WATERMELON MOSAIC VIRUS II IN CUCURBIT PLANTS
Title: Genome-wide approaches for the identification ofmarkers and genes associated with Water
melon mosaic virus 2 (WMV2) resistance in melon cultivars.
TARGET: TO INVESTIGATE THE RESPONSE OF VARIOUS MELON CULTIVARS BY
INOCULATINGWMV-II
 To develop and report a new marker that can be used in segregating resistant from
susceptible melon plants
 To evaluate the efficacy of severalgenome-wide approaches and identifying
markers and genes associate with WMV2 resistance.
 To analysis the biological and molecular variability of WMV-II by amplification
of their CP gene
 To Selection of breeding resources to foster virus-resistant melon cultivar
Experimental plan:
- Collection of plant materials and inoculation will be carryout in greenhouse.
- Phenotypic data analyses: Morphological and biological data (infectivity, symptoms,
weight of plant above and below soil) will be recorded. WMV2 will be evaluated for the
severity of melon cultivar symptoms and for viral titer estimating by relative in tissue
exudate methods and absolute RT-qPCR quantification in different season at least two
consecutive years
- Genotyping and genetic analyses: genotyping by sequencing (GBS) library will be
constructed in order to allowed the successfulgenotyping in all collecting accessions
- Association analyses: the coefficient of variation (significance difference) will be analyzed
with in Phenotypic data analyses and genetic analyses. the non-redundant markers that
significantly associated with cucurbit plant virus symptom severity will be analyzed
based on a benchmark of stringent significance threshold.
- Marker mapping and annotation: markers will be identifying based on the physical
location of all markers associated with WMV2 resistance, and other authors benchmark.
