Another way biological control has been developed as an alternative of chemicals to tock with plant
pathogenic fungi. Considering high presence of chitin in fungal cell wall, chitinase enzyme is camped as an
effective biocontrol agent against phytopathogenic fungi. Streptomyces bacteria are able to produce various chitinase enzymes, chitinases produced by streptomyces belong to the families 18 and 19 glycosyl hydrolases.
The antifungal activity is mostly shown by fomily 19 Chitinases. In comparison with bacterial family 18
chitinases, the specific hydrolyzing activity of chitinase 19 against soluble and in soluble chitinous substrates
has been markedly higher. Considering the importance of family to investigate antifungal potential of
streptomyces bacteria isolated from east Azarbijan region soils based on molecular identification of family 19
chitinase. encoding gene in these bacteria.
To aim the purpose 110 soil samples were collected from East Azarbaijan and 310 strepomyces
isolates were selected using macroscopic and microscopic observations. DNA genomic of all of the isolates
were extracted and PCR reactions was done using chitinase 19 designed primers as marker.
Totally isolates were selected with molecular selection and antagonistic test were done. One of the isolates
exhibit the most strong antifungal activity.
The strain was identified using 16srDNA gene, and the chitinase encoding gene were amplified partially to
prove the PCR selection. Finally the bacterium were introduced as potentially biological fertilizer.

— The microbiological content of Lettuce (a vegetable), commonly vended in the Benin metropolis of Edo state were evaluated. Five vending locations were chosen for the study. Whole and soft rot samples were purchased and analysed for microbiological composition. Results showed high counts in soft rot samples in lettuce. Nutrient agar plated lettuce samples had bacterial counts in the range of 2.0x 103 to 4.7x10 7. Pseudomonas species was the dominant species found in lettuce samples. Bacillus species was isolated from one location in the lettuce samples. Mac Conkey agar plated lettuce plated had bacterial counts in the range of 2.3 x 10 3 to 5.7x 10 7. Enterobacter species, E. coli, and Klebsiella species were the dominant species isolated. Though, Proteus species was isolated from lettuce samples obtained from location five only. The study observes that consuming soft rot samples could pose a risk of introducing pathogens to the consumer due to their high microbial counts and could be detrimental to the health of the consumer.
Effect of Compost Extract Fortified with Tempe on Chili Mosaic Virus DiseaseIJEABJ
Mosaic disease caused by multiple infections of viruses in one of the most devastating virus diseases of chili pepper (Capsicum annuum) in Indonesia. Improving plant resistance by treatment with exogenous bioactive compounds is promisingly developed for plant protection in organic chili production. We demonstrated the suppressive effects of a fermented water extract of compost fortified with over-fermented tempe (TCE) on mosaic disease and its aphid vector. TCE was applied weekly by foliar spraying at 0.2 and 2.0 % on potted Capsicum growing in field. The result showed that treated plant sprayed with TNF preparation exhibited a significantly slower disease progression as represented by a lower area under disease progress curve compared to control plant. Disease suppression was obtained at concentration as low as 0.2%. TCE-treated plants were significantly less colonized by Aphis gossypii than control plant.

In the present work an attempt has been made to carry out screening for the preliminary antibacterial activity of different plants used by Sugali tribes of Yerramalis forest. Fifteen plants were selected for preliminary screening for their antibacterial potentiality, The antibacterial activity was done against four bacterial strains, viz., Bacillus cereus, Staphylococcus aureus, Klebsiella pneumoniae, Escherichia coli,.The preliminary screening experiment revealed that methanol extracts were more potent than the aqueous extracts. The most susceptible bacteria were K. pneumoniae and the most resistant bacteria were E. coli. Bauhinia racemosa L. exhibited remarkable antibacterial activity.

Plant growth promoting rhizobacteria (PGPR), are associated with roots, found in the rhizosphere and can directly or indirectly enhance the plant growth. In this study soil was collected from rhizosphere of chickpea fields of different areas of Rawalpindi division of Pakistan. PGPR were isolated, screened and characterized. Eight isolates of rhizobacteria (RHA, RPG, RFJ, RC, RTR, RT and RK) were isolated from Rawalpindi division and were characterized. The antagonistic activity of these PGPR isolates against root infecting fungi (Fusarium oxysporum and Verticillium spp.,) was done and production of indole acetic acid (IAA), siderophore and P-solubilization was evaluated. The isolates RHA, RPG, RFJ, RC, RRD and RT were found to be positive in producing siderophore, IAA and P-solubilization. Furthermore, most of the isolates showed antifungal activity against Fusarium oxysporum, and Verticillium spp. The rhizobacterial isolates RHA, RPG, RFJ, RC, RRD, RTR, RT and RK were used as bio-inoculants that might be beneficial for chickpea cultivation as the rhizobacterial isolates possessed the plant growth promoting characters i.e. siderophore, IAA production, phosphate solubilization. In in vitro tests, Pseudomonas sp. and Bacillus spp. inhibited the mycelial growth of the fungal root pathogens. The isolates (RHA and RPG) also significantly increased (60-70%) seed germination, shoot length, root length of the chickpea. The incidence of fungi was reduced by the colonization of RHA and RPG which enhanced the seedling vigor index and seed germination. The observations revealed that isolates RHA and RPG is quite effective to reduce the fungal root infection in greenhouse, and also increases seed yields significantly. These rhizobacterial isolates appear to be efficient yield increasing as well as effective biocontrol agent against fungal root pathogen.
Prevalence, occurrence and biochemical characterization of Xanthomonas campes...INNS PUBNET
Xanthomonas campestris pv. vesicatoria the causal organism of bacterial spot in tomato results in heavy losses both in the form of quality and. In this study a survey was carried out to report the incidence of bacterial spot disease of tomato in district Swat. We reported maximum disease incidence in tehsil Kabal (71.66%), followed by Charbagh (61.66%) and Barikot (58.33%). For resistant screening a total of 13 tomato germplasms were screened against the disease. The foliar severity ranged from 3.33% to 73.33%, while severity for fruits was ranged from 18.33% to 30.66%. In case of phenotypic data the highest numbers of fruits obtained were 34, plant height 79.5cm and fruit weight was 470 grams/ten tomatoes. While the lowest average numbers of fruits were 6.67, plant height 45.7cm and fruit weight recorded was 215.67 grams/ten tomatoes. Line 1288 showed highest level of resistance followed by Red-stone. However, line 9708 showed highest susceptibility when exposed to artificial inoculation. Our study showed that bacterial spot is a major issue in some part of Pakistan and germplasm screening are linked to increased host resistance and could offer an important contribution to future integrated bacterial spot management programs.

Plant endophytes may be intercellular or intracellular depending upon their location in the plant tissue because they are present inside the cells or in the intracellular space, respectively. Isolation of endophytic bacteria has been reported from both monocot and dicot plants, ranging from woody trees, such as teak and pear, to herbaceous crop plants such as mustard and maize. The aim of this study was the isolation of endophytes from potato and their antagonist effect against Fusarium oxysporum. Endophytic fungi were isolated from leaves, stems and roots of healthy Potato plant derived from Chak No.359/E.B Village, Tehsil Burewala. Isolation of endophytic fungi from plant parts was done according to the method described by Petrini. The media used in the present study was the Potatodextrose agar (PDA) for fungus and nutrient agar medium for maintaining bacterial stains. F.oxysporum was taken from the Plant pathology lab of UAF sub-campus Burewala-Vehari . The results of the experiment clearly revealed that the stems, root and leaf of the potato plants under present investigation had the maximum colonization frequency for fungal endophytes. Fusarium oxysporum showed rapid growth 5-7cm in5 days. Fusarium oxysporum was white and growing rapidly that later produced dark violet pigments in PDA. Erwinia showed light green, circular, shining, slimy, smooth characteristics. The isolate strain of Bacillus showed rodshaped, fuzzy white or slightly yellow circular and irregular characteristics.

Plant Growth promoting rhizobacteria are a heterogeneous group of bacteria that can be found in the rhizosphere, at root surfaces and in association with roots. They benefit plants through Production of plant hormones, such as auxins, asymbiotic N2 fixation, solubilization of mineral phosphates, antagonism against phytopathogenic microorganisms by production of antibiotics, siderophroes, Chitinase and other nutrients ability to effectively colonize roots are responsible for plant growth promotion. An experiment was conducted in the field of National Institute of Agronomic Research of Meknes. Morocco. The experiment was a completely randomized design with six replicates. There were four treatments viz. T1: (control; N0 -PGPR), T2: (N0 +2027-2), T3: (N0 +2066-7) and T4: (N0+2025-1). The results indicated that a remarkable increase in root growth, namely length, the diameter of the rod and the total chlorophyll. A total of three different bacteria colonies were isolated and proceed with in vitro screening for plant growth promoting activities; phosphate solubilization, nitrogen fixation, indole acetic acid (IAA), ammonia production and antimicrobial enzymes (cellulose, chitinase and protease) activity. Among the three bacterial strains, all bacterial strains are able to produce ammonia, IAA production and nitrogen fixation activity, one strain phosphate solubilizing activity, two strain are able to produce cellulase syntheses, Protease activity and Chitinase activity.
Potentials of Compost Tea of Certain Botanicals for Minimizing Root- Knot and...Premier Publishers
Two greenhouse experiments were conducted on eggplant to assess the impact of compost tea derived from plant residues namely rice hull (RHC), rice straw (RSC), tomato (TC), potato (PC), citrus (CC), and guava(GC) as well as city waste (CWC) compost on eggplant biomass and reproduction of Meloidogyne incognita and Rotylenchulus reniformis. Screened composts were introduced to plants as drenching application. Results indicated that percentage of increase in total plant fresh weight of eggplant infected with such nematodeswere more pronounced (P<0.05) with compost teas of RSW. Drenching the soil with RHC (Rf=0.7) and PC (Rf=0.9) were also effective in suppressing densities of M. incognita as well as number of galls, and eggs/ root. However, population densities and fecundity of R. reniformis were significantly reduced following the introduction of GC. Only, total phenol showed remarkable increment in plants treated with PC, GC and RSC compared to untreated inoculated plants.

Potential of Entomopathogenic Bacillus thuringiensis as Plant Growth Promotin...Agriculture Journal IJOEAR
Abstract— Bacillus thuringiensis has been used as an effective bioinsecticide because it produces the proteins Cry and Cyt, which are highly toxic to insects in certain situations. However, recently, B. thuringiensis was used as a biological control agent that can suppress plant disease. In this study, the antagonistic activities of B. thuringiensisAS17 japonensisand AS18 kurstaki against the fungal pathogen Fusarium oxysporum f.sp. lycopersici (FOL) were examined using a dual culture technique. Furthermore, B. thuringiensis strains suppressed the development of wilt symptoms caused by FOL in tomato plants. After inoculating six strains of B. thuringiensis suspension following inoculation of FOL, the development of wilt symptoms became less than control, especially with B. thuringiensisAS17 japonensis and AS20 CR371-H. Furthermore, we proved that B. thuringiensis strains are plant growth promoting rhizobacteria (PGPR) that can promote plant growth. Seed germination and shoot elongation were promoted by treating the tomato seeds with a bacterial culture filtrate and a bacterial suspension.

Antibacterial Activity of Leaf Methanolic Extract of S. Caryophyllatum (L.) A...iosrjce
S.caryophyllatum (L.) Alston belongs to the family Myrtaceae is an endangered species. It possesses
traditional as well as pharmacological properties. The objective of the present investigation was to find out the
antibacterial activity of S. caryophyllatum leaf methanolic extract against some human pathogenic bacteria. It
was followed by Disc Diffusion method using gram positive and gram negative bacterial strains such as
Staphylococcus aureus, Bacillus cereus, Bacillus subtilis, Bacillus megaterium, Sarcina lutea, Esherichia coli,
Pseudomonas aeruginosa, Klebsiella Spp., Salmonella typhi and Proteus mirabilis. The result showed that the
inhibitory effect on Bacillus subtilis (24mm) was high when compared to E. coli (21mm) and Bacillus cereus
(20mm).This effect on the bacterial strains may be due to the presence of secondary metabolites present in the
leaf methanolic extract of Syzygium caryophyllatum.

Plant growth promoting rhizobacteria (PGPR), are associated with roots, found in the rhizosphere and can directly or indirectly enhance the plant growth. In this study soil was collected from rhizosphere of chickpea fields of different areas of Rawalpindi division of Pakistan. PGPR were isolated, screened and characterized. Eight isolates of rhizobacteria (RHA, RPG, RFJ, RC, RTR, RT and RK) were isolated from Rawalpindi division and were characterized. The antagonistic activity of these PGPR isolates against root infecting fungi (Fusarium oxysporum and Verticillium spp.,) was done and production of indole acetic acid (IAA), siderophore and P-solubilization was evaluated. The isolates RHA, RPG, RFJ, RC, RRD and RT were found to be positive in producing siderophore, IAA and P-solubilization. Furthermore, most of the isolates showed antifungal activity against Fusarium oxysporum, and Verticillium spp. The rhizobacterial isolates RHA, RPG, RFJ, RC, RRD, RTR, RT and RK were used as bio-inoculants that might be beneficial for chickpea cultivation as the rhizobacterial isolates possessed the plant growth promoting characters i.e. siderophore, IAA production, phosphate solubilization. In in vitro tests, Pseudomonas sp. and Bacillus spp. inhibited the mycelial growth of the fungal root pathogens. The isolates (RHA and RPG) also significantly increased (60-70%) seed germination, shoot length, root length of the chickpea. The incidence of fungi was reduced by the colonization of RHA and RPG which enhanced the seedling vigor index and seed germination. The observations revealed that isolates RHA and RPG is quite effective to reduce the fungal root infection in greenhouse, and also increases seed yields significantly. These rhizobacterial isolates appear to be efficient yield increasing as well as effective biocontrol agent against fungal root pathogen.

Xanthomonas campestris pv. vesicatoria the causal organism of bacterial spot in tomato results in heavy losses both in the form of quality and. In this study a survey was carried out to report the incidence of bacterial spot disease of tomato in district Swat. We reported maximum disease incidence in tehsil Kabal (71.66%), followed by Charbagh (61.66%) and Barikot (58.33%). For resistant screening a total of 13 tomato germplasms were screened against the disease. The foliar severity ranged from 3.33% to 73.33%, while severity for fruits was ranged from 18.33% to 30.66%. In case of phenotypic data the highest numbers of fruits obtained were 34, plant height 79.5cm and fruit weight was 470 grams/ten tomatoes. While the lowest average numbers of fruits were 6.67, plant height 45.7cm and fruit weight recorded was 215.67 grams/ten tomatoes. Line 1288 showed highest level of resistance followed by Red-stone. However, line 9708 showed highest susceptibility when exposed to artificial inoculation. Our study showed that bacterial spot is a major issue in some part of Pakistan and germplasm screening are linked to increased host resistance and could offer an important contribution to future integrated bacterial spot management programs.

Plant endophytes may be intercellular or intracellular depending upon their location in the plant tissue because they are present inside the cells or in the intracellular space, respectively. Isolation of endophytic bacteria has been reported from both monocot and dicot plants, ranging from woody trees, such as teak and pear, to herbaceous crop plants such as mustard and maize. The aim of this study was the isolation of endophytes from potato and their antagonist effect against Fusarium oxysporum. Endophytic fungi were isolated from leaves, stems and roots of healthy Potato plant derived from Chak No.359/E.B Village, Tehsil Burewala. Isolation of endophytic fungi from plant parts was done according to the method described by Petrini. The media used in the present study was the Potatodextrose agar (PDA) for fungus and nutrient agar medium for maintaining bacterial stains. F.oxysporum was taken from the Plant pathology lab of UAF sub-campus Burewala-Vehari . The results of the experiment clearly revealed that the stems, root and leaf of the potato plants under present investigation had the maximum colonization frequency for fungal endophytes. Fusarium oxysporum showed rapid growth 5-7cm in5 days. Fusarium oxysporum was white and growing rapidly that later produced dark violet pigments in PDA. Erwinia showed light green, circular, shining, slimy, smooth characteristics. The isolate strain of Bacillus showed rodshaped, fuzzy white or slightly yellow circular and irregular characteristics.

Plant Growth promoting rhizobacteria are a heterogeneous group of bacteria that can be found in the rhizosphere, at root surfaces and in association with roots. They benefit plants through Production of plant hormones, such as auxins, asymbiotic N2 fixation, solubilization of mineral phosphates, antagonism against phytopathogenic microorganisms by production of antibiotics, siderophroes, Chitinase and other nutrients ability to effectively colonize roots are responsible for plant growth promotion. An experiment was conducted in the field of National Institute of Agronomic Research of Meknes. Morocco. The experiment was a completely randomized design with six replicates. There were four treatments viz. T1: (control; N0 -PGPR), T2: (N0 +2027-2), T3: (N0 +2066-7) and T4: (N0+2025-1). The results indicated that a remarkable increase in root growth, namely length, the diameter of the rod and the total chlorophyll. A total of three different bacteria colonies were isolated and proceed with in vitro screening for plant growth promoting activities; phosphate solubilization, nitrogen fixation, indole acetic acid (IAA), ammonia production and antimicrobial enzymes (cellulose, chitinase and protease) activity. Among the three bacterial strains, all bacterial strains are able to produce ammonia, IAA production and nitrogen fixation activity, one strain phosphate solubilizing activity, two strain are able to produce cellulase syntheses, Protease activity and Chitinase activity.
Potentials of Compost Tea of Certain Botanicals for Minimizing Root- Knot and...Premier Publishers
Two greenhouse experiments were conducted on eggplant to assess the impact of compost tea derived from plant residues namely rice hull (RHC), rice straw (RSC), tomato (TC), potato (PC), citrus (CC), and guava(GC) as well as city waste (CWC) compost on eggplant biomass and reproduction of Meloidogyne incognita and Rotylenchulus reniformis. Screened composts were introduced to plants as drenching application. Results indicated that percentage of increase in total plant fresh weight of eggplant infected with such nematodeswere more pronounced (P<0.05) with compost teas of RSW. Drenching the soil with RHC (Rf=0.7) and PC (Rf=0.9) were also effective in suppressing densities of M. incognita as well as number of galls, and eggs/ root. However, population densities and fecundity of R. reniformis were significantly reduced following the introduction of GC. Only, total phenol showed remarkable increment in plants treated with PC, GC and RSC compared to untreated inoculated plants.

Potential of Entomopathogenic Bacillus thuringiensis as Plant Growth Promotin...Agriculture Journal IJOEAR
Abstract— Bacillus thuringiensis has been used as an effective bioinsecticide because it produces the proteins Cry and Cyt, which are highly toxic to insects in certain situations. However, recently, B. thuringiensis was used as a biological control agent that can suppress plant disease. In this study, the antagonistic activities of B. thuringiensisAS17 japonensisand AS18 kurstaki against the fungal pathogen Fusarium oxysporum f.sp. lycopersici (FOL) were examined using a dual culture technique. Furthermore, B. thuringiensis strains suppressed the development of wilt symptoms caused by FOL in tomato plants. After inoculating six strains of B. thuringiensis suspension following inoculation of FOL, the development of wilt symptoms became less than control, especially with B. thuringiensisAS17 japonensis and AS20 CR371-H. Furthermore, we proved that B. thuringiensis strains are plant growth promoting rhizobacteria (PGPR) that can promote plant growth. Seed germination and shoot elongation were promoted by treating the tomato seeds with a bacterial culture filtrate and a bacterial suspension.

ANTIFUNGAL ACTIVITY OF SELECTED MEDICINAL PLANT EXTRACS AGAINST PLANT PATHOG...IJSIT Editor
The aim of this work was to find an alternative to chemical fungicides currently used in the control
plant pathogenic fungi Rhizoctoniasolani ,ColletotrichummusaeandFusariumoxysporum,. The antifungal
activity of the methanol extracts of six medicinal plants used in native medicine in Sri Lanka is reported.All
plant extracts were screened for their fungistatic, fungicidal activities and minimum inhibitory dilution (MID)
against above fungi. The media amended with methanol and recommended fungicide for respective fungal
strain were consider as negative and positive control respectively.Results showed that radial growth in all the
three tested organisms was significantly impaired (p<0.05) by the addition of the extracts in the culture
medium used. The test fungi differed in their reaction to the different extracts but on the whole, growth
inhibition increased with the concentration of each extract. The most active extracts, shows a marked effect of
the 20% methanol extracts from sweet flag with inhibition values of 91%, 86% and 84 % for F. oxysporum,R.
solani and C.muceawhereas those from wild basil inhibited the growth of the same pathogens by 89%, 84%
and 74%.The results showed minimal inhibitory concentrations (MIC) were 5 % (v/v) for sweet flag and wild
basil and 20% (v/v) for all other plant crude extracts. Out of six plants extract screened, wild basil and sweet
flag showed more than 80% fungal inhibition after 6 hour immersion and other extracts could not exceed
60% inhibition after any exposure time. The study revealed that methanol crude extract of sweet flag and
wild basil exhibit strong fungistatic and fungicidal activities against tested fungi. These results support the
potential use of these plant extracts in the management of diseases caused by tested plant pathogenic fungi.
Evaluate the Efficiency of Gamma Irradiation and Chitosan on Shelf-Life of St...IJEABJ
Chitosan play an important role as an antifungal against Botrytis cinerea and the effect was a concentration dependent. The obtained results of in vitro experiment demonstrated that chitosan (4%) decreased radial growth of B. cinereato 2 %. Invivo the severity of infection reduced from 59.8 and 100.0 to 9.7, 33.8 and 40.1 in first, second and third week’s storage periods at 13C, respectively. Also, chitosan coating (4%) significantly caused an increase in fruit firmness whereas TSS was decreased with an increase by increasing in storage time. However,Vitamin C gave fluctuated results by increasing storage time. Gamma irradiation at 2.5 KGy reduced severity (%) of infected fruits from 55.5, 100 and 100 to 31.7, 45.9 and 49.9 and in healthy fruits severity (%) reduced from 48.9, 100 and 100 to 23.3, 25.1 and 29.1 in different storage periods 1, 2 and 3 weeks, respectively. Similarly, chitosan as well as gamma irradiation combination induced a significant increase of peroxidase enzyme (POD) activity. Induced changes in surface morphology and damage of cell structure caused by using chitosan shown by scanning electron microscopy. Also, gamma irradiation causes changes in hyphea structure and in surface morphology but combination of gamma irradiation with chitosan was more effective in altering fungus morphology and cell structure damage and no spore forming. This providing the efficiency of combination on reducing disease severity (%) of strawberry.

ABSTRACT- Small Cardamom (Elettariacardamomum L. Maton) is one of the major spice crops of India, which were
the world’s largest producer and exporter of cardamom till 1980. There has however been a reduction in production,
mainly because of Katte disease, caused by cardamom mosaic virus (CdMV) a potyvirus. Viral diseases can be managed
effectively by early diagnosis using serological methods. In the present investigation, CdMV isolates were sampled from
Mudigere, Karnataka, ultra purified, and electron micro graphed for confirmation. Polyclonal antibodies were raised
against the virus and a direct antigen coating plate Enzyme linked immunosorbent Assay (DAC-ELISA) and Dot-ELISA
(DIBA) standardized to detect the virus in diseased and tissue cultured plants. Early diagnosis in planting material will aid
in using disease free material for better yields and hence increased profit to the farmer. Key-words- Cardamom mosaic virus (CdMV), Electron microscopy, Direct antigen coating Enzyme linked

— Qualitative and quantitative analysis of selected mycotoxins has been performed in extracts of Conidiobolus coronatus pathogenic fungus cultivated under optimal and stress conditions. Furthermore, the analyses of these compounds in post-incubation filtrates were done. For identification purposes the analytical method allows identification and quantitation of selected mycotoxins including beauvericin , fumonisin B1, enniatin A and B and destruxin A based on high performance liquid chromatography coupled with tandem mass spectrometry was developed. Only beauvericin was detected in very low amounts in C. coronatus mycelium extract cultivated under optimal condition. In the extract of C. coronatus mycelium grown on LB 12.3 ± 0.1 µg/g of beauvericin was determined, while in the extract of C. coronatus mycelium grown on MM medium beauvericin content was lower and amounted 4.6 ± 0.1 µg/g. Also the presence of beauvericin was confirmed in postincubaction filtrate extract (MM). The content of this compound was 2.2 ± 0.1 µg/g. In other extracts beauvericin was not detected. In addition, in the tested extracts other compounds were not detected.

Ethanol (CH3CH2OH), or beverage alcohol, is a two-carbon alcohol
that is rapidly distributed in the body and brain. Ethanol alters many
neurochemical systems and has rewarding and addictive properties. It
is the oldest recreational drug and likely contributes to more morbidity,
mortality, and public health costs than all illicit drugs combined. The
5th edition of the Diagnostic and Statistical Manual of Mental Disorders
(DSM-5) integrates alcohol abuse and alcohol dependence into a single
disorder called alcohol use disorder (AUD), with mild, moderate,
and severe subclassifications (American Psychiatric Association, 2013).
In the DSM-5, all types of substance abuse and dependence have been
combined into a single substance use disorder (SUD) on a continuum
from mild to severe. A diagnosis of AUD requires that at least two of
the 11 DSM-5 behaviors be present within a 12-month period (mild
AUD: 2–3 criteria; moderate AUD: 4–5 criteria; severe AUD: 6–11 criteria).
The four main behavioral effects of AUD are impaired control over
drinking, negative social consequences, risky use, and altered physiological
effects (tolerance, withdrawal). This chapter presents an overview
of the prevalence and harmful consequences of AUD in the U.S.,
the systemic nature of the disease, neurocircuitry and stages of AUD,
comorbidities, fetal alcohol spectrum disorders, genetic risk factors, and
pharmacotherapies for AUD.

Inhibitory Effects of Aqueous Onion and Garlic Extracts on Growth
and Keratinase Activity in Tricophyton mentagrophytes
2. Abstract
The effect of onion and garlic extracts on fungal growth and keratinolytic
activity was studied in Tricophyton mentagophytes as one of the major
etiological agents of human and animal dermatophytosis in Iran and other
parts of the world.
To findout the best keratin producer for further steps,culture conditions of
30 strains of T.mentagrophytes isolated from human dermtophytosis were
optimized on specific solid and liquid media.
Extracellular keratinase activity was in range of 0.28 to 2.18u/mg in
optimal conditions .
The growth of T.mentagophytes was inhibited due to presence of various
concentrations of onion and garlic extracts i.e. it was decreased up to
100% for both extracts 10% v/v concentrations. These substances may
have potential values for treatment of human and animal dermatophytes.
3. INTRODUCTION
Dermatophytes are fungal organisms that require keratin for growth. This fungi
can cause superficial infections of hair, skin and nails. Dermatophytes are
spread by direct contact from other people, animals and soil.
Keratinases are the key enzymes in fungal invasion of skin and have been
mostly studied in dermatophyte species belonging to the trichophyton and
microsporum genera.
T. Mentagrophytes is a well-known producer of keratinases and this ability is
probably essential for invasion of host tissues. This fungus is unique among
the dermatophytes and some other zoonotic organisms for its high prevalence
and distribution and also for several pathogenic varieties.
Indeed, drug resistance problems have already occurred for dermatophytes
after several applications. Thus, many research programs have been conducted
to find out new natural and synthetic compounds with antifungal properties and
minimum side effects.
4. Two naturally occuring phytochemicals i.e. onion and garlic have
antimicrobial activities which prevent the growth of viral diseases.
In the present study, 30 isolates of T. mentagrophytes obtained from
patients with dermatophytosis have been studied. This is the first report
on inhibition of fungal keratinase as an important virulence factor by
aqueous onion and garlic extracts.
5. Methods
Screening of keratinolytic activity
Screening of keratinolytic activity on agar plates.
Preparation of aqueous onion and garlic extracts.
Submerged cultivation.
Keratinase activity assay.
6. Agar plate Screening
Screening of 30 T.mentagrophytes isolates
for keratinase production on solid mineral
medium showed that all of the examined
isolates were able to produce extracellular
keratinase at different levels.
Keratinolytic activity was assessed based on
the observation of a clear zone around the
fungal colony on the plate .
The diameter of clear zone was in the range
of 9.5to 47.5 mm among the isolates.
There was a significant difference in
keratinolytic activity on solid medium among
some isolates .
7. Optimization of keratinolytic activity
in submerged shaken cultures
The fungus was cultured on this medium and keratinase activity
was measured after incubation in different culture conditions.
The maximum keratinolytic activity was obtained for 12 days
cultures maintained at 32 C.
The fungal growth rate was between 1.5 to 23.5mg dry weight of
50 ml cultures among the isolates.
Extracellular keratinase specific activity was measured as 0.28 to
2.18 u/mg protein among the isolates (Fig. 2). Total keratinase
activity of the isolates was ranged between 0.65 to 4.02 u/50 ml
culture media.

9. Effects of aqueous onion and garlic extracts on
fungal growth and keratinase production.
T.mentagrophytes cultured in the presence of different concentrations of
both extracts and the results showed that fungal growth was significantly
inhibited by both onion and garlic extracts in all concentrations as
compared with controls.
Minimum inhibition for onion and garlic extracts was measured as 59% and
35% at 0.62% v/v concentrations, respectively and maximum
concentrations of both extracts at 10 % v/v .
Aqueous onionand garlic extracts at the lowest concentrations (0.62 %,
v/v) caused approximately 26% and 58% inhibition in enzyme activity by
the fungus.
The maximum inhibition of keratinolytic activity of 5% (v/v) concentration
of onion and garlic extracts was 57.54% and 71.36%, respectively .

11. Conclusion
. The data shows that onion and garlic extracts can be used as
potential candidates for preparation of anti-dermatophytic
drug formulations and thus may be useful in the treatment of
different kinds of dermatophytosis in human and animals .
