Origin of Protein
 The term protein is derived from a Greek
word proteios meaning holding the first place.
Berzelius( Swedish chemist) suggested the
name proteins to the group of organic
compounds that are utmost important to life.
Mulder (Dutch chemist) in 1838 used the term
proteins for the high molecular weight
nitrogen-rich and most abundant substances
present in animals and Plants .
Standard Amino Acids
• As many as 300 amino acids occur in nature
of these , only 20- known as standard
amino acids are repeatedly found in the
structure of proteins isolated from
different forms of life – Animal , plant and
microbial.
• Selenocysteine - the 21st amino acid
• Pyrrolysine - the 22nd amino acid
AMINO ACIDS
• Amino acids are a group of organic
compounds containing two functional
groups- amino Acid and carboxyl.
• The amino group (-NH2) is basic while the
carboxyl group (-COOH) is acidic in nature

• The amino acids are termed as Alpha-amino
acids, if both the carboxyl and amino groups
are attached to the same carbon atom, as
depicted below-

CLASSIFICATON OF AMINO ACID
• There are different ways of classifying the
amino acids based on the structure and
chemical nature, nutritional requirement
metabolic fate etc.

Composition of protein
• Proteins are predominantly constituted by five
major elements in the following proportion.
Carbon 50 - 55%
Hydrogen 6 - 7.3%
Oxygen 19 - 24%
Nitrogen 13 - 19%
Sulfur 0 – 4 %
• Besides the above, proteins may also contain
• Other elements such as P, Fe, C u, l, Mg, Mn , Zn
etc.
Primary Structure
The linear sequence of amino acids
forming the Backbone of proteins
(polypeptide)
Secondary structure
 The folding of
the N-C-C back
bone of the
polypeptide Chain
using weak
hydrogen bonds .
 The spatial
arrangements of
protein by twisting
of the polypeptide
chain.
Secondary structure
This produces the alpha helix and beta pleating
 The length of the helix or pleat is determined by certain
amino acids that will not participate in these structures
(e.g. proline)
TERTIARY STRUCTURE
• The three-
dimensional
arrangemen
t of protein
structure is
referred to
as tertiary
structure.
QUATERNARY STRUCTURE
• Some of the proteins
are composed of two
or more polypeptide
chains referred to as
subunits.
• The spatial
arrangement of these
subunits is known as
quaternary
structure.
Summary of Classification of protein
Functions of proteins
 Proteins perform a great variety of specialized and
essential functions in the living cells.
 These functions may be broadly grouped as static
(structural) and dynamic.
 CATALYTIC: enzymes
 TRANSPORT: haemoglobin
 COMMUNICATION: hormones (e.g. insulin) and
neurotransmitters
 CONTRACTILE: actin , myosin, dynein (in microtubules)
 PROTECTIVE: Immunoglobulin, fibrinogen, blood clotting
factors
 TOXINS: snake venom
 STRUCTURAL: cell membrane proteins , keratin (hair) ,
collagen

Origin of Protein  The term protein is derived from a Greek word proteios meaning holding the first place. Berzelius( Swedish chemist) suggested the name proteins to the group of organic compounds that are utmost important to life. Mulder (Dutch chemist) in 1838 used the term proteins for the high molecular weight nitrogen-rich and most abundant substances present in animals and Plants .

  • 8. Composition of protein • Proteins are predominantly constituted by five major elements in the following proportion. Carbon 50 - 55% Hydrogen 6 - 7.3% Oxygen 19 - 24% Nitrogen 13 - 19% Sulfur 0 – 4 % • Besides the above, proteins may also contain • Other elements such as P, Fe, C u, l, Mg, Mn , Zn etc.

  • 10. Secondary structure  The folding of the N-C-C back bone of the polypeptide Chain using weak hydrogen bonds .  The spatial arrangements of protein by twisting of the polypeptide chain.
  • 11. Secondary structure This produces the alpha helix and beta pleating  The length of the helix or pleat is determined by certain amino acids that will not participate in these structures (e.g. proline)
  • 12. TERTIARY STRUCTURE • The three- dimensional arrangemen t of protein structure is referred to as tertiary structure.
  • 13. QUATERNARY STRUCTURE • Some of the proteins are composed of two or more polypeptide chains referred to as subunits. • The spatial arrangement of these subunits is known as quaternary structure.

Functions of proteins  Proteins perform a great variety of specialized and essential functions in the living cells.  These functions may be broadly grouped as static (structural) and dynamic.  CATALYTIC: enzymes  TRANSPORT: haemoglobin  COMMUNICATION: hormones (e.g. insulin) and neurotransmitters  CONTRACTILE: actin , myosin, dynein (in microtubules)  PROTECTIVE: Immunoglobulin, fibrinogen, blood clotting factors  TOXINS: snake venom  STRUCTURAL: cell membrane proteins , keratin (hair) , collagen
