especially bitterness
