OU-DST-PURSE PROJECT PROGRAMME. (September 2011 –2014)
Designation: Project Fellow. Worked as a Project fellow in the project entitled “Molecular
characterization of endangered medicinal plants, Saraca asoca and Commiphora wightii and
their conservation through Micropropagation” at OU-DST-PURSE Osmania University,
Hyderabad,
Ph.D. Thesis Summary (March 2011- 2014).
• To collect ecotypes of Ashok and Guggul from different places of South India and raise
them in the Botanical Garden.
• Standardized the protocols for efficient in vitro plant regeneration via multiple shoot from
different explants (nodal, inter nodal and shoot tip) of Saraca asoca and Commiphora
wightii.
• To carry out chemical analysis (HPLC) of the collected ecotypes for the principal
chemical constituents to help identify the high-yielding elites.
• To characterize the ecotypes by using molecular markers (RAPD).
• To develop workable protocols for micropropagation of the elites to aid conservation.

Plant tissue culture: Standardization of plant regeneration system from various explants
through direct and indirect organogenesis, somatic embryogenesis, and synthetic seed
preparation.
HPLC, TLC and analytical techniques and Phytochemical analysis techniques
Plant genomic DNA Isolation and purification.
Standardization of annealing temperature in PCR for amplification of selected genes
using gradient system.
Performed RAPDs to analyze the difference between transgenic and control plants using
Operon primers.
Expert in antimicrobial activity and antioxidant activity techniques.
Known Techniques in Plant Anatomy, Taxonomy and morphology studies.
