Project title: Synthesis, purification and characterization of
i) calix[n]phyrin (n=4, 6) conjugates with Tröger’s base (TB) for sensing cations or anions, and
ii) TB-Tris(mannose) cluster
Accomplishments: Multi-step synthesis of Calix[n]phyrin-Tröger’s base (TB) (n= 4, 6) conjugates
was performed with the aim to construct a possible molecular tweezer for sensing cation or anion
(Scheme 1).
Multistep synthesis of a selectively functionalized D (+)-mannose derivative with the aim to construct
a TB-mannose cluster conjugate which is supposed to be water soluble and would interact with
various biological entities like protein, amino acid etc.

Accomplishments: αβ-CD heteroduplex was synthesized by oxidative coupling between α-CD dithiol
(6-step synthesis) and β-CD dithiol (6-step synthesis). Selective synthesis of the aforementioned
heteroduplex was also performed in the presence of custom made template. Method of its separation
and purification was developed using chromatography - gradient elution: LC-MS (waters), reverse
phase-preparative/analytical, flash etc.). In water, the duplex showed high binding affinities
(determined by VP-ITC technique) with several probe guests.

Dissertation title: Modified cucurbit[n]urils: synthesis and supramolecular interactions.
Accomplishments: My Ph.D. dissertation comprised of four projects: two major independent and two
collaborative projects.
Major projects: Two new water soluble functionalized cucurbit[n]urils (A and B, CB[n], n = 6) were
synthesized, purified and characterized and their supramolecular interactions with organic guests were
studied. Binding modes of the macrocycle B (hexamethylcucurbit[6]uril, MeCB[6]) with viologen
guests (MeV2+
and MeV+
) in solution and solid state were determined. Macrocycle B exhibited strong
binding affinity toward MeV2+
guest (K = 105
M-1
) in pure water. 1
H NMR and UV-vis titration
experiments showed remarkable weakening of the B-MeV2+
complex in the presence of NaCl.
Synthesis of macrocycle A was the first example of substituted CB[6] with one substituent at the
methylene bridge position. New method of synthesis, isolation and purification was developed. The
results introduced a milestone concept in the chemistry of CB[n] compound and further extend its area
of application.
Collaborative projects: In one project, new method of deprotection of a propylene glycoluril dimer
(C) was introduced. In the second project, a CB[6] based supramolecular shuttle (rotaxane) was
constructed based on the complexation studies between CB[6] and a bispyridinium ethylene guest.

Thesis title: Synthesis of Schiff bases, thiadiazoline derivatives and some heterocyclic compounds as
probes for pharmacological studies especially as Anti-cancer agents.
Accomplishments: A series of aldehyde-Δ2
-1, 3, 4-thiadiazolines (Scheme 1) were synthesized from
oxidative cyclization of the aldehyde thiosemicarbazones in acetic anhydride in hot condition. In
addition, some of the aldehyde thiosemicarbazone underwent cyclization with p-substituted
phenacylbromide to give the phenylthiazolyl hydrazone (Scheme 3). Some of the synthesized
compounds were shown to have cytotoxic and antibacterial activity.
