وفهموا من هذا الحديث أن الجنين يكون نطفة في أربعين يوما و يكون علقة في أربعين يوما و يكون مضغة في أربعين يوما فيكون المجموع مائة وعشرين يوما ،وهذا الفهم يخالف ظاهر القرآن و يخالف أحاديث أخرى و يخالف الحس و المشاهدة و يخالف علم الأجنة أضف إلى ذلك أن الحديث لم ينص أن تلك الأطوار الثلاثة تحدث في مائة وعشرين يوما فكان لابد من إيضاح الفهم الصحيح الموافق لظاهر القرآن و جميع الأحاديث النبوية الثابتة و يوافق الحس و المشاهدة و علم الأجنة .

10. التحفيز الخلوي أو الجنيني : Embryonic Induction : التحفيز الخلوي هو الذي يحدد مصير الخلايا لكي تعطي ذلك النسيج أو العضو المتمايز . التحفيز الخلوي أو الحنيني هو قدرة خلايا معينة في الجنين على تحديد وتحويل خلايا أخرى . إذا كان عملية التمايز الخلوي تتطلب عملية التحديد فإن التحديد يتم عن طريق التحفيز الجنيني للخلايا . فالتحفيز الجنيني هو الذي يحدد مصير الخلايا لكي تتمايز وتعطينا الأنسجة والأجهزة والأعضاء المختلفة . إذا النسيج الحاث يصدر رسالة محفزة تؤدي إلى تحديد خلوي على المستوى الجزيئي في النسيج المستجيب فيحدث التمايز الخلوي أو الجنيني فيما بعد . كما هو معلوم فإن الأجسام الصبغية أو الكروموسومات و المورثات أوالجينات التي تحملها في جميع خلايا الجنين هي نفسها التي جاءت من البويضة الملقحة ، لكن التحفيز الجنيني يعمل على تنشيط الجينات أو المورثات الخاصة بتكوين الخلايا العضلية شكلا ووظيفة مثلا بينما تبقى المورثات الأخرى مثبطة أو غير نشطة والتي ليس لها علاقة بالنسيج العضلي كالجينات الخاصة بالسمع أو البصر مثلا والعكس صحيح . والسؤال الذي يتبادر إلى الذهن الآن هو كيف تتم عملية التحفيز الخلوي أو الجنيني للخلايا لكن لكي نفهم ذلك لابد قبل ذلك من توضيح دور بعض المكونات الخلوية والعوامل التي تتحكم في عملية التمايز الخلوي .

23. 4- دور العوامل البيئية في عملية التمايز الخلوي والجنيني Ecological factors - تلعب البيئة دور أخر في عملية التمايز الخلوي والجنيني فهي تمثل الوسط الذي يتم إمداد الجنين خلاله بالمواد الغذائية و حمايته من العوامل البيئية الداخلية أو الخارجية . العوامل البيئية الداخلية : إذ يمكن أن تكون من بيئة الجنين نفسه كالوراثة والجينات والخلل الذي يحدث بها من طفرات أو من الأم وما تنقله من عوامل وتخزنه داخل البويضة أو أثناء الحمل وما توفره من غذاء وحماية أو عوامل بيئية خارجية : كالحرارة والرطوبة والضوء والهواء وتغير الأس الهيدروجيني والإشعاعات والملوثات بمختلف أنواعها الكيميائية والطبية الفيروسات والأمراض و نقص أو زيادة عنصر من العناصر المهمة في بيئة نمو الأجنة؛ كل فهذه العوامل تعمل على انحراف في النمو والتمايز الخلوي للأجنة . ـ تأثير الحرارة على تمايز الأجنة : عند تنمية جنين الضفدعة في درجتين حراريتين مختلفين وذلك بوضع قطن على جزء من الجنين أثناء مرحلة التفلج وتزويدها بالحرارة أو البرودة بالتقطير عليها بمحلول البيئة درجة حرارة مختلفة عن النصف الأخر بزيادة أو نقص 5 م فإن ذلك يؤدي إلى نمو شاذ كما في ( الشكل 10-2 ). كذلك عند تحضين بيض الدجاج عند درجة حرارة اقل من 35 م أو أكثر من 40 م درجة مئوية فإن نمو الأجنة يتوقف . وقد تطول مدة فقس البيض عند انخفاض درجة الحرارة أو تقصر عند ارتفاعها إذا كانت ضمن معدل الحرارة المناسب لنمو الأجنة داخل البيض .

27. المنظمات الجنينينة (Embryonic Organizer ) المنظم هو قدرة نسيج معين على تحويل أو تحفيز نسيج أو أنسجة أخرى لكي تتمايز وتعطي أعضاء أثناء مراحل التكوين الجنيني المبكرة . ففي البرمائيات وجد أن الشفة الظهرية لثقب المفلجة ( البلاستوبور ) وعقدة هنسن في أجنة الطيور والثدييات هي المنظمات الجنينينة في هذه الأجنة حيث تعمل على تحفيز تكوين الأنبوبة العصبية فيها . وذلك من خلال تجارب نقل وزراعة أجزاء من الأجنة إلى أجنة أخري . المنظمات الجنينينة في أجنة البرمائيات : من خلال التجارب التي آجراها هانس سبيمان على أجنة البرمائيات المبكرة وذلك عند فصل البويضة المخصبة إلى جزئي بحيث يحتوي أحدها على منطقة الهلال السنجابي أو الرمادي ( Gray crescent ) والأخرى لا تحتوي علية فإن الأولى تنمو وتكون جنين كامل التكوين بينما الأخرى لا تنمو ( شكل 12-2). كما أن نقل وزراعة أجزاء من منطقة الهلال الرمادي لجنين الضفدعة على جنين أخر تؤدى إلى تحفيز تكوين أنبوبة عصبية على الجنين المستقبل . لقد استطاع كل من هانس سبيمان وطالبته هيلد مانقولد عام 1924 م ( Hans Speman & Hilde Mangold ) من نقل الشفة الظهرية لطور المبطنة ( الجاسترولة ) المبكرة لجنين العلاجم ( Newt ) داكن اللون من نوع تريتوريس تنيوتس Triturus taeniatus) ) وزراعة في منطقة الأكتوديرم البطني لمبطنة جنين أخر ذو لون فاتح من نوع تريتوريس كريستاتس ( T riturus cristatus ) فإنها لم تتوقف عند تكوين ثقب أخر للمفلجة ( Blastopore ) بل كونت عملية تبطين أخرى وتكوين جنيني أخر في محيط نسيج المنطقة التي زرعت فيها ( الشكل 13-2 ).

30. وعندما غير ووضع الشفة الظهرية لمبطنة أو لجاسترولة متأخرة ( Late blastopore ) فقد عملت على تحفيز لتكوين الجزء الخلفي للأنبوبة العصبية شكل (14 أ -2) ولمعرفة بما يسمي الخاصية المكانية للتحفيز ( Regional specificity of induction) فعند نقل وزراعة الجزء الأمامي فقط لقطعة من خلايا الحبل الظهري في طور الأنبوبة العصبية ونقلها الى جنين في طور المبطنة الجاسترولة المبكرة , فإنها تؤدي إلى تكوين رأس لجنين ثانوي على الجنيني المستقبل ، بينما نقل وزراعة خلايا الجزء الخلفي لقطعة من نسيج الحبل إلى جنين في طور المبطنة المبكرة وتحت الطبقة الخارجية فإنها تعمل على تكوين الذيل أو الجزء الخلفي لجذع وزعنفة ذيلية كحنين ثانوي على الجنين المستقبل ( الشكل 14 ب -2).

32. شكل ( 17-2) يوضح بأن عملية التحفيز غير مرتبطة بالجنس : تجربة أجرها العالم سبيمان ومساعدة أوسكار حيث قاما بنقل وزراعة أجزاء من النسيج الميزنشيمي ( والذي سوف يحفز لتكوين الممص الفمي ) لمنطقة الطبقة المتوسطة أو الميزوديرم من طور المبطنة الجاسترولة لجنين الضفدعة وزراعتها مكان منطقة الزوائد الفميه لمبطنة أو جاسترولة العلجوم ( لنيوت Newt ) والعكس نقل أجزاء من النسيج الميزنشمي ( والذي سوف يحفز لتكوين الزوائد الفمية ) لمنطقة القطعة المتوسطة أو الميزوديرم لمبطنة أو الجاسترولة العلجوم وزراعتها على مبطنة أو جاسترولة الضفدعة ، فالنتيجة كان أن تكون ممص فمي ( من النسيج الطلائي ) للعلجوم أو النيوت وزائد فمية للنيوت على جنين الضفدعة . من هذا يتضح أن النسيج الطلائي للنيوت استجاب للمحفز الصادر من النسيج المزنشيمي لجنين الضفدعة فتحول إلى ممص فمي والعكس تم تحفيز تكوين الزائد الفمية على جنين الضفدعة بواسطة النسيج الميزنشيمي المحفز للنيوت . يرقة الضقدعة يرقة التيوت
33. المنظمات الحنينية في أجنة الطيور : لقد اكتشف وادنقتون عام 1933 م (Waddington 1933) المنظمات الجنينية في أجنة الطيور ألا وهي عقد هنس (Hensen node) فهي تعمل على تكوين جنيني أخر ( أنبوبة عصبية وجذع للجنين ) فعندما قام بنقل عقدة هنسن من جنين دجاج في طور المطنة أو الجاسترولة المبكرة وزراعتها على القرص الجنين لجنين البط بنفس العمر تقريبا فأعطت تكوين جنين ثانوي على الجنين المستقبل ( انظر الشكل 17-2 ). فعقدة هنسن منظم جنيني كالشفة الظهرية في البرمائيات فالخلايا التي تهجر من طبقة السطحية عبر عقدة هنسن سوف تكون منطقة الرأس والطبقة الداخلية ( الأندوديرم ) و الطبقة المتوسطة لخلايا ( أوميزودير ) الحبل الظهري بينما الخلايا التي تهجر من سطح القرص الجنيني عبر المنطقة الخلفية للخط البدائي (Primitive streak) سوف تكون منطقة خلايا الطبقة المتوسطة أو الميزوديرم الجانبي والبطني (Ventral and lateral mesoderm) ثم عمد إلى عمل مجموعة من التجارب كان الهدف منها معرفة اتجاه محور الجنين الثانوي ومدى علاقة النسيج الحاث بالنسيج المستجيب ( المانح والعائل (Donor and host فقد نقل عقدة هنسن من جنين وزرعها بالقرب من عقدة هنسن الأساسية للجنين العائل وفي اتجاه محور الجنين الأساسي فقد تكون جنين ثانوي اتجاه رأسه باتجاه الجنين الأساسي . وفي تجربة أخرى زرع عقدة هنسن المنقولة بعيدا عن عقد هنسن الأساسية للجنين العائل مرة باتجاه الجنين العائل ومره عكسه اتجاهه فأعطت أجنة ثانوية اتجاها حسب اتجاه عقدة هنسن المنقولة .
34. - وفي تجربة أجريت على نقل وزراعة أجزاء في أجنة الطيور ( الدجاج ) لدراسة العلاقة بين النسيج الحاث والنسيج المستجيب فقد تم نقل وزراعة نسيج الطبقة المتوسطة ( ميزوديرم ) الجناح مكان الطبقة المتوسطة ( ميزوديرم ) الرجل وتحت الطبقة الخارجية ( أكتوديرم ) في جنين الكتكوت المبكر ؛والعكس بدل النسيج الميزوديرمي لموقع الجناج بنسيج الطبقة المتوسطة ( ميزوديرم ) الرجل ووضع أيضا تحت الطبقة الخارجية ( اكتوديرم ) أو أدمة الجناح ؛فالنتيجة أنة تكونت رجل مكان الجناح وجناح مكان الأرجل ( الشكل 18-2) ومن هذه التجربة نستنتج انه على الرغم من أن الطبقة المتوسطة وهي في المرحلة الجنينية المبكرة تكاد تكون واحدة إلا أنها أعطت تحفيز مختلف فالأمامية تعطي ريش وجناح والخلفية تعطي حراشف و أرجل . أو بما يعرف الخاصية المكانية أو الموقعية للمحفز ( Regional Specificity of Induction ) .

36. المنظمات الجنينية في أجنة الثدييات : نظرا لطبيعة نمو جنين الثدييات فإن التجارب التي أجريت أغلبها على مرحلة التفلج أو على شكل زراعة خلايا نسيجية . فعند تدمير إحدى الفلجتين لجنين الفأر فإن الفلجة الأخرى تنمو وتعطي جنين كامل . كذلك عند فصل طور الخليتين أو الأربع خلايا لجنين الأغنام وتنميتها إلى طور المفلجة أو البلاستولة ثم نقلها إلى أم مستقبلة و مهيأة فسيلوجيا للاستقبال الأجنة فإن كل فلجة تعطي جنيني كامل من تجارب زراعة الخلايا على أجنة الثدييات فقد وجد أن عند زراعة عقد هنسن ( من مقدمة الخط البدائي ) لجنين الثدييات مع خلايا طبقة خارجية أكتوديرمية فإنها تحثها على تكوين خلايا عصبية . شكل (19-2) المنظمات الجنينية في أجنة الثدييات : فصل فلجات الجنين في مراحل التفلج المبكر للأغنام تؤدي إلى أن تكون كل فلجة جنين كامل .
37. استنتاجات على الحث الجنيني والمنظمات الجنينية : - من تعريف المنظم أو عملية التحفيز بأن هناك نسيج يسمى بالمحفز يؤثر على نسيج أخر فيحدد مصيره ويعمل على تحويله إلى نسيج أخر متمايز فيسمى بالنسيج المستجيب . - في بداية عملية التمايز هناك بما يسمى بالمنظم الابتدائي ( الشفة الظهرية في أجنة البرمائيات وعقدة هنسن في أجنة الطيور والثدييات ) تعمل على تكوين المحفز الأولي ( خلايا الحبل الظهري ) والذي بدور يعمل على تحفيز تكوين الأنبوبة العصبية في الخلايا التي تعلوه في الجنين . - أن هذا المحفز الابتدائي يفقد تأثيره بتقدم عمر الجنين أو يؤدي مهمته في الوقت المبكر للنمو وكلما زاد عمر الجنين قل التأثير والتأثر بالمحفز . - فالمحفز يعمل في مرحلة معينة من مراحل النمو الجنيني وكذلك النسيج المستجيب يتأثر بهذا المحفز في تلك الفترة من تأثير النسيج الحاث . تفسير عمل المنظمات الجنينية والحث الجنيني أولا : الاتصال الخلوي Cellular Contact: لقد أجريت عدة تجارب لمعرفة كيف تحدث عملية التأثير بين النسيج الحاث والنسيج المستجيب وكانت أول التجارب التي تفسر هل الاتصال الخلوي المباشر بين النسيج الحاث والنسيج المستجيب ضروري لحدوث عملية الحث الجنيني ومن ثم حصول التمايز الخلوي للنسيج المستجيب .
38. - فكما هو معلوم أن وجود العلاقة والتقارب بين خلايا القطب الحيواني ( الطبقة الخارجية أو الاكتوديرم ) وخلايا القطب الخضري ( الطبقة الداخلية أو الاندوديرم ) ضروري لتكوين خلايا الطبقة المتوسطة ( الميزوديرم ) - وكذلك وجود خلايا الحبل الظهري تحت خلايا الطبقة الخارجية أو الأكتوديرم ضروري لتكوين الأنبوبة العصبية . - فقد دلت تجارب زراعة الخلايا والأنسجة أن زراعة خلايا الحبل الظهري أو خلايا القطع الجسمية Somites كل على حدي لا تؤدي إلى تكوين نسيج غضروفي لكن عند زرعتهما مع بعض فإن ذلك يؤدي إلى تكوين النسيج الغضروفي ( الشكل 20-2) - ولمعرفة هل الاتصال الخلوي المباشر بين النسيج الحاث والنسيج المستجيب ضروري لعملية التحفيز فقد تم وضع غشاء رقيق غير منفذ مصنوع من السلوفان ( Cellophane ) بين الكأس البصرية ( النسيج الحاث ) والطبقة الخارجية ( الأكتوديرم ) المواجه لها ( النسيج المستجيب ) وهل ستعمل الكأس البصرية على تحفيز تكوين عدسة العين من الطبقة الخارجية ( أو الأكتوديرم ) فوجد بأنة لا تتكون عدسة العين . - وفي تجربة أخرى استخدم غشاء منفذ وهو عبارة عن شريحة من الأجار حيث تسمح بمرورالجزيئات ووضعت بين الكأس البصرية والطبقة الخارجية المواجهة لها ( الأكتوديرم ) فكانت النتيجة أن تكونت عدسة العين ( الشكل 21-2).
39. فتجارب الغشاء غير المنفذ تفيد بأن الاتصال الخلوي بين النسيج الحاث والنسيج المستجيب ضروري لحدوث عملية التحفيز . كذلك في تجارب الغشاء المنفذ وعند فحص الأغشية المنفذة بالمجهر الإلكتروني وجد أن هذه الأغشية بها خملات دقيقة ( Microvillies ) مما أدى إلى أن العلماء استنتجوا أن الاتصال الخلوي المباشر بين النسيج الحاث والنسيج المستجيب ضروري لحدوث عملية التحفيز الجنيني . ثانيا : التأثير الكيميائي ( Chemical Actions ): إن التجارب الحديثة تدل على أن الاتصال الخلوي المباشر بين النسيج الحاث والنسيج المستجيب غير ضروري لحدوث عملية الحث الجنيني ، إنما تعتمد عملية الحث على انتقال جزيئا ت كيميائية من النسيج الحاث إلى النسيج المستجيب وذلك من خلال التجارب الأتيه : فقد أعيدت نفس التجارب السابقة للاتصال الخلوي لكن استخدمت أغشية ذات ثقوب دقيقة جدا لا تسمح بتكوين خملات دقيقة خلالها ووضعت بين النسيج الحاث والنسيج المستجيب ثم اخذ النسيج المستجيب ( نسيج الطبقة الخارجية الأكتوديرم ) وزرع لوحده في بيئة تنمية فتحول إلى نسيج عصبي أي حدثت عملية التحفيز؛ كذلك فحص الأغشية بالمجهر الإلكتروني فلم يلاحظ وجود الخملات في الثقوب . عند زراعة خلايا القطب الخضري أو الطبقة الداخلية ( الأندوديرم ) في بيئة تنمية ثم رفعها ووضع خلايا القطب الحيواني أو الطبقة الخارجية ( الأكتوديرم ) في نفس البيئة فإنها تتحول إلى خلايا الطبقة المتوسطة ( الميزوديرم ) الشكل 23-2).
40. أعيدت نفس فكرة التجربة السابقة ولكن تم تنمية نسيج الشفة الظهرية في البيئة ثم أزيلت من البيئة وتم وضع نسيج للطبقة الخارجية ( الأكتوديرم ) في نفس البيئة فتحولت إلى نسيج عصبي . ولتفسير ذلك من التجارب السابقة أتضح أن البيئة قد احتوت على مواد أو جزيئات أفرزت من النسيج الحاث ( الشفة الظهرية ) وعند تنمية النسيج المستجيب في نفس البيئة وبعد إزالة النسيج الحاث عملت هذه المواد أو الجزيئات على تحفيز النسيج المستجيب ( الطبقة الخارجية أو الأكتوديرم ) و تحويله إلى نسيج عصبي . وعند زراعة أي نسيج آخر غير الشفة الظهرية في البيئة فإنه لا يعمل على تحول خلايا الطبقة الخارجية ( الأكتوديرم ) إلى نسيج عصبي ؛ مما يدل أيضا على أن نسيج الشفة الظهرية هي التي تمتلك المواد الخاصة والتي تعمل على الحث العصبي . شكل (22-2) يوضح التجارب التي تدل على أهمية الأتصال الخلوي في عملية التمايز : ( أ ) تجربة وضع الغشاء غير المنفذ ( السلوفان ) بين الشفة الظهرية والطبقة الخارجية ( الأكتوديرم ) لا يتكون . | نسيج من الطبقة الخارجية اكتوديرم . ( غشاء غير منفذ ( من السلوفان ) نسيج طبقة خارجية ( اكتوديرم ) نسيج شفة ظهرية ___ نسيج حاث زراعة الأكتوديرم غشاء منفذ ( من الأقار ) نسيج طبقة خارجية ( أكتوديرم )__ يتمايز إلى نسيج شفة ظهرية خلايا عصبية ب ) وفي حالة استخدام الغشاء المنفذ بينهما فقد تكون نسيج عصبي .
41. الشكل (20-2): يوضح العلاقة بين النسيخ الحاث والنسيج المستجيب أن زراعة خلايا الحبل الظهري أو خلايا القطع الجسمية Somites كل على حدي لا تؤدي إلى تكوين نسيج غضروفي لكن عند زراعتهما مع بعض فإن ذلك يؤدي إلى تكوين النسيج الغضروفي . خلايا حبل ظهري لوحدها خلايا قطع جسدية لوحدها خلايا حبل ظهري + خلايا قطع جسدية نسيج غضروفي

44. طبيعة المادة الكيميائية : - و لمعرفة طبيعة المادة الكيميائية التي تصدر من النسيج الحاث إلى النسيج المستجيب فقد قام العلماء على إجراء التجارب التالية : - بعد رفع النسيج الحاث ( الشفة الظهرية ) من البيئة أضيف أنزيم التربسين ( والذي يعمل على هضم المادة البروتينية ) إلى البيئة التي تم تنمية النسيج الحاث فيها ثم وضع النسيج المستجيب ( الأكتوديرم ) فوجد انه لا يتحول إلى نسيج عصبي ( الشكل 24-2) - ومن ذلك استنتج الباحثون أن طبيعة المادة المفرزة هي عبارة عن بروتين يفرز من النسيج الحاث ( الشفة الظهرية ) إلى النسيج المستجيب ( الأكتوديرم ). الشكل ( 24-2) تجارب توضح طبيعة المادة المحفزة للمنظم الجنيني :
45. كيف يؤثر المحفز التأثير أو العامل الموضعي ( Paracrine Factors ) : عند دراسة كيفية انتقال المواد المحفزة من النسيج الحاث إلى النسيج المستجيب ففي الدراسة التى آجراها جروبستين ( Grobestein ) 1956 م وأخرون على تمايز الأنيبيبات الكلوية والأسنان فقد وجد أن عملية التحفيز تتم حتى في حالة وجود الحواجز ذات الثغور أو عديمة الثغور في حالة وضعها بين النسيج الحاث والنسيج المستجيب . في بعض المحفزات تحتاج للثغور ومحفزات أخري تحتاج للاتصال بينها وبين النسيج الحاث حتى تتم عملية التأثير فعندما يتم إفراز مادة محفزة كالبروتين مثلا من النسيج الحاث وتنتقل عبر مسافة قصيرة ( عبر الفراغ أو الفسحة بين الخلوية التي تفصل النسيج الحاث عن النسيج المستجيب ) إلى النسيج المستجيب و يحدث فيها تغيرا أو تأثيرا فهذا يسمي التأثير الموقعي أو المحفز المحلي ( Paracrine Factors Or Growth and Differentiation Factors GDFs) ). لكن عندما تنتقل المادة المحفزة من النسيج الحاث إلى النسيج المستجيب عبر تيار الوعاء الدم كما في يتم عند تحفيز تكوين الأنيبيبات الكلوية والأسنان حيث تنتقل المادة المحفزة من النسيج الحاث إلى النسيج المستجيب بواسطة تيار الدم بالضبط كما تأثر الهرمونات فهذا يسمى بالتأثير الهرموني ( Endocrine factor )
46. الخاصية الجزيئية لعملية التحفيز ( Molecular Specificity of Induction ). - يفرز النسيج الحاث المادة المحفزة ( البروتين مثلا ) وتنتقل هذه المادة إلى النسيج المستجيب فترتبط بمستقبل على غشاء الخلايا المستجيبة وهذا الارتباط بدوره ينشط عوامل معينة داخل سيتوبلازم الخلايا المستجيبة والتي تنقل الإشارة إلى داخل نواة الخلايا المستجيبة فترتبط بالمادة الوراثية مما ينشط مورث أو مجموعة جينات معينة في الخلايا المستجيبة فتحدد مصيرها فتصدر هذه المورثات المنشطة شفرة معينة من النواة الى السيتوبلازم عبر الأحماض النووية المرسلة ( mRNA ) حيث يتم ترجمتها فيه فيحدث التغير والتمايز لهذا الخلايا كإستجابة لتحفيز المادة المنظمة الصادرة من النسيج الحاث
