For bacterial populations cultured in vitro ii is important to study their homogeneity of growth, where the majority of cells display identical growth rate, cell size and content. Recent insights, however, have revealed that even cells growing in exponential growth phase can be heterogeneous with respect to variables typically used to measure cell growth. Bacterial heterogeneity has important implications for how bacteria respond to environmental stresses, such as antibiotics. The phenomenon of antimicrobial persistence, for example, has been linked to a small subpopulation of cells that have entered into a state of dormancy where antibiotics are no longer effective. While methods have been developed for identifying individual non-growing cells in bacterial cultures, there has been less attention paid to how these cells may influence growth in colonies on a solid surface. In response, we have developed a low-cost, open-source platform to perform automated image capture and image analysis of bacterial colony growth on multiple nutrient agar plates simultaneously. The descriptions of the hardware and software are included, along with details about the temperature-controlled growth chamber, high-resolution scanner, and graphical interface to extract and plot the colony lag time and growth kinetics. Experiments were conducted using a wild type strain of Escherichia coli K12 to demonstrate the feasibility and operation of our setup. By automated tracking of bacterial growth kinetics in colonies, the system holds the potential to reveal new insights into understanding the impact of microbial heterogeneity on antibiotic resistance and persistence.

On the other hand, electron microscopy is useful for not only assisting virologists in identifying disease-causing viral agents, but may perhaps provide definitive identification of these agents. Hazelton and Gelderblom (2003)1 have made the argument that electron microscopy should be the diagnostic tool of choice in many viral outbreaks because of the rapidity and fidelity of the result.

1. Yes, i do agree with Hazelton and gelderblom that the electron microscopy should be the diagnostic tool of choice. I agree with this because after reading some articles i have found that the electron microscopy is fast and realiable. When you are trying to identify a disease or viral outbreak, you are going to need something that will give you fast results that you can trust. I also think when using the electron microscopy that you should use another tool to back your findings.
2. I agree with the statement, electron microscopy has two advantages over enzyme-linked immunosorbent assay and nucleic acid amplification tests. After a simple and fast negative stain preparation, the undirected, “open view” of electron microscopy allows rapid morphologic identification and differential diagnosis of different agents contained in the specimen. Details for efficient sample collection, preparation, and particle enrichment are given. Applications of diagnostic electron microscopy in clinically or epidemiologically critical situations as well as in bioterrorist events are discussed. Electron microscopy can be applied to many body samples and can also hasten routine cell culture diagnosis. To exploit the potential of diagnostic electron microscopy fully, it should be quality controlled, applied as a frontline method, and be coordinated and run in parallel with other diagnostic techniques. This just show that Gram staining is the first step identify a bacteria, when electron microscopy will make a more result to understand where and how the bacteria was produce. I feel that electron microscopy is just a more advance way to diagnosis the reasoning on how a bacteria was caused.

Introduction: Bacterial transformation is the process by which a DNA fragment is inserted into a bacterium. This process explains the genetic variability of bacteria and the resistance that some of these bacteria develop to antibiotics. Bacterial transformation has a wide range of applications, being a widely used technique in the laboratory. ACTIVITY 1 a) The videotape “New drugs against new threats” and the virtual experiment “Find a target for a drug” explain the process drawn below. In groups, find information on other different ways to introduce a gene into a cell and prepare an explanation with drawings to share with your classmates.
