Introduction In this investigation pUC19 plasmids were used as the vector due to
its small size of 2686bp, high uptake efficiency by the host and fast replication
time. Important features of this plasmid include the origin of replication and
multiple cloning sites (MCS). The origin of replication allows the plasmid to
replicate inside the host bacterium. The MCS is located within the lacZ gene and
contains unique sites for the Xbal EcoRI restirction enzymes to cut and produce
sticky ends for the CIH 1 gene to bind to. Furthermore, the pUC19 plasmid also
contains an ampiccilin resistance gene so only transforemed E.coli are able to
remain viable when spread on the agar plates that also has the addition of ampiccilin.
The lacZ gene encodes the ОІ galactosidase enzyme which aids in indentifying the
recombinant E.coli from the non recombinant cells (Coventry University 2016). The
addition of 5 bromo 4 chloro 3 indoyl ОІ thiogalactoside (X gal) in the agar mix acts
as an artifical substrate for the enzyme so it produces blue E.coli colonies when
hydrolised by the ОІ galactosidase meaning those specific colonies do not contain the
plasmid with the CIH 1 insert (non recombinant E.coli) (Coventry University 2016).
Isopropyl ОІ D thiogalactoside (IPTG) an artifical transcription inducer of the lacZ
gene is also added into the agar mix, binding to the repressor gene and inactivating it.
Therefore when the CIH 1 gene is incorporated within the plasmid MCS and inserted
into the E.coli

Jochebed and Miriam (Moses mother and sister respectively) showed a great deal of
courage when facing an edict from a powerful Pharaoh.
On the other hand, the Egyptian leader was not courageous. He had complete
control of the Hebrew slaves, and yet he feared them so much he ordered the death
of all newborn baby boys.
Into this chaos and turmoil, Moses is born. Desperate to save her son, Jochebed
kept him hidden. Every whimper from this baby must have brought fear into her
heart, but her faith kept her strong. She knew Moses was special and God had great

Bletchley Park was a location set up by the British government dedicated entirely
to breaking enigma. However, they were extremely secretive about the work going

creek and west creek catchments. In January 2011 there was a main flooding event

surprise that the video game industry is not immune. Common red flags like
Copyright infringement, using the likeness of others, or 1st amendment concerns, are
volatile problems that can hinder the progress with any studio. Learning how to
identify major issues such as these can help a company avoid legal ramifications in
the long run. Three interesting cases that highlight these issues and can be used as
great examples are: Tetris v. Xio, Davis/Dupree/Keller/Ferragamo v. Electronic Arts
and Schwarzenegger v. Entertainment Merchants Association.
The first case I wanted to point out was Tetris v. Xio. This was a case that dealt with
copyright infringement issues. Tetris Holdings, LLC, own the popular tile matching
puzzle game Tetris. They found themselves in a legal battle with Xio Interactive
Inc., the creators of Mino, a puzzle game of a similar look and feel. Tetris, LLC
accused Xio Inc. of creating a game that was strikingly similar to their title. Xio
argued that they had only copied the non protectable elements of the game and

Introduction
My wheelchair simulation experience took place on Monday, September 26th. I
started in my on campus dorm around 6pm. After wheeling around for a half hour,
I began making my way to CafГ© 210 for dinner and drinks with a friend. The
textbook mentions that participants will experience that even the best laid plans
will lead them into the unexpected (Dattilo, 2012, p. 74) and it was especially true
throughout my simulation. I realized that making it to CafГ© was a very lofty
goal and given that it was raining, made it nearly impossible. I had to call the friend
I was meeting and change where we were meeting, deciding on the much closer
Inferno. We spent about two hours eating and catching up before pulling out a
deck of cards and playing a few card games for another hour. After our long dinner,

conscious reaction to Schoenberg s serial music and a synthesis of many styles of
music while including serial elements from the Viennese school. This connection
