1. Metaloproteinases (MMPs) are enzymes that maintain and degrade the extracellular matrix during tissue repair and remodeling. MMPs participate in wound healing, angiogenesis, and cancer metastasis.
2. MMPs are classified into subgroups based on their protein targets, such as collagens, gelatinases, and matrilysins. They have domains for signaling, catalytic activity, and substrate binding.
3. MMP activity is regulated by tissue inhibitors of metalloproteinases (TIMPs) and imbalanced MMP/TIMP expression contributes to cancer progression and metastasis by continuously remodeling the extracellular matrix.

2. Rol de las Metaloproteinasas
Las metaloproteinasas son enzimas fundamentales para el mantenimiento estructural de
la matriz extracelular, así como para su degradación en situaciones donde se requiere un
proceso de reparación tisular. Las metaloproteasas de matriz participan en la curación de
heridas, la angiogénesis y la metástasis de las células tumorales.
Características:
- Tienen la capacidad de eliminar proteínas dañadas y de degradar componentes de
la MEC.
- Pueden ser inhibidas por Inhibidores Tisulares de las Metaloproteinasas (TIMPs).
- En cuanto a su participación en los procesos vasculares, están relacionadas con la
proliferación y apoptosis de las fibras musculares lisas y de las células
endoteliales.
- Las metaloproteinasas, tienen la capacidad de intervenir en el desarrollo de las
células durante la embriogénesis.
Estructura y clasificación:
Es posible agrupar las distintas MMPs en seis subfamilias diferentes como colagenasas,
estromalisinas, elastasas, gelatinasas, metaloproteinasas asociadas a la membrana
plasmática y matrilisinas. Su clasificación depende de la proteína diana de cada enzima
particular.
3. Estructura de las MMPs
1. Péptido señal o predominio: necesario para el desplazamiento intracelular
2. Dominio propeptídico: Formado por secuencia peptídica con residuo de cisteína
3. Dominio Catalítico carboxiterminal.
4. Dominio de tipo hemopexina: interviene en la unión a sustratos específicos de las
MMPs y con los inhibidores
5. Dominio Transmembran
Actividad Biológica
- Varias MMPs y MT-MMPs ejercen una importante acción activadora sobre otras
proMMPs y ellas mismas son activadas por otras proteasas.
4. - El PDGF (factor de crecimiento derivado de plaque-tas) produce aumento de
expresión de MMP-1 y cuando actúa conjuntamente con TGF-β produce
sobreexpresión de MMP3 y TIMP-1
- El EGF (factor de crecimiento epidérmico) induce la expresión de MMP-1 en
fibroblastos de piel humana.
- VEGF (factor de crecimiento de endotelio vascular) y FGF-2 (factor de
crecimiento de fibroblastos), son factores angiogénicos que pueden inducir la
expresión de MMPs y facilitar la diseminación metastásica
- En la artritis reumatoidea, la MMP-13 tiene un papel importante en la destrucción
del cartílago articular.
Inhibición
1. Los inhibidores específicos de MMPs son los TIMPs. Se unen a las MMPs de
manera no covalente en complejos estequeométricos (1:!), formando un complejo
reversible y de alta afinidad con dichas proenzimas.
2. En el hombre se han reconocido cuatro moléculas con actividad inhibitoria de
MMPs4. Entre las cuatro, inhiben todas las MMPs conocidas hasta el momento
difiriendo entre ellas en cuanto a su solubilidad, regulación y en su interacción
específica con la proenzima. No actúan con la misma efectividad frente a todas
las MMPs.
3. Los TIMPs son proteínas multifactoriales involucradas en varias actividades
biológicas independientes del efecto inhibitorio sobre las MMPs.
4. Intervienen en la inducción de la proliferación celular, inhibición de la apoptosis,
migración celular, invasión y angiogénesis.
5. El balance entre MMPs y sus inhibidores se ve alterado en el cáncer debido a la
pérdida de las condiciones fisiológicas de equilibrio, la MEC se ve remodelada
continuamente.
6. Hay inhibidores inespecíficos de MMPs, como la macroglobulina α2, una
glucoproteína presente en plasma y fluidos celulares, o la α1-antitripsina (α1- AT).
MMPS durante la Cicatrización
Las MMPs están implicadas en situaciones donde hay daño epitelial, endotelial y se
requiera un proceso de cicatrización. El proceso de cicatrización se puede dividir en tres
fases, inflamatoria, proliferativa y de regeneración en estas fases:
- Las células inflamatorias liberan MMPs, las cuales proporcionan el debridamiento
inicial de las proteínas de la matriz extracelular y de los tejidos que fueron dañados
o desnaturalizados durante la herida
- Las nuevas células endoteliales capilares migran hacia el lecho de la herida
isquémica generando tejido de granulación. Los miofibroblastos secretan las
MMPs que ayudan a la formación de nuevo tejido cicatrizal, mientras que las
5. células epiteliales las utilizan para migrar debajo de la costra y sobre la superficie
del nuevo lecho de la herida
- Durante la fase de remodelamiento, los fibroblastos secretan las MMPs para
ayudar a remover las porciones iniciales e irregulares de la matriz del tejido de
neoformación Las MMPs utilizadas en este proceso de cicatrización son: MMPs
1, 2, 3, 8, 9, 13, 10 donde sus expresiones manejan el proceso de cicatrización.
Metaloproteinasas y Cáncer
- Las MMPs son las principales mediadoras en las alteraciones observadas en el
microambiente tumoral durante la progresión del cáncer.
- En el crecimiento tumoral tienen un rol importante en la degradación del tejido
conectivo y componentes de membrana basal.
- Las MMPS son las responsables de la actividad proteolítica durante la progresión
tumoral.
- Las MMPs pueden jugar distintos papeles durante la progresión del cáncer,
dependiendo del estadio del tumor
- Se ha comprobado que los niveles de MMPs pueden afectar la conducta invasora
del tumor y su habilidad para metastatizar.
- Hay sobreexpresión de ADAMs en diversos tumores malignos, y su papel en el
cáncer y la diseminación a través de metástasis se relaciona con su actividad
proteolítica.
