• Place the microscope on a clean, flat surface

B. Preparing Microscope Slides
• Wash your hands
• Use a lint-free cloth for cleaning
• Start with prepared slides
• Place the slides on the stage
• Secure the slide with the stage clips
• Turn on the microscope

• Adjust the eyepiece
• Open the diaphragm to its widest setting
• Start with the LPO lens
• Center the specimen on the stage
• Focus the slide

1. What are some common mistakes people make when
using a microscope?
– Touching the lens may cause the specimen to appear blurry
– Incorrectly handling the microscope. (Note: Always handle the
microscope by gripping the arm and placing your palm on the
bottom of the base)
– Using the coarse adjustment knob in High Power Objective (Note:
You shall never do this as it would cause damage and crack to your
lens and your slide.)
2. How do I know if the microscope is properly illuminated?
– The specimen is properly illuminated if we could see the specimen
clearly and with a good resolution.
3. Why is it important to start with LPO lens?
– Because LPO or Low power objective has the higher field of view.
Hence, it is easier to locate the specimen on the slide than if you
start with a higher power objective.
