Introduction
• Phototoxicity – is defined as a toxic acute response from a substance applied to the body which is either elicited or increased after
subsequent exposure to light, or that is induced by skin irradiation after systemic administration of a substance. (Oecd432).
• It is an acute light-induced response, occurs when photoreactive chemicals are activated by solar lights and transformed into
products cytotoxic against skin cells
• Organic chemicals like benzene or heterocyclic compounds drugs possession of up-absorbing
• Absorption of photons and absorbed energy (hv) by photoactive chemicals results in molecular changes or generates reactive
oxygen species
• Symptoms like skin irritation, erythema, pruritis, and edema similar to exaggerated sunburn.
• Test method, in vitro and in chemico assays as well as in vivo.
Medications: Some medications, such as certain antibiotics, diuretics, and non-steroidal anti-inflammatory drugs
(NSAIDs), can cause phototoxic reactions when exposed to sunlight or artificial light sources.
Chemicals: Exposure to certain chemicals, such as psoralens found in some plants, can cause phototoxicity. Other
chemicals commonly found in cosmetics, fragrances, and cleaning products may also cause phototoxic reactions.
Plants: Certain plants, such as giant hogweed, wild parsnip, and common rue, contain photosensitizing compounds that
can cause phototoxicity when they come into contact with the skin and are exposed to sunlight.
Foods: Certain foods, such as limes, figs, and celery, contain psoralens that can cause phototoxicity when consumed in
large quantities and exposed to sunlight.
UV radiation: Exposure to UV radiation from the sun or tanning beds can cause phototoxicity. UV radiation can damage
skin cells and increase the risk of skin cancer.
It is important to note that the severity of phototoxic reactions can vary depending on the individual's sensitivity to the
specific substance, the duration and intensity of exposure, and other factors such as age and overall health
CAUSES
Absorption of light: A phototoxic substance absorbs light energy, particularly in the UV-A and UV-B ranges.
Generation of reactive species: The absorbed energy causes the phototoxic substance to generate reactive
oxygen species (ROS), such as singlet oxygen and superoxide radicals.
Cell damage: The ROS produced by the phototoxic substance can damage various cellular components, such as
proteins, lipids, and DNA.
Inflammation: The damage caused by the ROS can trigger an inflammatory response, which can exacerbate the
damage and lead to tissue injury.
Photosensitivity: In some cases, phototoxicity can result in photosensitivity, which is an increased sensitivity to
light that can cause a rash, blistering, or other skin reactions upon exposure to sunlight.
All substances are not phototoxic, the severity of phototoxic reactions can vary widely depending on the
substance and the amount and duration of exposure to light. Some common phototoxic substances include
certain medications (such as tetracyclines and some diuretics), fragrances, and plant extracts (such as citrus oils)
MECHANISM OF PHOTOTOXICITY
Modes of phototoxicity
1. Direct mode is distinguished
by the existence of unstable
species which is derived from
excited state and reacts directly
with the endogenous molecules
2. Indirect mode - secondary
photoproducts of the phototoxic
compound react with the
biological targets
*Photochemical reaction*
Photoactive chemicals absorb photons and the absorbed energy (hv)
results in molecular changes of the chemical itself or generates reactive
oxygen species that can cause toxicity to the target tissues .
Intermediates of photochemical reactions include radicals and radical
ions that can directly inflict cytotoxicity.
Photo-excited molecule can also transfer energy to adjacent oxygen that
results in the formation of highly cytotoxic singlet oxygen.
Superoxide anion and hydrogen peroxide can be also generated by
photoexcitation and transfer of energy to neighbouring water molecules
and due to it toxic event occurs.
In vitro test - 3T3 NRU assay and 3D epidermis model
In vivo test - Guinea pig, mouse or pigmented rats
In chemico test
3T3 Neutral red uptake assay
Todaro in 1968. 3T3 designation stands for “3-day transfer, inoculum 3 × 10^5 cells” in 20 cm2 dish,
and this cell is relatively stable, readily available and easy to handle.
Approved by OECD and endorsed as OECD TG 432 in 13th April 2004.
the relative reduction in cell viability after the exposure to the test article in the presence versus
absence of UV/VIS irradiation.
TEST METHODS FOR PHOTOTOXICITY AND ANIMAL
ALTERNATIVES
Principle of 3T3
comparison of cell viability in the presence or absence of UV/Vis irradiation as determined with vital dye, neutral red,
which is a weak cationic dye that readily penetrates cell membranes and accumulating intracellularly in lysosomes of
viable cells.
Base cell-line is Balb/c 3T3 cell, which is mouse fibroblast developed from mouse embryos by G.T.Todaro in 1968.
Dermal fibroblast target cells of phototoxicity providing a solid rationale for the employment of 3T3 cells.
To decide whether the test article is phototoxic or not concentration-response shall be obtained in the presence and in the
absence of irradiation
Photo-Irritation-Factor (PIF) or Mean Photo Effect (MPE) shall be calculated
PIF is the ratio of IC50 (concentration that decreases cell viability by 50%) of non-
irradiated over irradiated
When IC50 cannot be obtained, MPE
IF < 2 or an MPE < 0.1 predicts: “no phototoxicity”. A PIF > 2 and < 5 or an MPE >
0.1 and < 0.15 predicts: “probable phototoxicity” and PIF > 5 or an MPE > 0.15
predicts: “phototoxicity”
Erythrocyte Hemolysis test
Cellular membranes are vulnerable to photochemically generated ROS and radicals.
UVA-induced damages of erythrocyte and resultant hemolysis.
Sheep red blood cells (SRBC) are incubated with chemicals and irradiated with UVA at 20 J/cm2.
Irradiation, SRBCs were incubated in the dark for 2 hr at room temperature and then for a further 1 hr at 37o C.
Hemolysis were measured with Drabkin’s reagent the measurement of UV absorbance at 540 nm.
Extent of phototoxicity was assessed by the release of haemoglobin from SRBC
Photohemolytic activity (%) = ADE − AD /C × 100
ADE: optical density of exposed drug solution with erythrocytes
AD: optical density of exposed drug solution without erythrocytes
C: optical density of 100% hemolytic control solution
Phototoxicants like ciprofloxacin, norfloxacin or enoxacin significantly increase photohemolytic activity beyond
20% at 100 μg/mL
In vitro human 3D epidermis model
To overcome the limitations of in vitro cell-based methods this model is investigated for
phototoxicity testing
Its test principle is similar to 3T3 NRU test, namely the assessment of the difference of tissue
viability between in the presence and absence of UV/VIS irradiation
water-insoluble materials can be tested
measurement of cytokine production like IL-1β (Interleukin-1β) , comet assay and histological
examination are possible that can be considered into the further evaluation of photoallergenicity
and photocarcinogenicity
In vivo methods employing guinea pig, mouse or pigmented
rats –
Animals are exposed to chemicals topically or systemically and irradiated with appropriate dose of
UVA
Scoring of erythema and edema from 0 to 4 is summed up and maximum score during 72 hrs of
observation is averaged per animal to generate irritation index.
Phototoxicity index is being obtained by the equation
“Irritation index of UVA irradiated - Irritation index of non-irradiated site”
Phototoxicity index over 0.6 indicates the potential of phototoxicity.
Ear thickness may be measured to estimate edema in mouse tests.
sacrifices of animals, expenses and time required to conduct the test poses many problems especially
in the era of wide-spread awareness of animal welfare and ethics.
In chemico methods for phototoxicity evaluation

phototoxic potential of a chemical can be evaluated in chemico Method..
This assay also does not require live cells or tissues, but plasmid is required.
Plasmid is dissolved in buffer and mixed with test articles.
mixture has been irradiated with UV, samples are subjected to electrophoresis. Quantity of breakage DNA is
analysed by fluorescent-based technology.
UV induced phototoxic compound results opening DNA strands and it is dependent on drug concentration and
UV irradiation dose
CONCLUSION
skin cancers, photoaging and hyper-pigmentation are in the trend of increasing frequency and
severity. As the awareness of the safety increases.
Current in vitro test methods still leave many points to be improved,
further attention and efforts from toxicologist and photo chemists is necessary which will provide a
better solution in near future. To avoid incidence like phototoxicity.

Phototoxicity / Phototoxicity testing

Introduction • Phototoxicity – is defined as a toxic acute response from a substance applied to the body which is either elicited or increased after subsequent exposure to light, or that is induced by skin irradiation after systemic administration of a substance. (Oecd432). • It is an acute light-induced response, occurs when photoreactive chemicals are activated by solar lights and transformed into products cytotoxic against skin cells • Organic chemicals like benzene or heterocyclic compounds drugs possession of up-absorbing • Absorption of photons and absorbed energy (hv) by photoactive chemicals results in molecular changes or generates reactive oxygen species • Symptoms like skin irritation, erythema, pruritis, and edema similar to exaggerated sunburn. • Test method, in vitro and in chemico assays as well as in vivo.

Medications: Some medications, such as certain antibiotics, diuretics, and non-steroidal anti-inflammatory drugs (NSAIDs), can cause phototoxic reactions when exposed to sunlight or artificial light sources. Chemicals: Exposure to certain chemicals, such as psoralens found in some plants, can cause phototoxicity. Other chemicals commonly found in cosmetics, fragrances, and cleaning products may also cause phototoxic reactions. Plants: Certain plants, such as giant hogweed, wild parsnip, and common rue, contain photosensitizing compounds that can cause phototoxicity when they come into contact with the skin and are exposed to sunlight. Foods: Certain foods, such as limes, figs, and celery, contain psoralens that can cause phototoxicity when consumed in large quantities and exposed to sunlight. UV radiation: Exposure to UV radiation from the sun or tanning beds can cause phototoxicity. UV radiation can damage skin cells and increase the risk of skin cancer. It is important to note that the severity of phototoxic reactions can vary depending on the individual's sensitivity to the specific substance, the duration and intensity of exposure, and other factors such as age and overall health CAUSES
  • 4. Absorption of light: A phototoxic substance absorbs light energy, particularly in the UV-A and UV-B ranges. Generation of reactive species: The absorbed energy causes the phototoxic substance to generate reactive oxygen species (ROS), such as singlet oxygen and superoxide radicals. Cell damage: The ROS produced by the phototoxic substance can damage various cellular components, such as proteins, lipids, and DNA. Inflammation: The damage caused by the ROS can trigger an inflammatory response, which can exacerbate the damage and lead to tissue injury. Photosensitivity: In some cases, phototoxicity can result in photosensitivity, which is an increased sensitivity to light that can cause a rash, blistering, or other skin reactions upon exposure to sunlight. All substances are not phototoxic, the severity of phototoxic reactions can vary widely depending on the substance and the amount and duration of exposure to light. Some common phototoxic substances include certain medications (such as tetracyclines and some diuretics), fragrances, and plant extracts (such as citrus oils) MECHANISM OF PHOTOTOXICITY

  • 6. Modes of phototoxicity 1. Direct mode is distinguished by the existence of unstable species which is derived from excited state and reacts directly with the endogenous molecules 2. Indirect mode - secondary photoproducts of the phototoxic compound react with the biological targets
  • 7. *Photochemical reaction* Photoactive chemicals absorb photons and the absorbed energy (hv) results in molecular changes of the chemical itself or generates reactive oxygen species that can cause toxicity to the target tissues . Intermediates of photochemical reactions include radicals and radical ions that can directly inflict cytotoxicity. Photo-excited molecule can also transfer energy to adjacent oxygen that results in the formation of highly cytotoxic singlet oxygen. Superoxide anion and hydrogen peroxide can be also generated by photoexcitation and transfer of energy to neighbouring water molecules and due to it toxic event occurs.

Approved by OECD and endorsed as OECD TG 432 in 13th April 2004. the relative reduction in cell viability after the exposure to the test article in the presence versus absence of UV/VIS irradiation. TEST METHODS FOR PHOTOTOXICITY AND ANIMAL ALTERNATIVES

Principle of 3T3 comparison of cell viability in the presence or absence of UV/Vis irradiation as determined with vital dye, neutral red, which is a weak cationic dye that readily penetrates cell membranes and accumulating intracellularly in lysosomes of viable cells. Base cell-line is Balb/c 3T3 cell, which is mouse fibroblast developed from mouse embryos by G.T.Todaro in 1968. Dermal fibroblast target cells of phototoxicity providing a solid rationale for the employment of 3T3 cells. To decide whether the test article is phototoxic or not concentration-response shall be obtained in the presence and in the absence of irradiation Photo-Irritation-Factor (PIF) or Mean Photo Effect (MPE) shall be calculated

PIF is the ratio of IC50 (concentration that decreases cell viability by 50%) of non- irradiated over irradiated When IC50 cannot be obtained, MPE IF < 2 or an MPE < 0.1 predicts: “no phototoxicity”. A PIF > 2 and < 5 or an MPE > 0.1 and < 0.15 predicts: “probable phototoxicity” and PIF > 5 or an MPE > 0.15 predicts: “phototoxicity”
  • 11. Erythrocyte Hemolysis test Cellular membranes are vulnerable to photochemically generated ROS and radicals. UVA-induced damages of erythrocyte and resultant hemolysis. Sheep red blood cells (SRBC) are incubated with chemicals and irradiated with UVA at 20 J/cm2. Irradiation, SRBCs were incubated in the dark for 2 hr at room temperature and then for a further 1 hr at 37o C. Hemolysis were measured with Drabkin’s reagent the measurement of UV absorbance at 540 nm. Extent of phototoxicity was assessed by the release of haemoglobin from SRBC Photohemolytic activity (%) = ADE − AD /C × 100 ADE: optical density of exposed drug solution with erythrocytes AD: optical density of exposed drug solution without erythrocytes C: optical density of 100% hemolytic control solution Phototoxicants like ciprofloxacin, norfloxacin or enoxacin significantly increase photohemolytic activity beyond 20% at 100 μg/mL
