Centrifugation is a technique used for the separation of particles from a solution according to their size, shape, density, viscosity of the medium and rotor speed.
The particles are suspended in a liquid medium and placed in a centrifuge tube. The tube is then placed in a rotor and spun at a define speed.
Separation through sedimentation could be done naturally with the earth gravity, nevertheless, it would take ages. Centrifugation is making that natural process much faster.
Rotation of the rotor about a central axis generates a centrifugal force upon the particles in the suspension
PROCESS OF CENTRIFUGATION
The centrifuge consists of essentially a container in which a mixture of solid and liquid or two liquids is placed and rotated at high speeds.
The mixture is separated into its constituent parts by the action of centrifugal force on their densities. A solid or liquid of higher specific gravity is thrown outward with greater force. Therefore, it is retained at liquid. Iayer of pure liquid.

INTRODUCTION
1. Centrifugation is widely used in bulk drug industry to separate crystalline material from
suspension.
2. Centrifugation is used for isolation of bacterial cells. Actinomycetes mycelium and smes from
liquid grow 1h.
3. Centrifugation is used for separation of blood cells from blood.
4. Ultracentrifuge is used for separation of virus particle which arc utilize in industrial application .
5.Centrifugation is used for separating drug present in blood. urine and tissue fluid. This is essential
for evaluation of pharmacokinetic parameters and bioequivalence studies.
6. Centrifugation is used to obtain pure form of insulin hy selective precipitating other fraction of
protein and separating them by ultraccntrilugation.

PRINCIPLE OF CENTRIFUGATION
Thc centrifuge involves the principle of
sedimentation. where the acceleration at centripetal
force causes
denser substances to separate along the radial
direction at the bottom of the tube and the lighter
object will tend move up thc tube. The particle having
size more than 5 um are separated by simple filtration
process
while the partirte having size 5 um or less do not
sediment under gravity. Thc centrifugal force is used
to separate them.
