Pemeriksaan alkaloid
• ± 4 gram sampel (2 lembar daun) dirajang atau potong kecil-kecil
• Masukkan secukupnya kedalam lumpang
• Tambahkan sejumput pasir, gerus hingga homogen
• Lalu, tambahkan 10 mL kloroform
• Tambahkan 10 mL kloroform amoniak 0,05 N gerus hingga warna hijau
• Kemudian saring dengan kapas, ambil dengan pipet tetes
• Masukkan ke dalam tabung reaksi besar dan tambahkan H2SO4 2N
• Kocok perlahahan hingga terbentuk dua lapisan lalu, ambil lapisan asam
• Masukkan kedalam tabung reaksi kecil dan tambahkan 2 tetes preaksi mayer
• Amati hasilnya, jika terbentuk kabut putih/ endapan putih berarti hasilnya positif
Cara Kerja
*
•Pemeriksaan Flavonoid, Fenolik, Saponin, Terpenoid, Steroid
•± 4 gram sampel (2 lembar daun) dirajang atau potong kecil-kecil
•Masukkan kedalam tabung reaksi besar, ditambah etanol sampai terendam
•Panaskan diatas bunsen hingga larutan setengah
•Kemudian saring dengan kapas yang dimasukkan kedalam pipet tetes kecil dan di tampung dengan tabung reaksi besar
•Lalu, tambahkan kloroform dan aquadest sebanyak 1 : 1 kedalam tabung reaksi
•Kocok dan diamkan hingga terbentuknya dua lapisan yaitu lapisan atas kloroform dan lapisan bawah aquadest
Flavanoid (Sianidin Test) :
•Masukkan 1 mL lapisan airnya kedalam tabung reaksi, tambahkan logam Mg sedikit dan ditambah 2 tetes H2SO4 Pekat (
hasil positifnya terbentuk warna orange/ merah/ kuning)
Fenolik :
•Masukkan 1 mL lapisan airnya kedalam tabung reaksi, tambahkan 2 tetes FeCl3 1% (hasil positifnya terbentuk warna biru
kehitaman)
Saponin :
•Masukkan 1 mL lapisan airnya kedalam tabung reaksi, lalu kocok biarkan selama 5 menit (hasil positifnya terbentuknya
busa)
Terpenoid & Steroid :
•Ambil lapisan kloroform lalu masukkan kemudian saring dengan pipet yang sudah dimasukkan kapas yang berisi norit
•Lalu, hasil saringan yang bewarna bening diletakkan diplat tetes dan keringkan
•Tambahkan 1 tetes anhidrat asetat dan H2SO4 Pekat (pereaksi Liebermann Burchard)
•Setelah itu amati perubahan yang terjadi, hasil positifnya jika terbentuk warna merah berarti mengandung terpenoid,
bewarna hijau berarti mengandung steroid dan jika bewarna ungu berarti mengandung terpenoid dan steroid
