Cefpodoxime proxetil and potassium clavulanate calculation by BISHNU ADHIKARI
1. Label claim: Each Film coated tablets contains:
Cefpodoxime proxetil eq. to cefpodoxime 200 mg
Clavulanate potassium equivalent to clavulanic acid 125 mg
For cefpodoxime proxetil as per company Certificate of Analysis (COA)
738 µg/mg as on Anhydrous basis (As Cefpodoxime)
0.738 mg = 1 mg
200 mg = 1/0.738*200
= 271.0 mg (Cefpodoxime Proxetil)
For PotassiumClavulanate with MCC (1:1)
Potassium Clavulanate (C8H8KO5) = 237.25 (Molecular Weight)
Clavulanic acid (C8H8NO5) = 199.0 (Molecular Weight)
199 = 237.25
Now as per label claim: 125 mg we need.
=237.25/199*125
= 149.026 mg (Potassium Clavulanate)
For 100% , Potassium Clavulanate with MCC (1:1) , it should divide by assay
= 149.026/99.18*100
= 150.26 mg.
Now required to add water content also i.e. 0.74%
= 150.26 + 0.74
=151.37 mg required of potassium clavulanate with MCC.
Similarly,
For PotassiumClavulanate with Syloid (1:1)
For 100% Potassium Clavulanate with Syloid (1:1)
=149.026/99.59*100
=149.64mg + 0.64% (water content in %)
= 150.598 mg required of potassium clavulanate with syloid.
