Research undertaken during Ph.D.
During my doctoral research I have gained expertise in Cotton and Tobacco tissue culture and its
genetic transformation using Agrobacterium method. Also developed insect resistant cry2AX1
transgenic cotton plants through somatic embryogenesis, in the Coker 310 background and did its
molecular characterization, which included PCR, RT-PCR, Southern blot hybridization analysis,
qualitative and quantitative ELISA and insect bioassay. Besides this I gained experience in transgenic
elite event characterization in advanced T3 generation progeny for stability of gene expression using
PCR, southern blot analysis, ELISA and Helicoverpa armigera insect bioassay to assess the bio-
efficacy Bt protein. Further I worked on development of new plant expression constructs for
enhanced expression of cry2AX1 (Bt) gene in plant and their expression validation in tobacco.
Besides, I have experience in various bioinformatics tools viz. nucleotide sequence assembly tools,
sequence alignments tools, construction of phylogenetic trees.
