This document discusses genetics and late onset Alzheimer's disease. It notes that late onset Alzheimer's disease (LOAD) is the most common form of Alzheimer's in older individuals. The Apolipoprotein E (APOE) gene, specifically the APOE*4 allele, is a major genetic risk factor for late onset Alzheimer's and accounts for about 30% of risk. However, the APOE*4 allele alone is not sufficient to cause Alzheimer's, suggesting other genetic or environmental factors also influence risk. Recent genome-wide studies have provided evidence of additional potential Alzheimer's risk genes on several chromosomes, with the strongest links found on chromosomes 12, 10, 9 and 6.
