• Our institute priority is to scale up and implement research results to end users
such as to farmers, private & commercial Tissue Culture (TC) rather than
trying to publish them. As a result, the above research results have not yet been
published.
10. 1. Proposed study plan
Title: Study on Biocontrol Mechanism of Endophytic Bacterial wilt
(Ralstonia solanacearum) using morphological, biochemical, and
molecular techniques.
STUDY PLAN
11. 1.1 INTRODUCTION
• Bacterial wilt is one of the major diseases of tomato and
other solanaceous plants.
• The disease is caused by the bacterium Ralstonia solanacearum,
previously known as Pseudomonas solanacearum which is most
damaging plant pathogens.
• Strains of this pathogen affect more than 200 plant species in over
50 families throughout the world like Potato , tomato banana,
ginger , tobacco , sweet pepper, olive, rose and soybean etc (Rieter
et al.2012; Das et al. 2017; El-deeb et al. 2013 and Elbeltagy et al
2000).
• Endophytic bacteria may have certain metabolic properties, such as
promoting plant growth, controlling soil-borne pathogens, or
helping host plant to defeat stress responses to environmental abuse.

• Biocontrol of bacterial wilt pathogen of tomato using endophytic
bacteria is one of control method to support sustainable agriculture
(NAWANGSIH et al. 2011).
• Several groups of bacteria have been explored for biocontrol of
pathogens. Plant Growth-Promoting Rhizobacteria (PGPR) are one
of the most studied biocontrol agents.
• However, PGPR are usually effective at laboratory or green house
scale, often fail to provide effective disease suppression in a field.
Therefore, this study focus on obtaining comprehensive pictures of
another group of endophytic bacteria from the tomato plants that
used as a biocontrol agents.
13. 1.2 OBJECTIVES
1.2.1 General Objectives
To Study the Biocontrol Mechanism of Endophytic Bacteria based on
morphological, biochemical, and molecular techniques
1.2.2 Specific objectives
• To select & characterize endophytic bacteria isolated from tomato stems,
and
• To investigate whether they can act as plant growth-promoter or as
biocontrol agents of tomato bacterial wilt disease.
14. 1.3 METHODOLOGY
• First Isolate and purify endophytic bacteria by
growing on Nutrient Agar (NA) from tomato
plants with no apparent disease symptoms.
• Then the Pathogenicity of Endophytic Bacteria
will be test by Inoculation of other host plants.
• Screening of endophytic bacteria for antibiosis
using in-vitro competition mechanism of culture
assay
• Finally Characterization & Identification Selected
Endophytic Bacteria will be analyzed based on
morphological (microscope and colony
appearances) biochemical properties, and
molecular analyses (Extracting DNA and
comparing sequence analysis of their 16S rRNA
gene)

15. Furthermore, I have commitments and interest to work on collaborative

• Functional analysis of plant (soybean) genes by molecular reverse genet
