This review examines two randomized controlled trials that compared oral vitamin B12 to intramuscular vitamin B12 for treating vitamin B12 deficiency. The trials included a total of 108 participants and followed them for 90 days to four months. Both trials found that high oral doses of vitamin B12 (1000 mcg and 2000 mcg) were as effective as intramuscular administration in improving hematological and neurological outcomes. The authors concluded that high daily oral doses of vitamin B12 may be as effective as intramuscular injections for treating vitamin B12 deficiency in the short term.

We therefore conclude that there is no current evidence that treatment with clioquinol (PBT1) has any significant effect on cognition and in particular memory (as measured by the ADAS-Cog scale) in patients with Alzheimer's dementia. This drug has now been withdrawn from development. The trial of PBT2 showed it was safe after 12 weeks of treatment but demonstrated no overall significant effect on cognition or memory.
