This document describes a method for measuring glycogen hydrolysis using salivary amylase. Glycogen is incubated with diluted saliva for various time periods. The reaction is stopped by adding DNS reagent which detects reducing sugars. Absorbances are read and used to calculate the percentage of glycogen hydrolyzed over time, allowing interpretation of the enzymatic reaction rate. Tube 9 containing acid-hydrolyzed glycogen serves as a positive control representing 100% hydrolysis. Plotting percentage hydrolysis versus time will show the kinetics of glycogen breakdown by salivary amylase.
