3. RESEACRH EXPERIENCE AND INTEREST
My research interest lies in the structural and biochemical characterizations of challenging proteins
such as multi-subunit complexes and mammalian proteins. I have followed my interest since last eight years
and have gained sufficient experience in this area. During my research career I have developed skills of
working independently as well as in team, making hypotheses, designing experiments, analyzing data,
writing grant application and papers and presenting research work to a variety of audience in form of posters
and oral presentations.
I worked as Extended Senior Research Fellow from December 2014 to December 2015 in the lab of
Dr. Nimisha Sharma (GGS Indraprastha University, New Delhi, India). The lab works on elucidating molecular
mechanism of transcription regulation in yeast by studying the biochemical and biophysical properties of
different RNA polymerase subunits. My project was based on deciphering the biochemical and biophysical
properties of yeast ELL and EAF proteins. I worked on expression and purification optimization of
recombinant ELL and EAF. Manuscript on this work is currently in progress.
Before joining Dr. Nimisha’s lab, I was a doctoral student in Dr. Debandra K. Sahoo’s lab in CSIR-
Institute of Microbial Technology (Chandigarh, India) from August 2008 till December 2014. The lab works
on multiple domains ranging from nanoparticle to structural studies of proteins. My doctoral work focused
on the structural and functional characterizations of the human SIRT1 protein and screening of activity
modulators of SIRT1 using computational and in vitro screening.
SIRT1 is a large tri-domain protein with a conserved central catalytic domain. The protein remains
conserved from archae to humans. However, its size varies considerably among the organism due to flanking
N- and C-terminal domains, with SIRT1 being the largest. Regulation of its activity has been found to play a
critical role in controlling multiple disease like type 2 diabetes, cancer, senescence. I performed structural
studies on SIRT1 using Small Angle X-ray scattering and computational modelling to decipher the global
shape of protein in solution. This data was then used for screening of various nutraceutical libraries for the
activity modulation of the protein. A manuscript on the above studies is under communication.
Simultaneously, I also assisted Dr. Raman Parkesh with the creation of EpiDbase: a database for the
epigenetic modulators, by helping in the development of website and searching the available literature for
various reported modulators. This work has been published in the Database journal in 2015.
I have also an opportunity to workwith human genetics under the supervision of Prof. R.N.K. Bamezai
at Jawahar Lal Nehru University. This work has been published in Plos One journal in 2013.
