3. Aim: Carbohydrates Fermentation Test

.Material Required:-
• Suitable Basal Medium(Peptone Water):-
• Peptone water composition
Indegrident Amount
• Peptone 10.00 gm
• Nacl 5.00 gm
• Distilled Water 1000 ml
• PH 7.4
• Sugar(1% or 0.5gm)may be :-
• Glucose

4. • Suitable indicator:-phenol red indicator (0.008gm)
• Bacterial culture for (18 to 24 hrs / solid or liquid broth medium)
• Durham’s tube
• Inoculating loop
• Incubator
• Bunsen burner
• Sterile test tube
• Note:- (peptone water is an broth medium and used for non-fastidious
organism)
• Note:-(phenol red is commonly used as PH indicator in carbohydrates
fermentation test and also used Andrade’s indicator (sodium hydroxide 1
acid fuchsin)
• Note:-(Durham tube is used in microbiology to detect the production of GS
by microorganisms and made of clear tubular glass and size:1 to 3 inch)

7. Introduction :
1.Carbohydrates fermentation test is detect the ability of
microorganisms to ferment a specific carbohydrates and ferment can be
used to differentiate among bacteria
2.This test is used for the presence of acid and gas produced from the
fermentation of a single particular carbohydrates
3.This test is biochemical test
4.Carbohyrate fermentation is the process of microorganisms used to
produce energy

8. Principle:-
• Carbohydrates fermentation test is used to determine the ability of a
bacterium to ferment a specific carbohydrate (sugar) Producing acid
or acid with gas

10. Procedure:-
1. Prepare the peptone(broth medium)
2. Then, add sugar(1% or 0.5gm) may be:- glucose , lactose and sucrose
3. Then, add phenol red indicator and mix well
4. Then, insert Durham’s tube at inverted position in test tube
5. Then, take a fresh culture medium (18 to 24 hrs/solid or liquid
medium)with the help of Inoculating loop
6. Then, mix the culture medium into test tube and apply cotton plug
7. Then, incubate At 37 degree celsius for 18 to 24 hrs
8. Then, observe result

11. INTERPRETATION:
• Positive:- broth medium color change to yellow and
gas bubbleis found in durham tube then,indicate acid
and gas production
• Negative:-broth medium colour red and no change
the color
• E.x:-pseudomonas.sp shows yellow color change in
glucose
• E.coli, klebsiella.sp ferment the sugars:-
glucose,lactose and sucrose
