Background: The current study aimed to elucidate the protective role of combined fenugreek and a-tocopherol against cadmium induced histopathological
changes in the testes.
Materials and methods: Thirty adult male albino rats divided into three equal
groups 10 rats each. Group I is the control group. Group II received 5 mg/kg/day
cadmium chloride. Group III received 5 mg/kg/day cadmium chloride and 150
mg/kg/day fenugreek and 100 mg/kg/day of a-tocopherol. The treatment of all
groups was done by oral gavage for 60 consecutive days. The testes were removed
and subjected to histopathological and ultrastructure study.
Results: Rats exposed to cadmium showed severe histopathological changes in
the testicular spermatogenic series, many vacuoles and multinucleated giant cells.
Treatment with fenugreek and a-tocopherol partially improved the morphological
changes, reduced tissue damage and rebuilt of the spermatogonia layer.
Conclusions: Fenugreek and a-tocopherol might represent a promising medicinal
combination to ameliorate the toxic effects of cadmium exposure. (Folia Morphol

Chlorophytum Ker Gawl, is a medicinally important plant genus employed since ancient time as a key component in Ayurvedic and Unani medicine. Genus represented with more than 217 species out of which 17 species have been reported from India. The main objective of this study is to evaluate molecular phylogeny of Chlorophytum species. In this study phylogenetic analysis of Chlorophytum species was carried out using AFLP marker. Total 16 selective primer combinations were scored as presence and absence of alleles for all the 17 species, resulting in total 938 allele, out of which 291 allele were found to be polymorphic. The percentage of polymorphism ranged from 18.3 in the combination E2M3 to 42 in the combination E1M1 .The phylogenetic tree is divided into two clade, each clade contains species with similar morphological characters. The expression of alpha-smooth muscle actin (α-SMA) was increased and proliferating cell nuclear antigen (PCNA) expression was decreased. Administration of CoQ10 and CS resulted in a significant improvement of hepatic and renal functional parameters, and an improvement of both α-SMA and PCNA.
Conclusion: It is concluded that pretreatment with CoQ10 and CS is associated with up-regulation of favorable protective enzymes and down-regulation of oxidative stress. That can be advised as a supplement to dox-treated patients.

Genetic Variability for Antioxidant Activity and Total Phenolic Content in Fo...CrimsonpublishersNTNF
Total phenolic content and antioxidant activity were evaluated in 139 diverse genotypes of four pulse crops including 54 genotypes of chickpea (Cicer arietinum), 37 lentil (Lens culinaris), 21 pigeonpea (Cajanus cajan), and 26 blackgram (Vigna mungo). Results indicate significant genotypic variation (p < 0.01) for total phenolic content (TPC) as well as antioxidant activity (AOA). Amongst the four major pulse crops tested, maximum mean phenolic content was recorded in blackgram genotypes (7.01mg GAE/g), followed by lentil (3.46mg GAE/g), pigeonpea (3.32mg GAE/g) and chickpea (2.30mg GAE/g). In general, the Mediterranean landraces of lentil had higher phenolic content as compared to the other lentil varieties and breeding lines. Amongst the chickpea genotypes the phenolic content ranged from 0.40 to 5.63mg GAE/g and comparatively higher value for phenolic content was recorded in desi types (2.67mg GAE/g) as compared to the Kabuli types (1.05mg GAE/g). The antioxidant activity (AOA) was assayed in mature dry seeds utilizing DPPH (2,2-Diphenyl-1-picryl hydrazyl) radical scavenging assay which ranged from 1.73 to 19.14μmole Trolox/g tissue. As observed for TPC, highest AOA was also recorded in blackgram genotypes (19.14μmole Trolox/g tissue), followed by pigeonpea (2.72μmole Trolox/g tissue), chickpea (2.05μmole Trolox/g tissue) and lentil (1.73μmole Trolox/g tissue). Highly significant genotypic as well as phenotypic correlation (p<0.01) was recorded between phenolic content and antioxidant activity in chickpea, lentil as well as blackgram (rG values ranging from 0.268 to 0.850 and rP from 0.253 to 0.817), however, surprisingly the values were non-significant in case of pigeonpea. Strongest genotypic correlation was recorded in chickpea (rG=0.850), followed by lentil (rG =0.744), and blackgram (rG =0.268). High broad-sense heritability (h2bs) (0.89 to 0.97) for phenol content was recorded which indicates that substantial portion of total variation for phenolic content is due to genetic effects.

Cancer is a second leading cause of death after heart attack, in developing as well as undeveloped
countries. It is caused by unregulated growth and metastasis of the abnormal cancer cells.
Cancer can be cured by radiation, immunotherapy and chemotherapy, among them; chemotherapy is a
good treatment for cancer, in which chemotherapeutic drug is used. The anticancer activity of newly
synthesized compounds (13a-j) was carried out on four different types of human cancer cell lines like
MCF-7 (breast), A549 (lung), Colo-205 (colon) and A2780 (ovarian) by the MTT method, and compared
to etoposide used as a positive control. Among them, compound 13g with electron-withdrawing
(3,5-dinitro) group, exhibited more promising activity in all cell lines (MCF-7 = 0.10±0.076 μM, A549
= 0.17±0.039 μM, Colo-205= 0.13±0.022 μM and A2780 = 0.87±0.027μM). This compound may act
as lead drug in cancer chemotherapy. In future, this compound can be examined for clinical studies.
Abstract
Objective(s):
Gold nanoparticles (GNPs) command a great deal of attention for biomedical applications nowadays. The data about the degree of toxicity and the accumulation of gold nanoparticles in-vivo is not enough to judge.
Materials and Methods:
A total of 32 healthy male Wistar rats were randomly divided into 4 including: three GNP-treated and one control group. Groups 1, 2 and 3 received 0.5 cc of a solution containing 5, 10, and 100 ppm Au daily via intraperitoneal (IP) injection for 7 days, respectively. The control group was treated with 0.5 cc normal saline with same procedure. Then, several biochemical parameters such as serum glutamate oxaloacetat transaminase (SGOT) and serum glutamate pyrvate transaminase (SGPT) were evaluated at 2, 7 and 14 days after the last injection. After 14 days, all the rats were sacrificed and liver, lung tissues were separated and evaluated.
Results:
SGOT two days after intervention was significantly greater in the group 2 than the control group. In liver histological assessment, in group 1, basophils were observed around the central veins, in group 2 fading and no observation of central veins was seen, and in group 3 hepatic damage was noticed. The lung histological results showed severe vascular hyperemia in group 1, air sacs damage in group 2, and complete air sacs destruction in group 3.
Conclusion:
The results showed extreme changes in the histopathology of lung and liver tissues caused by spherical nanogold with 5-10 nm size in all of three treatment groups.

new Schiff base [I] was prepared by refluxing Amoxicillin trihydrate and 4-Hydroxy-
3,5-dimethoxybenzaldehyde in aqueous methanol solution using glacial acetic acid as a catalyst. The
new 1,3-oxazepine derivative [II] was obtained by Diels- Alder reaction of Schiff base [I] with
phthalic anhydride in dry benzene. The reaction of Schiff base [I] with thioglycolic acid in dry
benzene led to the formation of thiazolidin-4-one derivative [III]. While the imidazolidin-4-one [IV]
derivative was produced by reacting the mentioned Schiff base [I] with glycine and triethylamine in
ethanol for 9 hrs. Tetrazole derivative [V] was synthesized by refluxing Schiff base [I] with sodium
azide in dimethylformamid DMF. The structure of synthesized compounds[I-V] was characterized
by their melting points, elemental analysis CHN-S and by their spectral data; FTIR and 1H NMR

Objective: To investigate the changes in the retina due to deltamethrin toxicity and the process in cell inflammation and apoptosis.
Study Design: Sixteen Wistar albino rats were randomly divided into two groups as control (n=8) and deltamethrin (n=8) groups. Saline was given to the control group, and 0.5 mL of 5 mg/kg deltamethrin was given to the deltamethrin group for 14 days each. Blood was collected for biochemical analysis. Retinal tissue was processed for histological examination.
Results: Compared to the control group, MDA levels were high while GSH and CAT levels were low in the deltamethrin group. Histopathological analysis showed spaces between the pigment epithelium, irregularity in the delimiting membrane, degenerated ganglion, cone and bacillus cell, pyknotic nuclei, thinned inner limitation membrane, and thickened vascular wall. The control group showed FAS expression in the pigment layer limiting membranes, in the nuclei of many cone and bacillus cells, and ganglion cells in the control group sections. In the deltamethrin group, FAS expression was observed in the inner and outer limiting membranes of the pigment epithelium, cone and bacillus cells, and ganglion cell nuclei. In the control group, negative NOS expression in the pigment epithelium and outer limiting membranes, internal limitation membrane, and ganglion cells in the cone and bacillus cell nuclei were observed. In the deltamethrin group, NOS expression was positive in the pigment epithelium, cone and bacillus, and ganglion cell nuclei.
Conclusion: We suggest that deltamethrin toxicity induced apoptotic process due to increased inflammation in the retina and may cause visual impairment as a result of neural damage.

Naturally Occurring Insecticides from the Marine Sponge Xestospongia testudin...Premier Publishers
The insecticidal potential of the marine sponge Xestospongia testudinaria extracts against the whitefly Bemicia tabaci and the aphid Aphis gossypii under laboratory conditions were examined after 24 and 72 hrs. of treatments. The bioassay-guided isolation afforded nine structurally diverse marine compounds: xestosterol (1), 18-bromooctadeca-(9E,17E)-dien-7,15-diynoic acid (2), 18,18-dibromo-(9E)-octadeca-9,17-dien-5,7-diynoic acid (3), 16-bromo(7E,11E,l5E)hexadeca-7,11,l5-trien-5,13-diynoic acid (4), methyl 18-bromooctadeca-(9E,17E)-dien-7,15-diynoate (5), 2-methylmaleimide-5-oxime (6), maleimide-5-oxime (7), 2'-deoxythymidine (8) and 2'-deoxyuridine (9). The chemical structures were determined on the basis of detailed spectroscopic analyses (ESI-MS and 1H, 13C-NMR). Maleimide-5-oxime (7) was the most potent one against 3rd instar nymphs of B. tabaci under laboratory conditions after 24 and 72 hrs of treatments, with LC50 values of 175.26 and 7.62 ppm, respectively. While against Aphis gossypii, 18-bromooctadeca-(9E,17E)-dien-7,15-diynoic acid (2) recorded the highest LC50 values of 1.54 and 0.001 ppm after 24 and 72 hrs of treatments against Aphis gossypii, respectively. Structure-activity relationship revealed the effectiveness of maleimides and brominated polyacetylenic lipids.
Insecticidal and Antifeedant Effects of Neem Seed and Scent Leaves on Dermest...Premier Publishers
This study was conducted to evaluate the insecticidal and antifeedant effect on the hide beetle (Dermestes marculatus) exposed to dried croaker (Pseudotolithus elongatus) flesh treated with 30%, 25% and 15% concentrations of scent leaves (Ocimum gratissimum) and neem seed (Azadirachta indica) extracts. Insecticidal effect was determined as daily percentage mortality of hide beetle larvae in each treatment, while weight loss of preserved fish was the measure of feeding inhibitory effect of the treatment during the 10 days exposure. With a total kill (100% mortality) of the insect larvae in 10 days by 30% neem seed concentration, neem seed proved to be more potent than scent leaf of equal concentration even though statistical analysis did not find any significant difference between the two treatments. The least weight loss of 7% obtained from 30% neem seed concentration was less than half the weight lost by fish treated with equal concentration of scent leaf, indicating the superiority of neem seed. The study has shown that high concentration of scent leafs and particularly neem seed, have strong insecticidal and antifeedant effect on hide beetle larvae and can be used to control this pest and preserve dried fish. A combination of neem seed and scent leaf together may prove more effective and needs to be investigated.
5 phytochemical analysis of bitter melon juice; antiproliferative and apopto...BIOLOGICAL FORUM
ABSTRACT: Osteosarcoma is one of the most common cancers among adolescents and young adults. Prognosis of osteosarcoma is particularly poor, suggesting critical needs for additional drugs to improve disease outcome. In this study, osteosarcoma cell SaOS-2 was used as an in vitro model to assess the effect of bitter melon (Momordica charantia) juice (BMJ) as an anticancer agent. Fruit juice of Momordica charantia was subjected to preliminary phytochemical analysis to identify various phytoconstituents present in them such as carbohydrates, starch, flavonoids, phenols, steroids, terpenoids, alkaloids, tanins and saponins. Cell viability (MTT) results revealed that 1 to 10% (v/v) of BMJ treatment significantly (p<0.05) reduced the proliferation of SaOS-2 cells in a dose dependent manner. Antiproliferative activity of BMJ was also coupled with morphological changes in the cells with the significant induction of apoptosis as quantified by DAPI stain. Reactive oxygen species (ROS) generation was also significantly induced by BMJ treatment. Furthermore, flow cytometric analysis revealed that BMJ induced cell cycle arrest in G2/M phase with the enhancement of apoptosis. The findings of this study suggest that BMJ modulates ROS generation and cell cycle arrest in G2/M phase of SaOS-2 cells which lead to inhibition of cell proliferation and induction of apoptosis of osteosarcoma. These findings suggest that BMJ could be a potential agent for osteosarcoma treatment and anticancer drug discovery.
Renal Toxicity Studies of Albino Rats Exposed to Common Emulsion Paint SolutionPremier Publishers
Emulsion paint solutions (EPS) are constantly used to decorate and protect different materials in the environment. The occupational exposure of professional painters and paint factory workers to EPS may predispose them to systemic toxicity. This work investigated the effects of EPS exposure on renal function indices in albino rats to assess potential renal toxicity. Twenty male albino rats weighing between 120-140g were collected and divided into four groups. Two groups were exposed to periodic intraperitoneal administration of 2.5mg/kg body weight (bw) of the EPS for 14days while the other two groups received the same dose of distilled water and served as control. The rats were sacrificed after 7and14days respectively, and their blood collected and analyzed for heavy metals, urea, creatinine and electrolytes, using standard methods. The rat kidneys were also excised, weighed and analyzed for total protein and histopathology. Results showed that bw, kidney weight, serum creatinine, urea and electrolytes decreased significantly(p<0.05) in the exposed groups compared to control. However, urea and Na+ increased significantly(p<0.05) after the initial 7days exposure. Kidney total protein increased by 20.83% and Pb, Ni, and Cd were detected in the exposed rats. Results of kidney histopathology showed intact renal tubules and glomeruli. These results suggest that exposure of albino rats to EPS caused significant alterations in the steady-state levels of renal function biomarkers. Therefore, prolonged exposure may exert deleterious effects on renal function.
POTENTIAL OF CURCUMIN IN PYRAMID CELL NECROSIS OF MICE (MUS MUSCULUS) DUE TO ...IAEME Publication
Mercury pollution in the aquatic environment can cause intoxication of marine
organisms and the formation of free radicals in the human body if consumed. Curcumin
is a natural ingredient that contains extracellular antioxidants to overcome the
formation of free radicals in the body. The purpose of the study was to determine the
effect of administering curcumin to the number of pyramid cells that were necrotic in
mice (Mus musculus) exposed to methylmercury. This experiment used a completely
randomized design with 4 groups of mice, i.e. 0.5 ml distilled water, 0.056 mg/kg
methylmercury, 0.0056 mg/kg methylmercury + 150 mg/kg curcumin and 0.0056 mg/kg
methylmercury + 300 mg/kg curcumin. Examination of total pyramid cell necrosis was
observed with a 400x magnification light microscope. In this study, there was a
decrease in the number of pyramid cell necrosis in mice. The decrease in the number
of necrotic cells was directly proportional to the increase in dose given. The treatment
results as positive controls showed the highest value with the number of pyramid cells
experiencing necrosis of 23.28. The highest decreases in the number of each treatment
were 10.32 in 150 mg/kgBW curcumin and 5.80 in 300 mg/kgBW curcumin. Curcumin
can reduce the number of pyramid cells that experience necrosis due to exposure to
methylmercury.

Abstract:
An inoculation of virulent Leishmania major is known as leishmanization (LZ) which is proven to be the most effective control measure against Cutaneous Leishmaniasis (CL). However, using LZ is restricted due to various side effects such as uncontrolled lesion development. In the present research, the efficacy of cationic nanoliposomes containing CpG oligodeoxynucleotides (CpG ODN) as an improved adjuvant delivery system was studied to diminish the lesion development and infection course of L. major after inoculation into the mice. BALB/c mice were inoculated subcutaneously (SC) with L. major plus empty DSPC, DSPC (CpG ODN), DSPC (Non CpG ODN), empty DMPC, DMPC (CpG ODN), DMPC (Non CpG ODN) or HEPES buffer. The results showed that group of mice received DMPC (CpG ODN) nanoliposomes developed a significantly smaller lesion and showed minimum number of L. major in the spleen and draining lymph nodes. In addition, using DMPC (CpG ODN) liposomes resulted in a Th1 type of immune response with a preponderance of IgG2a isotype which is concurrent with the production of DMPC (CpG) induced IFN-γ in the spleen of the mice. Taken together, the results suggested that immune modulation using DMPC (CpG ODN) nanoliposomes might be a practical approach to improve the safety of LZ

Genotoxicity of Goji Berry (Lyciumbarbarum) In Vivo Mammalian Cellsinventionjournals
Lyciumbarbarum (Gojji berry) belongs to family Salonaceae which is found in China and Himalayan. This herb is used to prevent various diseases and in medical treatments as an alternative medicine being widely used for its antioxidant and revitalizing potential effects. In recent years, Gojji has become increasingly popular in Europe and North America as a "superfruit" and dietary supplement. The belief that herbal products do not bring any risk to health, is part of popular culture. However the term "natural" assigned to many products cannot assure no health risk. The aim of this study was to evaluate the possible genotoxic effects of aqueous extract of Lyciumbarbarum (Gojji berry) by micronucleus test and comet assay. Thirty Rattus norvegicus were divided into three equal groups: 1) experimental group, submitted to Gojji berry (200mg/kg orally); 2) positive control group (cyclophosphamide), and; 3) negative control group (distilled water). Micronucleus Tests were done by smear method of bone marrow cells performed after 48h for acute, and 72h for chronic exposure. The comet assay was performed on peripheral blood taken from the tail of each animal 4h, and 24h after intervention. Cytotoxicity was assessed by observing the DNA damage measuring the percentage of DNA in the tail (% DNA- measurement of the proportion of the total DNA present in the tail) and the tail moment (TM-tail length times the percentage of DNA in the tail), calculated by 100 nucleoids per animal and the presence of micronuclei in 2,000 polychromatic erythrocytes per animal. Analysis of variance (ANOVA) followed by Tukey test at 5% significance was used comparing the results. The data showed no significant difference in the frequency of DNA damage and the number of micronuclei between the experimental group and the negative control group. The results also suggest that the aqueous extract of Lyciumbarbarum (Gojji berry) at the dose of 200 mg/kg showed no genotoxic effect, which could, to a certain point, justifies its use.

Cancer is a second leading cause of death after heart attack, in developing as well as undeveloped
countries. It is caused by unregulated growth and metastasis of the abnormal cancer cells.
Cancer can be cured by radiation, immunotherapy and chemotherapy, among them; chemotherapy is a
good treatment for cancer, in which chemotherapeutic drug is used. The anticancer activity of newly
synthesized compounds (13a-j) was carried out on four different types of human cancer cell lines like
MCF-7 (breast), A549 (lung), Colo-205 (colon) and A2780 (ovarian) by the MTT method, and compared
to etoposide used as a positive control. Among them, compound 13g with electron-withdrawing
(3,5-dinitro) group, exhibited more promising activity in all cell lines (MCF-7 = 0.10±0.076 μM, A549
= 0.17±0.039 μM, Colo-205= 0.13±0.022 μM and A2780 = 0.87±0.027μM). This compound may act
as lead drug in cancer chemotherapy. In future, this compound can be examined for clinical studies.
Abstract
Objective(s):
Gold nanoparticles (GNPs) command a great deal of attention for biomedical applications nowadays. The data about the degree of toxicity and the accumulation of gold nanoparticles in-vivo is not enough to judge.
Materials and Methods:
A total of 32 healthy male Wistar rats were randomly divided into 4 including: three GNP-treated and one control group. Groups 1, 2 and 3 received 0.5 cc of a solution containing 5, 10, and 100 ppm Au daily via intraperitoneal (IP) injection for 7 days, respectively. The control group was treated with 0.5 cc normal saline with same procedure. Then, several biochemical parameters such as serum glutamate oxaloacetat transaminase (SGOT) and serum glutamate pyrvate transaminase (SGPT) were evaluated at 2, 7 and 14 days after the last injection. After 14 days, all the rats were sacrificed and liver, lung tissues were separated and evaluated.
Results:
SGOT two days after intervention was significantly greater in the group 2 than the control group. In liver histological assessment, in group 1, basophils were observed around the central veins, in group 2 fading and no observation of central veins was seen, and in group 3 hepatic damage was noticed. The lung histological results showed severe vascular hyperemia in group 1, air sacs damage in group 2, and complete air sacs destruction in group 3.
Conclusion:
The results showed extreme changes in the histopathology of lung and liver tissues caused by spherical nanogold with 5-10 nm size in all of three treatment groups.

new Schiff base [I] was prepared by refluxing Amoxicillin trihydrate and 4-Hydroxy-
3,5-dimethoxybenzaldehyde in aqueous methanol solution using glacial acetic acid as a catalyst. The
new 1,3-oxazepine derivative [II] was obtained by Diels- Alder reaction of Schiff base [I] with
phthalic anhydride in dry benzene. The reaction of Schiff base [I] with thioglycolic acid in dry
benzene led to the formation of thiazolidin-4-one derivative [III]. While the imidazolidin-4-one [IV]
derivative was produced by reacting the mentioned Schiff base [I] with glycine and triethylamine in
ethanol for 9 hrs. Tetrazole derivative [V] was synthesized by refluxing Schiff base [I] with sodium
azide in dimethylformamid DMF. The structure of synthesized compounds[I-V] was characterized
by their melting points, elemental analysis CHN-S and by their spectral data; FTIR and 1H NMR
spectroscopy. Two cancer cell lines include: (RD) human pelvic rhabdomyosarcoma
and (L20B) the mice intestines carcinoma cell line
Objective: To investigate the changes in the retina due to deltamethrin toxicity and the process in cell inflammation and apoptosis.
Study Design: Sixteen Wistar albino rats were randomly divided into two groups as control (n=8) and deltamethrin (n=8) groups. Saline was given to the control group, and 0.5 mL of 5 mg/kg deltamethrin was given to the deltamethrin group for 14 days each. Blood was collected for biochemical analysis. Retinal tissue was processed for histological examination.
Results: Compared to the control group, MDA levels were high while GSH and CAT levels were low in the deltamethrin group. Histopathological analysis showed spaces between the pigment epithelium, irregularity in the delimiting membrane, degenerated ganglion, cone and bacillus cell, pyknotic nuclei, thinned inner limitation membrane, and thickened vascular wall. The control group showed FAS expression in the pigment layer limiting membranes, in the nuclei of many cone and bacillus cells, and ganglion cells in the control group sections. In the deltamethrin group, FAS expression was observed in the inner and outer limiting membranes of the pigment epithelium, cone and bacillus cells, and ganglion cell nuclei. In the control group, negative NOS expression in the pigment epithelium and outer limiting membranes, internal limitation membrane, and ganglion cells in the cone and bacillus cell nuclei were observed. In the deltamethrin group, NOS expression was positive in the pigment epithelium, cone and bacillus, and ganglion cell nuclei.
Conclusion: We suggest that deltamethrin toxicity induced apoptotic process due to increased inflammation in the retina and may cause visual impairment as a result of neural damage.

The insecticidal potential of the marine sponge Xestospongia testudinaria extracts against the whitefly Bemicia tabaci and the aphid Aphis gossypii under laboratory conditions were examined after 24 and 72 hrs. of treatments. The bioassay-guided isolation afforded nine structurally diverse marine compounds: xestosterol (1), 18-bromooctadeca-(9E,17E)-dien-7,15-diynoic acid (2), 18,18-dibromo-(9E)-octadeca-9,17-dien-5,7-diynoic acid (3), 16-bromo(7E,11E,l5E)hexadeca-7,11,l5-trien-5,13-diynoic acid (4), methyl 18-bromooctadeca-(9E,17E)-dien-7,15-diynoate (5), 2-methylmaleimide-5-oxime (6), maleimide-5-oxime (7), 2'-deoxythymidine (8) and 2'-deoxyuridine (9). The chemical structures were determined on the basis of detailed spectroscopic analyses (ESI-MS and 1H, 13C-NMR). Maleimide-5-oxime (7) was the most potent one against 3rd instar nymphs of B. tabaci under laboratory conditions after 24 and 72 hrs of treatments, with LC50 values of 175.26 and 7.62 ppm, respectively. While against Aphis gossypii, 18-bromooctadeca-(9E,17E)-dien-7,15-diynoic acid (2) recorded the highest LC50 values of 1.54 and 0.001 ppm after 24 and 72 hrs of treatments against Aphis gossypii, respectively. Structure-activity relationship revealed the effectiveness of maleimides and brominated polyacetylenic lipids.
Insecticidal and Antifeedant Effects of Neem Seed and Scent Leaves on Dermest...Premier Publishers
This study was conducted to evaluate the insecticidal and antifeedant effect on the hide beetle (Dermestes marculatus) exposed to dried croaker (Pseudotolithus elongatus) flesh treated with 30%, 25% and 15% concentrations of scent leaves (Ocimum gratissimum) and neem seed (Azadirachta indica) extracts. Insecticidal effect was determined as daily percentage mortality of hide beetle larvae in each treatment, while weight loss of preserved fish was the measure of feeding inhibitory effect of the treatment during the 10 days exposure. With a total kill (100% mortality) of the insect larvae in 10 days by 30% neem seed concentration, neem seed proved to be more potent than scent leaf of equal concentration even though statistical analysis did not find any significant difference between the two treatments. The least weight loss of 7% obtained from 30% neem seed concentration was less than half the weight lost by fish treated with equal concentration of scent leaf, indicating the superiority of neem seed. The study has shown that high concentration of scent leafs and particularly neem seed, have strong insecticidal and antifeedant effect on hide beetle larvae and can be used to control this pest and preserve dried fish. A combination of neem seed and scent leaf together may prove more effective and needs to be investigated.

ABSTRACT: Osteosarcoma is one of the most common cancers among adolescents and young adults. Prognosis of osteosarcoma is particularly poor, suggesting critical needs for additional drugs to improve disease outcome. In this study, osteosarcoma cell SaOS-2 was used as an in vitro model to assess the effect of bitter melon (Momordica charantia) juice (BMJ) as an anticancer agent. Fruit juice of Momordica charantia was subjected to preliminary phytochemical analysis to identify various phytoconstituents present in them such as carbohydrates, starch, flavonoids, phenols, steroids, terpenoids, alkaloids, tanins and saponins. Cell viability (MTT) results revealed that 1 to 10% (v/v) of BMJ treatment significantly (p<0.05) reduced the proliferation of SaOS-2 cells in a dose dependent manner. Antiproliferative activity of BMJ was also coupled with morphological changes in the cells with the significant induction of apoptosis as quantified by DAPI stain. Reactive oxygen species (ROS) generation was also significantly induced by BMJ treatment. Furthermore, flow cytometric analysis revealed that BMJ induced cell cycle arrest in G2/M phase with the enhancement of apoptosis. The findings of this study suggest that BMJ modulates ROS generation and cell cycle arrest in G2/M phase of SaOS-2 cells which lead to inhibition of cell proliferation and induction of apoptosis of osteosarcoma. These findings suggest that BMJ could be a potential agent for osteosarcoma treatment and anticancer drug discovery.
Renal Toxicity Studies of Albino Rats Exposed to Common Emulsion Paint SolutionPremier Publishers
Emulsion paint solutions (EPS) are constantly used to decorate and protect different materials in the environment. The occupational exposure of professional painters and paint factory workers to EPS may predispose them to systemic toxicity. This work investigated the effects of EPS exposure on renal function indices in albino rats to assess potential renal toxicity. Twenty male albino rats weighing between 120-140g were collected and divided into four groups. Two groups were exposed to periodic intraperitoneal administration of 2.5mg/kg body weight (bw) of the EPS for 14days while the other two groups received the same dose of distilled water and served as control. The rats were sacrificed after 7and14days respectively, and their blood collected and analyzed for heavy metals, urea, creatinine and electrolytes, using standard methods. The rat kidneys were also excised, weighed and analyzed for total protein and histopathology. Results showed that bw, kidney weight, serum creatinine, urea and electrolytes decreased significantly(p<0.05) in the exposed groups compared to control. However, urea and Na+ increased significantly(p<0.05) after the initial 7days exposure. Kidney total protein increased by 20.83% and Pb, Ni, and Cd were detected in the exposed rats. Results of kidney histopathology showed intact renal tubules and glomeruli. These results suggest that exposure of albino rats to EPS caused significant alterations in the steady-state levels of renal function biomarkers. Therefore, prolonged exposure may exert deleterious effects on renal function.

Cytotoxicity was assessed by observing the DNA damage measuring the percentage of DNA in the tail (% DNA- measurement of the proportion of the total DNA present in the tail) and the tail moment (TM-tail length times the percentage of DNA in the tail), calculated by 100 nucleoids per animal and the presence of micronuclei in 2,000 polychromatic erythrocytes per animal. Analysis of variance (ANOVA) followed by Tukey test at 5% significance was used comparing the results. The data showed no significant difference in the frequency of DNA damage and the number of micronuclei between the experimental group and the negative control group. The results also suggest that the aqueous extract of Lyciumbarbarum (Gojji berry) at the dose of 200 mg/kg showed no genotoxic effect, which could, to a certain point, justifies its use.
ABSTRACT- Mosquito-borne diseases have intruded the globe since immemorial time. The present scenario for
commanding the mosquitoes is aimed at application of target and stage-specific, cost-effective and biodegradable
phytoproducts. Plant extracts are safer for non-target organisms including man. Plant based formulations would be more
feasible environmental products with proven potential as insecticide. Therefore, in the present study of larvicidal
activity of biologically active compound Apigenin extracted from leaf of Jatropha gossypifolia against the filarial
vector, Culex quinquefasciatus was studied. Standard WHO protocols with minor modifications was adopted for the
larvicidal bioassay. The active compound Apigenin extracted through ethyl alcohol solvent from the leaf of Jatropha
gossypifolia plant of family Euphorbiaceae was administered for 24h or 96h to the larvae of Culex quinquefasciatus.
Exposure of larvae over 24h to sub-lethal doses (40% and 80% of LC50) of apigenin, significantly (P<0.05) altered the
level of total protein, total free amino acid, glycogen and activity of enzymes acetyl cholinesterase, acid and alkaline
phosphatase activity in whole body tissue of Culex quinquefasciatus larvae. The alterations in all these biochemical
parameters were significantly (P<0.05) time and dose dependent.

Bryophyllum Pinnatum: A Potential Attenuator of Cadmium-Induced Oxidative Str...IOSR Journals
Cadmium has been famously implicated in the stimulation of free radical production in biosystems resulting in oxidative deterioration of lipids, proteins and DNA, and initiating various pathological conditions in humans and animals. This study therefore, examined the antidotal and ameliorative capacity of crude ethanolic extract of Bryophyllum pinnatum on cadmium-induced oxidative stress using rabbit models. A total of fifteen rabbits (1.30±0.05kg) were used for the study. After two weeks of acclimatization, the rabbits were randomly rifted into three experimental groups- (N, CD & CB) with five animals per group. The control group (N) was injected normal saline intraperitoneally (3mg/kg body weight) and the test groups (CD & CB) were administered cadmium once daily by subcutaneous injection (3mg/kg body weight). The ethanolic extract of the plant was orally administered once daily at a dose of 100mg/kg body weight. The oxidative and antioxidative stress parameters were assessed in tissues. The results showed significant difference (p˂ 0.05)in treated groups relative to the control group with the exception of glutathione peroxidase activity in leg muscles. Therefore, the results obtained in this study confirmed the potency of the plant to annihilate cadmium toxicity in animals

Cladosporium cladosporioides is one of the promising entomopathogenic fungi acting as insect-pathogenic microorganism or can be used as a source of toxins against insect pests. Ethyl acetate extract of the secondary metabolites of C. cladosporioide was obtained, and its volatile constituents were characterized using GC/MS technique. Also, two major compounds were isolated and identified as 3-phenyl propanoic acid (6) and 3-(4β-hydroxy-6-pyranonyl)-5-isopropylpyrrolidin-2-one (7). It's worthy to mention that this isolated compound (7), is reported from C. cladosporioides for the first time. Also, the toxicity of the ethyl acetate extract of the secondary metabolites of C. cladosporioides against both adults and nymphs of cotton aphid, A. gossypii was determined. Data showed that C. cladosporioides ethyl acetate extract was most effective against nymphs showing LC50 of 24.5827 ppm, LC90 of 128.7385 ppm and toxicity index of 100%, while, it showed LC50 of 36.6959 ppm, LC90 of 154.4394 ppm and toxicity index of 76.69% against adults.
Atmospheric Exposure to Cr III Powder Causes Genotoxicity in Rattus Norvegicus.inventionjournals
Several chemical elements are responsible for altering the genetic integrity of living beings. The metal Cr stands out in this regard. It exists in two oxidation states, Cr VI and Cr III, and has been investigated as an important environmental and occupational contaminant. Although the former is considered carcinogenic, the latter is classified as safe, even for human use in food supplementation. However, most studies with Cr( III) have been carried out by different routes to how it is occupationally found – in the atmosphere. This study evaluated the genotoxicity of Cr(III) inhaled during 8 hours of exposure to the maximum concentration permitted by ATSDR. Fifteen male Rattus norvegicus were used in this study. There were 3 groups (n=5 per group); these were - group exposed to Cr (III) powder (S), the negative control group (NC) and the positive control group (PC). The animals were exposed to Cr aerosol particles at a flow rate of 9L/min and atmospheric concentration of 500μg/m3 for only 8 hours in this study. An increase in genotoxicity and mutagenicity in the group exposed to the metal powder was observed. These findings suggest that further studies should be carried out in order to establish safe levels of exposure to Cr III in work environments
ABSTRACT- Radioprotective mechanisms of Rutin (RUT) and Quercetin (QRT) against gamma radiation was studied
by investigating recovery of histopathology of intestinal mucosa and bone marrow in Swiss albino mice. These mice
were treated with RUT (10mg/kg.b.wt.) and QRT (20mg/kg.b.wt.) once daily for five consecutive days and exposed to
7.5 Gy of gamma radiation after the last administration. RUT and QRT treatment before exposure to 7.5 Gy of gamma
radiation. To assess the intestinal and bone marrow protective potential of RUT and QRT, histological analysis was
carried out by observing the villus height, crypt survival, number of goblet cells/villus section and dead cells/villus
section in the mouse jejunum and bone marrow cellularity at 24 hours post-irradiation. Mice exposed gamma radiation
caused a significant decline in the villus height and crypt number with an increase in goblet and dead cell number with a
significant decrease in bone marrow nucleated cells. The potent antioxidant nature of RUT and QRT mitigate the
oxidative stress induced by gamma radiation and thus protect the mice from gastrointestinal damage.

Hepatoprotective Effect of Cestrum parqui L. aerial parts and Phytochemical ...Jing Zang
This study deals with the investigation of hepatoprotective effect of 70% methanolic extract from Cestrum parqui aerial parts and determination of the bioactive components of the plant. The hepatoprotective effect of Cestrum parqui methanol extract (100, 500, 1000 mg/kg) was analysed on carbon tetrachloride (CCl4)-induced acute liver injury. The administration of a single dose of 40% CCl4 (1ml/kg b.w.) causes an increase in the activities of serum alanine aminotransferase (ALT) and aspirate aminotransferase (AST) enzymes and so pretreated orally of a dose from Cestrum parqui methanol extract (100, 500, 1000 mg/kg) and silymarin (200 mg/kg) for three consecutive days prior to The administration of a single dose of CCl4 significantly prevented the increase in the activities of these enzymes. Histological analysis showed that Cestrum parqui methanol extract at doses of 500 and 1000 mg/kg and silymarin reduced the incidence of liver lesions including vacuole formation, neutrophil infiltration and necrosis of hepatocytes induced by CCl4. The extract cause a negative result on the antioxidative enzymes, superoxide dismutase (SOD), glutathione peroxidase (GPx), glutathione reductase (GRd) and decreased malondialdehyde (MDA) level in liver, as compared to those in the CCl4-treated group and this suggests that the hepatoprotective activity of the extract is due to the antioxidant effect of the extract. Phytochemical analysis of the methanol extract from Cestrum parqui aerial parts showed that it contained different phytoconstituents, flavonoids, tannins, saponins, alkaloids, terpenes and carbohydrates.

BACKGROUND:
The current article aims to explore the protective potentials of α-tocopherol alone and the combination of allicin and vitamin B-complex against lead-acetate neurotoxicity on the cerebellar cortex.
MATERIALS AND METHODS:
Forty rats were divided into four groups (n=10). Group 1 was the control group. Group 2 received 10 mg/kg body weight (BW) of lead acetate. Group 3 was exposed to 10 mg/kg BW of lead acetate plus a combination of allicin (100 mg/kg BW) and vit. B-complex (40 mg/kg BW). Group 4 was administered lead acetate (10 mg/kg BW) and α-tocopherol (100 mg/kg BW). The animals received treatment for sixty days by oral gavage. All the groups were studied ultrastructurally and immunohistochemically with glial fibrillary acidic protein (GFAP).
RESULTS:
The affected groups revealed shrunken and degenerated Purkinje cells with irregular nuclei. The cytoplasm comprised several lysosomes, unhealthy mitochondria, and dilated Golgi saccules. The myelinated nerve fibers demonstrated breaking of the myelin sheaths, apparent vacuoles, and broad axonal spaces. Immunohistochemically, there was a tremendous surge in GFAP-positive astrocytes in the lead acetate-treated group. These histological and ultrastructural variations were ameliorated by the administration of α-tocopherol and the combination of allicin and vit. B complex. Moreover, an apparent decrease in the number of GFAP-positive astrocytes was obvious in the protected groups.
CONCLUSIONS:
Although both α-tocopherol and the combination of allicin and vit. B-complex can be used as possible adjuvant therapies to ameliorate nervous system ailments attributable to lead acetate, α-tocopherol showed more protective potential.

Background
Quercetin, a naturally occurring flavonoid known for its potent antioxidant properties, has been investigated for its potential in counteracting the harmful effects of lead (Pb) toxicity, which induces apoptosis and oxidative damage in various human tissues. This study aims to assess the reparative effects of quercetin on lead-induced testicular damage.
Methods
Four groups, each comprising ten adult male albino rats, were randomly assigned as follows: Quercetin group, Pb group, Pb + Quercetin group, and control group. All treatments were administered orally via gavage daily for a duration of 30 days. Evaluation of sex hormone levels (serum testosterone, FSH, and LH), cytokines and inflammatory mediators (IL-1β, TNF-α, MCP-1), lead concentration, oxidative and antioxidant stress markers (superoxide anion [O2−], MDA, SOD, CAT, GSH), and sperm characteristics were carried out.
Results
The results demonstrated a significant decline in sex hormones and antioxidants, accompanied by an increase in lead concentrations, cytokines, inflammatory mediators, and oxidative stress indicators (O2−, MDA), while SOD, CAT, and GSH levels were reduced. The Pb-intoxicated group exhibited a substantial increase in dead and abnormal sperm, along with significant reductions in sperm concentration and motility. Morphometrically, a marked decrease was observed in spermatogonia, primary spermatocytes, spermatids, and sertoli cells per seminiferous tubule, as well as epithelial height. Furthermore, coadministration of quercetin exhibited notable benefits. It significantly elevated testosterone levels (P < 0.001), testicular SOD, CAT, and GSH activities, while decreasing MDA levels (P < 0.001). Quercetin also mitigated the deleterious effects of lead toxicity on sperm parameters and restored morphometric variations, including epithelial height.
Conclusions
Quercetin supplementation alongside lead exposure showed a potential for ameliorating degenerative changes caused by lead toxicity in the testicles. This cotreatment effectively reduced oxidative stress, cytokine levels, inflammatory mediators, and restored biochemical alterations, thereby improving morphometric parameters.

Background:
The aim of the present work was to provide evidence about the anatomical variations as regard the origin, distribution, and branching pattern of the musculocutaneous nerve (MCN).
Materials and methods:
Brachial plexus was dissected in 40 upper limbs of 20 male adult cadavers. The pattern of the musculocutaneous nerve was photographed by a digital camera.
Results:
The location and length of the nerve branches between left and right arms were recorded and statistically analyzed. In (90%) of specimens the MCN originates from the lateral cord of the brachial plexus, in (5%) it arose from the median nerve (MN), while in the remaining (5%) specimen, it was absent. The musculocutaneous nerve pierced the coracobrachialis muscle in 90% of specimens, and in the remaining (10%) did not pierce it. The motor branches to biceps brachii muscle were categorized into: Type 1 (90%): one branch that divides to supply the two heads of biceps; Type 2 (5%): double branches, innervating each head of biceps separately. The motor branches to brachialis muscle were categorized into: Type 1 (82.9%): one branch; Type 2 (14.2%): double branches and Type 3 (2.9%): three branches that innervating brachialis muscle. Communications between the MCN and the MN were observed in 35% of specimens.
Conclusions:
The knowledge of the common and uncommon musculocutaneous nerve variations is important especially to the surgeons for carrying out surgical procedures in axilla and arm.

Objective: Natural compounds can act as metal chelators and oxygen free radical scavengers, which allows them to be used as bioactive antagonists to heavy metals neurotoxicity. The aim of the study to analyze the morphometric effects of Coriandrum sativum (C. sativum) on lead-induced neurotoxicity.
Materials and Methods: Forty Sprague-Dawley albino rats were divided into four equal groups (ten in each group): control group; coriander group: received aqueous C. sativum extracts (600 mg/kg BW for 60 days orally); lead (Pb) group: received a daily dose of lead acetate (Pb) (10 mg/kg BW for 60 days orally); Pb+ coriandrum group: received: aqueous C. sativum extract (600 mg/kg BW) prior to 10 mg/kg BW of Pb. The following parameters malondialdehyde (MDA), superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) were measured. Layers thickness and nuclei density were analyzed.
Results: Lead levels in blood and tissues were decreased significantly in the Pb group and those findings were corrected significantly (p=0.001) with C. sativum addition. Data exhibited an increase in oxidative stress marker MDA and a decrease in antioxidant enzymes activities (SOD, CAT, and GPx) significantly in the Pb group and those effects were reversed significantly (p=0.001) by C. sativum administration. The cerebellar cortex and all layers of the somatosensory cortex thickness and nuclei density were diminished significantly in the Pb group. The morphometrical measurements were corrected significantly (p=0.001) by C. sativum.
Conclusion: From the findings of the current study, Pb caused noticeable structural and functional variations in the cerebellar cortex and somatosensory cortex. C. sativum corrected these parameters as it possesses chelating and antioxidant potentials.
Background:
The anterolateral ligament (ALL) is a true well-defined ligament in the knee first described in 1879 by Segond. After the work of Claes et al., several studies were conducted about biomechanics and its role in stability of the knee. The anatomical existence of the ALL has been studied by and various radiographic diagnostic modalities and in cadavers. It originates from lateral femoral epicondyle and is inserted between Gerdy’s tubercle and the fibular head. There has been controversy about the existence of ALL in pediatric patients. The aim of this work was to confirm the presence of ALL in pediatric patients by using MRI.
Materials and Methods:
We reviewed the knee MRI scans of 100 pediatric patients (ages between one and 12 yr) who had no knee injury or congenital deformity and had been evaluated by an expert radiologist.
Results:
The ALL was detected in 90% of the pediatric patients with the use of MRI.
Conclusions:
The main finding of this study was that ALL can be seen in pediatric patients using MRI. Despite numerous studies, additional research is needed to further define the role of the ALL in knee function.

Background: In spite of its industrial usefulness and varied daily uses, lead (Pb) pollution is a widespread ecological problem that faces the humans in the 21th century. Pb was found to produces a wide range of toxic effects including neurotoxicity especially to the developing and young offspring. Recently, the utilisation of herbal plants has received a significant attention where there has been rising awareness in their therapeutic use; among these is the garlic. In light of the above, the current study is designed experimentally in female pregnant rats in order to investigate the beneficial role of garlic extract in the protection from the maternal and foetal cerebellar damage produced by administration of different doses of Pb during pregnancy.
Materials and methods: Positively pregnant female rats were divided into five groups; one control group, two Pb-treated groups (exposed to 160 and 320 mg/kg b.w. of Pb, respectively) and two groups treated with both Pb and garlic (exposed to Pb as previous groups together with 250 mg/kg b.w./day of garlic extract). Treatments started from day 1 to day 20 of pregnancy, where the mother rats of different experimental groups were sacrificed to obtain the foetuses. Pb level in the maternal and foetal blood and cerebellum was estimated by spectrophotometry. Specimens of the cerebellum of different mother and foetal groups were processed to histological and immunohistochemical staining for microscopic examination.
Results: The results showed that administration of Pb to pregnant rats resulted in a dose-dependent toxicity for both mothers and foetuses in the form of decrease in maternal weight gain, placental and foetal weights, brain weight and diminished foetal growth parameters, which were prominent in rat's group treated with larger dose of Pb. In Pb-treated rats, Pb level in blood and cerebellum was high when compared with the control group. The histopathological examination of the cerebellum of treated dams and foetuses showed marked alterations mainly in the form of Purkinje cell degeneration and lack of development of foetal cerebellum. Co-treatment of garlic extract along with Pb resulted in a significant decrease in Pb levels as compared with those treated with Pb alone with improvement of the histopathological changes.
Conclusions: This study was useful in evaluating the hazardous effects of uncontrolled use of Pb in general and in assessing the developmental and neurotoxicity of foetuses due to exposure during pregnancy in particular. Co-administration of garlic has beneficial effects in amelioration of Pb-induced neurotoxicity and reversing the histopathological changes of the cerebellum of mother rats and foetuses. Doxorubicin (DOX) is a chemotherapeutic agent used for treatment of different cancers and its clinical usage is hindered by the oxidative injury-related cardiotoxicity. This work aims to declare if the harmful effects of DOX on heart can be alleviated with the use of Coenzyme Q10 (CoQ10) or L-carnitine. The study was performed on seventy two female Wistar albino rats divided into six groups, 12 animals each: Control group; DOX group (10mg/kg); CoQ10 group (200mg/kg); L-carnitine group (100mg/kg); DOX+CoQ10 group; DOX+L-carnitine group. CoQ10 and L-carnitine treatment orally started 5days before a single dose of 10mg/kg DOX that injected intraperitoneally (IP) then the treatment continued for 10days. At the end of the study, serum biochemical parameters of cardiac damage, oxidative stress indices, and histopathological changes were investigated. CoQ10 or L-carnitine showed a noticeable effects in improving cardiac functions evidenced reducing serum enzymes as serum interleukin-1 beta (IL-1 β), tumor necrosis factor alpha (TNF-α), leptin, lactate dehydrogenase (LDH), Cardiotrophin-1, Troponin-I and Troponin-T. Also, alleviate oxidative stress, decrease of cardiac Malondialdehyde (MDA), Nitric oxide (NO) and restoring cardiac reduced glutathione levels to normal levels. Both corrected the cardiac alterations histologically and ultrastructurally. With a visible improvements in α-SMA, vimentin and eNOS immunohistochemical markers. CoQ10 or L-carnitine supplementation improves the functional and structural integrity of the myocardium.

Background:
The mortality rate of elderly patients who sustain a hip fracture is high compared to the general population. Identifying risk factors can help predict patients at risk of hip fracture to reduce the mortality rate. No studies have shown the mortality rate of patients with hip fractures in the western region of Saudi Arabia. Therefore, this study aimed to identify the risk factors associated with the mortality of patients with hip fractures admitted to the King Abdulaziz Hospital and compare the results with other studies.
Methods:
The mortality rate (within 1 yr or less) in 177 patients over the age of 60 yr who were admitted to the university hospital between July, 2007, and September, 2012, with hip fractures was retrospectively studied. The patients were assessed with regard to gender, age, type of hip fracture, and type of surgical intervention.
Results:
The overall mortality rate 1 yr after hip fracture was 12.43%, and the mean age was 77.77 yr old. The risk factors most associated with mortality were as follows: advanced age (71 to 80 and 81 to 90 yr old), male, peritrochanteric (extracapsular) fracture, and operative fixation with dynamic hip screw.
Conclusions:
The mortality rate of patients with hip fractures within 1 yr has a high-risk potential, especially for male patients over 71 yr of age with peritrochanteric (extracapsular) fractures. Surgical treatment with dynamic hip screw also was shown to be a risk factor between the different treatment options.

Systemic Lupus Erythematosus (SLE) and systemic sclerosis (SSc) are systemic inflammatory autoimmune disorders characterized by a large spectrum of clinical and laboratory features. The aim of the present study was to investigate the possible use of serum level of soluble intercellular adhesion molecule-1(sICAM-1) and soluble interleukin-2 receptor (sIL-2Ra) as biomarkers for monitoring of SLE and SSc disease activity. Moreover, it aimed to compare the specificity and sensitivity as well as cut-off value of both biomarkers in a sample of Egyptian patients. 50 SLE patients, 30 SSc patients and 60 age and sex matched healthy controls were enrolled in our study. sICAM-1and sIL-2Ra were measured in serum samples obtained from all participants. In addition to Erythosedimentation rate (ESR), complete blood count (CBC), Antineuclearantibodies (ANA) estimation, disease activity of both diseases were also assessed. sICAM-1and sIL-2Ra levels were higher in SLE and SSc patients versus control. Both parameters are correlated with each other as well as the activity parameters. A cut-off levels of 455.59 (ng/ml) &2525935 (pg/ml) in both SLE & SSs respectively was observed with the highest specificity and sensitivity. It could be concluded that sICAM-1 and sIL-2Ra are noninvasive biomarkers for SLE and SSc that could play a pathophysiologic role in development and progression of both diseases. ABSTRACT Anatomic characterization and fine structure of the human ligamentum flavum (LF), especially at different spinal levels, represent an attractive focus for the scientific and surgical application. Descrip-tive anatomical and structural study of LF at the cervical, thoracic and lumbar levels of the vertebral column in human cadavers is carried out here. The aim of the work is to clarify the anatomical features and fine structural differences in the human LF at different vertebral levels (cervical, thoracic and lumbar). Specimens of vertebral column were ob-tained from 34 human preserved cadavers. Their average age ranged between 56 and 69 years. Morphometric parameters including height, width and thickness of the ligament flavum at the mid-levels of cervical, thoracic and lumbar regions were measured. Sections obtained from different levels were stained with different stains. Morpho-metric measurements involved the relative elastic area, relative collagen area, elastic area% and collagen area% were measured.The results of the height, width and thickness of the LF at different spinal levels showed gradual increase in their mean values respectively. The LF midline gaps were found in the cervical, thoracic and lumbar regions. The morphometrical measure-ments showed that the average elastic area was highest in the cervical region and lowest in the tho-racic region. In the lumbar region, the percentages of both elastic area and the collagen area were nearly the same. The characterization of morpho-logical and histological aspects of the LF at differ-ent spinal levels will be of great importance for ap-plications in spinal surgery, biomechanical and physical rehabilitation of vertebral column.Keywords: Ligamentum Flavum – Spinal – Collagen and elastic fibers

Background:
Clinical testing has demonstrated the role of the anterolateral ligament (ALL) in controlling anterolateral laxity and knee instability at high angles of flexion. Few studies have discussed the association between an anterior cruciate ligament (ACL) injury and ALL injury, specifically after residual internal rotation and a post-ACL reconstruction positive pivot-shift that could be attributed to ALL injury. The goal of this study was to assess the correlation between ALL injury and ALL injury with concomitant ACL injury using MRI.
Material and Methods:
This was a retrospective study of 246 patients with unilateral ACL knee injuries from a database that was reexamined to identify whether ALL injuries occurred in association with ACL injuries. We excluded the postoperative reconstructed cases. The charts were reviewed on the basis of the presence or absence of diagnosed ACL injury with no regard for age or sex.
Results:
Of the 246 patients with ACL injury, there were 165 (67.1%) patients with complete tears, 55 (22.4%) with partial tears, and 26 (10.6%) with sprains. There were 176 (71.5%) patients with ALL and associated ACL injuries, whereas 70 (28.5%) did not have associated ACL injuries. There was a significant statistical relationship between ACL and ALL injuries (P<0.0001).
Conclusions:
There is high incidence of ALL tears associated with ACL injuries. Clinicians should be aware of this injury and consider the possibility of simultaneous ALL and ACL repair to prevent further knee instability.

Liver ischemia reperfusion is induced during sur-gical procedures like liver transplantation and re-section. Multiple mechanisms have been postulat-ed to liver damage following liver ischemia reperfu-sion injury, such as oxidative stress and inflamma-tory reactions. The present study declares the pos-sible mechanism of tadalafil, toward modulating the inflammatory response. Forty-eight rats were divided into 4 groups as follows; Sham group sub-jected to midline laparotomy only. Tadalafil group administered Tadalafil 10 mg/kg intraperitoneal 45 min before sham operation. I/R (Ischemia-reperfusion) group, rats undergo 60 min of hepatic ischemia followed by 60 min of reperfusion. Tada-lafil + I/R group rats undergo a similar pattern of I/R after the treatment with Tadalafil 10 mg/kg, 45 min before ischemia. At the end of the reperfusion, the blood samples were collected for estimation of biochemical markers including liver enzymes using colorimetric assay method and serum: TNF-α (tumor necrosis factor-α), IL-6 (interleukin 6) le-vels, ICAM- 1 (Intercellular Adhesion Molecule-1) were measured. Tissues were evaluated by semi-quantitative and morphometrical approaches. Ta-dalafil succeeded in restoring normal levels of liverenzymes and ameliorating the oxidative stress as evidenced by decreasing MDA and restoring redu-ced glutathione levels in liver tissue homogenate. Also, Tadalafil exhibits anti-inflammatory effects, as it significantly decreased the levels of TNF-α, IL6 and ICAM-1. The findings are supported by BCL-2, TNF-α immunomarkers. It is concluded that modulation of the inflammatory response might be one of the mechanisms of Tadalafil-mediated he-patoprotection, so it is recommended as an adju-vant therapy in liver surgery.Keywords: Ischemia/reperfusion injury – Oxidative stress – Apoptosis – TNF-α – BCL-2

Aluminum (Al) is a neurotoxic substance which has played an important role in the etiology, pathogenesis, and development of amyloid-β (Aβ) plaques. This study was carried out to evaluate the neuroprotective effect of aqueous cinnamon extract against aluminum chloride (AlCl3)-induced Alzheimer’s disease. Forty adult male albino rats, randomly divided into four equal groups. Control group; ACE200 group administered aqueous cinnamon extract (ACE) orally; AlCl3 group received daily intraperitoneal (i.p.) injection of AlCl3 for 60 days to induce neurotoxicity and AlCl3 + ACE200 group received a combination of AlCl3 and ACE in the same dose and route as previous groups. Aluminum administration significantly enhanced the memory impairment and the Aβ formation in the rat model. The cerebellum exhibited a significant reduced number of Purkinje cells, marked decrease in the density of dendritic arborization and prominent perineuronal spaces in the molecular layer. There was loss of dendritic spines, neurofibrillary degeneration, and appearance of neuritic plaques. Concomitant administration of AlCl3 and ACE displayed an observable protection against these changes with progressive improvement in memory and intellectual performance. In conclusion, ACE may play a protective role against formation of amyloid-β plaques in cerebellum.

SUMMARY: Knee osteoarthritis (OA) is a common disabling disease. Epidemiological studies have revealed various risk
factors for OA, including sex, aging, obesity, occupational illnesses, and chronic diseases. Here we evaluate the clinical, pathological,
and radiological findings of knee OA in a subset of Saudi patients who were subjected to total knee replacement (TKA). The study
population included 30 Saudi patients with knee OA who were operated by TKA (from June 2014 to December 2015) in the Department
of Orthopedics, Faculty of Medicine, King Abdulaziz University, Saudi Arabia. Patient’s clinical and radiological data were collected
from the hospital files. Pathological examination of the excised superior articular surface of tibia and femoral condyles were done.
Pearson Chi-squared analysis was used to test for differences between the variables in associated risk factors. There were more women
than men. Sixty per cent of patients were older than 60 years [mean age, 59.2 (females) and 61.7 (men) years-old]. All patients exceeded
obesity class 1, with females being more obese than males. Pathological examination of the superior articular surface of tibia and femoral
condyles showed high score lesions, which was more apparent in females than in males. Radiological findings showed that most lesions
were high grade. The findings of this study will help to understand the pathogenesis of OA and improve treatment decision making
relevant to TKA in knee OA in Saudi Arabia and elsewhere.

SUMMARY: The toxic effects of different doses of diclofenac sodium (DS) on the kidney on the postnatal period (0-7 days) by
morphometrical and immunohistochemical methods were investigated. For this purpose, 15 female adult wistar albino rats were used and
divided into 5 main groups. Group Ia served as normal control, physiologic group Ib received normal saline, group II received low dose (3.9
mg/kg), group III received medium dose (9 mg/kg) and group IV received high dose (18 mg/kg). Male offspring’s from 0-7 days after birth
were used in this study. On the 8th day of postnatal life, all animals were anesthetized. Then, the kidney samples were analyzed. Haematoxylin
and eosin staining showed degeneration and necrosis, apparent atrophy of the glomeruli, mononuclear cell infiltration, congested vessels,
increased fibrous tissue and distortion of the proximal convoluted tubules with interruption of the brush margin of the DS treated group.
Increased level of Caspase-3 and upregulation of TNF-α with different doses of DS. In light of our findings, DS may lead to adverse effects
that are dose-dependent in the prenatal subjected kidney to this drug.

ABSTRACT
Lead (Pb) toxicity is known to be a chief environmental health issue, especially for pregnant
women and young children. Today, the use of medicinal herbs in the treatment of many diseases
and different toxic agents has become highly accepted due to their effectiveness and lower costs.
Thymoquinone (TQ), which is extracted from Nigella sativa seeds, is a potent antioxidant and anti
inflammatory agent. This study was designed to explore the optional protectivity of TQ against
maternal and fetal oxidative stress and brain damage induced by Pb administration. Pregnant
rats were distributed into seven groups: control group, TQ group, DMSO group, two groups Pb
treated (160 and 320 ppm), and two groups Pb-treated (160 and 320 ppm) co-treated with TQ.
Administration started from gestation day 1 (GD1) to day 20 (GD20) through oral gavage once
daily. Lead administration caused a dose-dependent toxicity for both mothers and fetuses. Also,
the histopathological assessment of the brains from Pb-treated groups showed marked altera
tions. Co-treatment of with TQ and Pb caused a significant decrease in Pb levels as compared
with those treated with Pb alone and amelioration of histopathological changes in the brains. It
was concluded that co-treatment of TQ along with gestational Pb exposure could mitigate the
effects against Pb-induced maternal and fetal neurotoxicity.

This study was designed to address the protective effects of Zingiber officinale on the toxic outcomes of prenatal Cadmium administration on pregnancy outcome. Pregnant female Sprague-Dawley rats were randomly divided into four groups (eight rats/each), control group received distilled water, 2nd group treated with 8.8 mg of CdCl2/kg b. wt, 3rd group treated with 250 mg of Zingiber officinale/kg b. wt, and 4th group treated with 250 mg of Zingiber officinale/kg b. wt, followed by 8.8 mg of CdCl2/kg b.wt. Daily body weight of pregnant was recorded from GD1-GD20, and then pregnant rats were sacrificed at GD20. Samples of maternal and fetal livers and kidneys were processed for histological examination. Administration of Cd to pregnant rats showed adverse effects on pregnant mothers and their fetuses; reduced maternal weight gain, reduced absolute organ weights, reduced fetal growth parameters and placental weights together with altered histological appearance of the maternal and fetal livers and kidneys. While co-administration of Zingiber officinale showed an improvement of these toxic alterations. Zingiber officinale through its antioxidant activity could be beneficial against toxic outcomes of Cd exposure during pregnancy.

Background
Short morning exposure to high illuminance visible electromagnetic radiations termed as artificial daylight is beneficial for the mental health of people living in geographical areas with important seasonal changes in daylight illuminance. However, the commercial success of high illuminance light sources has raised the question of the safety of long hour exposure.
Methods
We have investigated the effect of the replacement of natural daylight by artificial daylight in Swiss mice raised under natural lighting conditions. Mice were monitored for neurotoxicity and general health changes. They were submitted to a battery of conventional tests for mood, motor and cognitive functions’ assessment on exposure day (ED) 14 and ED20. Following sacrifice on ED21 due to marked signs of neurotoxicity, the expression of markers of inflammation and apoptosis was assessed in the entorhinal cortex and neurons were estimated in the hippocampal formation.
Results
Signs of severe cognitive and motor impairments, mood disorders, and hepatotoxicity were observed in animals exposed to artificial daylight on ED20, unlike on ED14 and unlike groups exposed to natural daylight or conventional lighting. Activated microglia and astrocytes were observed in the entorhinal cortex, as well as dead and dying neurons. Neuronal counts revealed massive neuronal loss in the hippocampal formation.
Conclusions
These results suggest that long hour exposure to high illuminance visible electromagnetic radiations induced severe alterations in brain function and general health in mice partly mediated by damages to the neocortex-entorhinal cortex-hippocampus axis. These findings raise caution over long hour use of high illuminance artificial light.

Renal ischemia/reperfusion injury (IRI) represents the main reason for acute kidney injury (AKI). Dexmedetomidine (Dex) and Benincasa cerifera (BC) have wide benefits due to their anti-inflammatory and antioxidant properties. This study aims to illustrate the protective effects of BC and Dex on renal IRI in a diabetic model. Sixty adult male albino rats (Wistar strain), weighing 250–300 g, were included in the study. The rats were divided into four groups, as follows: sham group: (non-diabetic); diabetes mellitus (DM) + IRI group: streptozotocin (STZ)-induced diabetic rats exposed to renal IRI on day 30 after diagnosis of diabetes; DM + IRI + BC group: STZ-induced diabetic rats treated with BC (500 mg/kg) for 30 days after diagnosis of diabetes, then exposed to renal IRI; and DM + IRI + Dex group: STZ-induced diabetic rats treated with Dex (100 µg/kg intraperitoneally) 5 min before induction of ischemia on day 30 after diagnosis of diabetes, then exposed to renal IRI. Biochemical parameters, histopathological examination, and immunohistochemical markers were evaluated. A significant improvement in the biochemical, histopathological, and immunohistochemical parameters were observed in the DM + IRI + BC group, while the DM + IRI + Dex group showed improvements in renal IRI and dyslipidemia. The present study demonstrated that oxidative stress plays a chief role in renal IRI in the STZ-induced diabetic model. Treatment with BC achieved excellent ameliorative effects, while treatment with DEX improved renal IRI.

This study was planned to explore the protective role of curcumin (Cur) against maternal and fetal oxidative stress and cerebral damage induced by lead (Pb) during pregnancy. Positively pregnant female rats were divided into seven groups: control group, Cur group (300 mg/kg of Cur/b.wt.), DMSO group (50% DMSO), two Pb-treated groups (exposed to 160 and 320 mg/kg b.wt./day of Pb acetate, respectively), and two groups treated with both Pb and Cur (exposed to Pb as previous groups together with 300 mg/kg b.wt./day of Cur). Treatments through oral gavage once a day started from gestation day 1 (GD1) till day 20 (GD20), where the mother rats of different experimental groups were sacrificed to obtain the fetuses. Different chemical parameters were assessed. Brain specimens of mother and fetal groups were processed with examination. The results displayed that Pb administration to pregnant rats resulted in a dose-dependent toxicity for both mothers and fetuses. Also, there was a significant rise in lipid peroxidation and decreased antioxidant enzyme activities in the brains of the different Pb-treated groups. The histological examination of the brain of treated dams and fetuses showed marked alterations. Co-treatment of Cur along with Pb caused a significant decrease in Pb levels as compared with those treated with Pb alone, improving the oxidative condition with amelioration of the brain’s histopathological changes. Co-administration of Cur could have ameliorative effect against Pb-induced neurotoxicity through the reduction of oxidative stress and reversal of histopathological changes.

RESULTS: Overall life span (LS) was 2252.1±1742.5 days and cumulative 5-year survival (5YS) reached 73.2%, 10 years – 64.8%, 20 years – 42.5%. 513 LCP lived more than 5 years (LS=3124.6±1525.6 days), 148 LCP – more than 10 years (LS=5054.4±1504.1 days).199 LCP died because of LC (LS=562.7±374.5 days). 5YS of LCP after bi/lobectomies was significantly superior in comparison with LCP after pneumonectomies (78.1% vs.63.7%, P=0.00001 by log-rank test). AT significantly improved 5YS (66.3% vs. 34.8%) (P=0.00000 by log-rank test) only for LCP with N1-2. Cox modeling displayed that 5YS of LCP significantly depended on: phase transition (PT) early-invasive LC in terms of synergetics, PT N0—N12, cell ratio factors (ratio between cancer cells- CC and blood cells subpopulations), G1-3, histology, glucose, AT, blood cell circuit, prothrombin index, heparin tolerance, recalcification time (P=0.000-0.038). Neural networks, genetic algorithm selection and bootstrap simulation revealed relationships between 5YS and PT early-invasive LC (rank=1), PT N0—N12 (rank=2), thrombocytes/CC (3), erythrocytes/CC (4), eosinophils/CC (5), healthy cells/CC (6), lymphocytes/CC (7), segmented neutrophils/CC (8), stick neutrophils/CC (9), monocytes/CC (10); leucocytes/CC (11). Correct prediction of 5YS was 100% by neural networks computing (area under ROC curve=1.0; error=0.0).

haemorrhages and necrosis, appear to be involved
in the histopathological mechanism of these lesions
[22]. Moreover, it is well known that prolonged Cd
exposure leads to carcinogenic effects on the male
reproductive organs [29] and leads to a reduction in
reproductive capabilities [48].
Vitamin E is the collective name for a band of fat-
soluble substances with unique antioxidant activities
that is available in 8 chemical forms (a-, b-, g- and
d-tocopherol, a-, b-, g- and d-tocotrienol) that have
diverse degrees of biologic activity [12]. The chemi-
cal form a-tocopherol is the most recognised and
found in many food items, including nuts, seeds and
vegetable oils [21]. The body forms reactive oxygen
species (ROS) endogenously which comprise both free
radical and non-free radical oxygen such as hydrogen
peroxide (H2O2), superoxide (O2-) and hydroxyl radical
(OH) [57]. Alpha-tocopherol causes the elimination of
the free oxygen radicals and the inhibition of the per-
oxidation [19]. It acts by eliminating lipid peroxyl and
alkoxyl radicals, so it suppresses the chain reaction
of lipid peroxidation and promotes the production
of scavenger antioxidant enzymes [18].
Numerous reports in several labs are going on to
evaluate the preventive influence of different natural
substances toward poisonous metals [24]. Fenugreek
seed (Trigonella foenum-graecum L. cultivar Baladi,
family: Leguminosae) is herb of the Legume family
that is broadly grown in Egypt and India. Used with
meal as a spice and in medication where it demon-
strates antioxidant impact in diabetes mellitus due to
the existence of various ingredients [20, 53].
The aim of this study is to evaluate the cytopro-
tective effects of fenugreek and a-tocopherol on Cd
chloride induced testicular damage.
MATERIALS AND METHODS
Ethical approval. This study was conducted after
receiving the approval of the Medical Research Eth-
ics Committee, Faculty of Medicine, King Abdulaziz
University.
Animals. Thirty male adult Wistar rats aged 120
days that weighed 175–200 g and were obtained
from the animal house were weighed and randomly
placed into three groups (n = 10). The rats were
housed individually in stainless steel cages with con-
trolled temperature (22 ± 2°C) and humidity (55 ±
10%) and 12/12 h cycle of light and darkness with
access to food and drinking water ad libitum. The
experimental procedures were performed in accor-
dance with the international guidelines for the care
and the use of animals in a laboratory.
Chemicals. Cadmium chloride (CdCl2, 99% pure)
and a-tocopherol were purchased from Sigma-Aldrich
Chemicals Co. (St. Louis, MO, USA).
Fenugreek powder preparation. Fenugreek (Trig-
onella foenum-graecum L.) was purchased from local
herbal market. Fifty grams of fenugreek-dried seeds
were crushed, boiled in 1 L of distilled water for half an
hour. Then the extracts were filtered. The powder was
mixed at 5% w/w in standard rat pelleted diet (i.e. 5 g
of dry fenugreek in 95 g of rat food). The dose used in
the current work was selected according to a previous
research [4]. The chemical constituents of fenugreek
are carbohydrate (49.81 g%), protein (27.31 g%), fat
(7.11 g%), ash (3.67 g%), steroidal saponins (2 g%),
flavonoids (399.67 mg%), trigonelline alkaloid (380
mg%) and polyphenols (251.71 mg%) [4, 26].
Experimental design. Group I received 1 mL of
normal saline and served as the control group. Group
II received 5 mg/kg/day CdCl2 (1/15 LD50) [43]. Group
III received 5 mg/kg/day CdCl2 and 150 mg/kg/day
fenugreek and 100 mg/kg/day of a-tocopherol [6].
The prophylactic combination started 1 week before
CdCl2 administration in the current study to acquire
the benefits of both curative and preventive regi-
mens. All groups were treated by oral gavage for 60
consecutive days.
Testis histology. At the end of the experiment,
the animals were anesthetised, weighed and sac-
rificed by decapitation. The testes were removed
and weighed. Subsequently the testes were fixed in
10% neutral buffered formalin, processed for 5-µm
paraffin sections and stained with haematoxylin and
eosin (H&E). For each specimen, at least 3 to 5 slides
were examined using an Olympus BX53 microscope
equipped with a DP73 digital camera (Olympus, To-
kyo, Japan) [34, 38, 54].
Immunohistochemistry. Paraffin sections were
de-waxed in xylene, rehydrated and pretreated with
3% of hydrogen peroxide to block endogenous pero-
xidase activity. Microwave-assisted antigen retrieval
was performed for 20 min. Slides were then incu-
bated overnight at 4oC with the primary antibody
(Anti CD68+ for macrophages [1:50], Anti CD3+ for
T-lymphocytes [1:200] and Anti CD20+ for B-lymp-
hocytes [1:300]) (Rabbit Polyclonal Antibody from
Neo-Markers, Lab Vision Corporation) and sections
were incubated with biotinylated IgG and then with
streptavidin-peroxidase conjugate (Zymed Corp.). Sec-

tions were then washed with phosphate-buffered sa-
line (PBS) and incubated with 3-3’ diaminobenzidine
tetrachloride (DAB) for 5 min to detect immunoreacti-
vity. Sections were counterstained with haematoxylin.
Negative control sections were prepared by omitting
the primary antibody. Positive control standard labo-
ratory slides were used for all stains to validate the
success of the technique. All slides were examined
under light microscopy and the presence of labelled
cells was documented. The absence of staining was
recognised as a negative result (−) while the presence
of brown staining was recognised as a positive result
(+) [54].
Morphometry. Five slides of the testes of each
group (6 fields per slide), seminiferous tubules round
or almost round were selected indiscriminately and
analysed. The mean seminiferous tubule diameter was
obtained by measuring across the minor and major
axes. The seminiferous epithelium height was measu-
red for the same tubules. The epithelium height was
calculated as the space between the tunica propria
and the edge of the lumen and two diametrically
opposed readings were taken with a digital ruler
on each cross section using their mean value [33].
The measurements were taken at magnifications of
×100 and ×200 using the Image-Pro Plus v6.0 (Media
Cybernetics, Maryland, USA) and an Olympus BX53
microscope. In addition, the percentage of the immu-
nopositive cells in the testicular tissue was evaluated.
Ultrastructure study. Testes samples of approxi-
mately 1 mm3 were obtained and immersed in 2.5%
of glutaraldehyde in a 0.1 M buffer phosphate at 4°C
for 3 h and post-fixed in 1% of osmium tetraoxide.
After dehydration in a graded series of ascending con-
centrations of ethanol, the tissues were embedded
in epon 812. Semithin sections were prepared from
the blocks, stained using toluidine blue and observed
with the Olympus microscope. Demonstrative areas of
semithin sections were chosen. Ultrathin sections of a
50–60 nm of thickness were cut using ultramicroto-
me (NOVA, LKB 2188, Bromma, Sweden) and uranyl
acetate and lead citrate were used to stain the tissues
and then inspected using a Philips 201 transmission
electron microscope at 60–80 kV in a transmission
electron microscope unit (Philips Industries, Eindho-
ven, Netherlands) [37].
Statistical analysis. Quantitative data were ex-
pressed as the mean ± standard deviation of dif-
ferent parameters for the treated groups. The data
were analysed using the one way analysis of varian-
ce (ANOVA) after testing Levene’s test verified the
equality of variances in the samples (homogeneity
of variance) (p > 0.05). In the parametric data which
equality was assumed we used Tukey’s post-hoc test
for comparison between variable, meanwhile in pa-
rametric parameters in which equality of data were
not assumed (CD3, CD20, CD68) we use Dunnett’s T3
test for comparison between groups. The statistical
analysis was performed using SPSS version 22. The
values were considered significant when p < 0.05.
RESULTS
Body and testes weight. Rats received CdCl2
revealed a significant reduction in the body wei-
ght and testis weight in comparison with control
group. Administration of combined fenugreek and
a-tocopherol with CdCl2 improved body weight and
testes weight (Table 1).
The histological evaluation of the testes of the
control group shows the normal architecture of the
seminiferous tubules (Fig. 1A). CdCl2 treated group
showed disintegration, disorganisation and the loss
of the typical cytoarchitecture of the tubules (oval
or circular) with a separation of the spermatogonia
from the basal lamina, a widening of the interstitial
spaces and congestion of the capillaries. Some tubu-
les showed vacuoles of different sizes inside and in
between the spermatogenic cells, spermatogenic cells

sloughed, detached and lost the support of Sertoli
cells so they could no longer survive and there was
appearance of giant cells with multiple nuclei (Fig. 1B
C). Combined fenugreek and αa-tocopherol with CdCl2
minimised the degenerative changes evidenced by the
reappearance of the spermatogonia near the basal
lamina as well as the restoration of the structure of
the seminiferous tubules and no cavities or vacuoles
were detected nearly in all exposed animals (Fig. 1D).
Toluidine blue stained sections. The testes of
the control group reveal the typical histological pic-
ture of the seminiferous tubules (Fig. 2A). The group
treated with CdCl2 showed disorganisation of the
seminiferous tubules with an obvious diminution of
the epithelial layers, extensive degenerative vacuolisa-
tion and intercellular empty spaces in between the
cells. Also, the spermatozoa disappeared, the sper-
matids exfoliated, the primary spermatocytes’ density
was diminished and the spermatogonia noticeably
deteriorated and depleted (Fig. 2B, C). Combined
fenugreek andαa-tocopherol with CdCl2 showed an
apparent improvement of the architecture of the
seminiferous tubules with a partial restoration of a
low germinal epithelium more or less in all exposed
animals and minimal or no vacuolations among the
germ cells (Fig. 2D, E).
Morphometric analysis. The mean seminiferous
tubular diameter showed variations in the different
groups in comparison with the control group. The data
showed that there was a significant decrease in the
mean diameter of the seminiferous tubules in the CdCl2
treated group in comparison with the control group,
however there was an improvement in the diameter
of the seminiferous tubules of the group treated with
combined fenugreek and αa-tocopherol and CdCl2. The
mean germin:al epithelial height of the CdCl2 treated
group showed a significant decrease in comparison
with the control group. The group treated with CdCl2
and combined fenugreek andαa-tocopherol showed sig-
nificant improvement in the epithelial heights (Table 2).
Immunohistochemistry study. The data showed
that there were significant increases in the area per-
centage of CD3, CD20 and CD68 in the CdCl2 treated
group in comparison with the control group (Table 3).
Ultrastructure picture. The transmission electron
micrographs of the control group reveal a normal
structural organisation of the seminiferous tubules
and the interstitium. The CdCl2-treated testes re-
vealed several vacuoles of variable sizes inside and in
between the germ cells and an apparently thickened
basal lamina with a myoid cell. Sertoli cells with an
irregular euchromatic nucleus and the characteris-
tic indentation of the nucleus resting on the basal
lamina with numerous vacuoles and an increase in the
density of the cytoplasmic inclusions, also unhealthy
damaged cells were observed. The primary spermato-

swollen mitochondrial sheaths surrounds the central
axoneme. Spermatids with rounded or oval nuclei that
have disturbed chromatin and peripherally arranged
cytes with disrupted chromatin are separated by the
variable sizes of the intercellular spaces and the vacu-
olated distorted mitochondria. In addition, several
vacuoles of variable sizes all over the cytoplasm were

the mitochondria with lost cristae and acrosomal cap.
Leydig cells with oval, indented nuclei with disrupted
chromatin are noticed and apparent electron dense
bodies, vacuoles, empty spaces, lymph vessels and
ruptured vacuolated mitochondria. Unhealthy cells
characterised by heterochromatin condensation and
shrinkage of both cytoplasm and nucleus, which is
an indication of phagocytosis (Fig. 3A–D).
While the recovery group showed minimal vacu-
olation, some mitochondria were still affected and
others are healthy mid pieces of the sperm were
nearly improved and restored nearly in all exposed
animals. Reconstruction of the structure of the cross-
sections of the mid pieces with axoneme surrounded
by a mitochondrial sheath is noticed. Leydig cells
with oval euchromatic nuclei and peripherally located
heterochromatin. The cytoplasm contains multiple
lysosomes. Improvement of the appearance of germ
cells is observed. The Spermatids with rounded or
oval euchromatic nuclei and peripherally arranged
mitochondria with preserved cristae and acrosomal
cap were observed. Minimal empty spaces in the af-
fected intercellular bridge are still noticed (Fig. 4A–D).
DISCUSSION
Cadmium is a poisonous heavy metal that is attri-
buted to it reduced elimination rate [51]. Male genital
organs are obviously affected during environmental
exposure to Cd as proved by bad quality of seminal
fluid and sterility [7]. In the current study, it is ob-
served that administration of CdCl2 lowered testicular
weight and body weight and this is related to reduced
androgen level [52]. Moreover, CdCl2 decreases total
sperm count and raises anomalous sperm forms [35]
and induces necrotic destruction of testicular tissues
[25]. In addition, the undifferentiated spermatogenic
cell count is declined [40].
With light microscopic examination, CdCl2 treated
group showed degenerative alterations in the testi-
cular histology with damage of the supporting cells
of Sertoli that ultimately causes irreversible loss and
detachments of spermatogonia and this agreed with

a previous research [58]. Morel et al. [36] found that
Leydig cells’ morphology is altered with disrupted
chromatin followed by reduction in the cell number
and is irreversibly substituted by inflammatory cells
and this agreed with the current findings that are
supported by the immunomarkers increased expres-
sion. It is noted that fenugreek consumption restored
serum testosterone level and testicular weight [45].
The fenugreek being an antioxidant, it performs a cru-
cial role in restoring testosterone level by eliminating
ROS which is known to stop steroidogenesis [45, 60].
The cell type that is damaged is mainly in the early
stage of spermatogenesis and the degree of germ
cell damage is related to the dose of Cd exposure [9].
This is consistent with the current findings that the
spermatogonia are the primary cells that are affected.
In contrast, another researcher observed that the
exfoliated sloughed germ cells were spermatocytes
and spermatids [44].
Administration of Cd in the present study pro-
duced atrophy, vacuolations, cellular debris and the
seminiferous epithelium was sloughed off at many
points. These changes are consistent with the findings
of the author who demonstrated that a single dose
of Cd induced severe necrosis and degeneration of
the seminiferous tubules with a complete loss of the
centrally located spermatozoa [2].
In the current research, a wide interstitium and
congested capillaries were noticed. Burukoğlu and
Bayçu [10] observed several congested thickened
blood vessels and homogeneous oedematous ma-
terial in the interstitium which is infiltrated by diffe-
rent cells such as leukocytes and macrophages. The
thickened blood vessel wall and oedema are due to
cytokines secreted by macrophages situated in the
peritubular zones [1, 44].
Ultrastructurally, the changes of the thickness of
basal lamina observed in the present research could
be explained by the study of Shokri et al. [50] who
found that certain stimulants might encourage the
myoid cells to yield more collagen and extracellular
matrix, which cause the thickening of the basal lamina.

In the current work, numerous intercellular areas
were noticed in-between the germinal cells lining the
seminiferous tubules. This is attributed to loosening
of cell–cell connections and these gaps caused shrin-
kage of both germ and Sertoli cells [14]. A previous
study observed disarrangement of germ cell structure
and function that is due to elevated level of ROS in
the membrane lipids [16, 56].
In CdCl2 treated group, Sertoli cells showed irregu-
lar euchromatic nuclei with variable damage of germ
cells [23] and this agreed with the current findings.
The present study revealed ultrastructural alteration
of sperms as shown in the transverse sections of the
mid-pieces of sperm that revealed disrupted axoneme
and a distorted swollen mitochondrial sheath.
The mechanism of Cd induced injury involves the
production of free radicals, which modify mitochon-
drial activity [13]. Therefore, other authors have po-
stulated that antioxidants could be a critical aspect of
the efficient management of Cd intoxication [11, 16].
Current work revealed that fenugreek admini-
stration reduces the unhealthy consequences of Cd
on testicular tissues that are attributed to enhan-
cement of antioxidant potential due to minimising
lipid peroxidation [28]. Moreover, an experimental
research demonstrated that consumption of herbal
substance that contains polyphenols and flavonoids
exerts antioxidant role and suppresses low density
lipoprotein oxidation [55].
Some researchers have discussed the immunohi-
stochemistry of the testicular architecture and they
observed that macrophages and lymphocytes were
limited around the basal lamina and in the interstitial
spaces [32]. Other investigator observed that immu-
ne response infiltration of monoclonal antibodies for
B- and T-lymphocytes and macrophages occasionally
occurred in the lumen of the testes [26]. This is in
accordance with our findings that CD68 immunore-
activity (macrophages) showed significant increase
that is improved with the administration of combined
fenugreek and a-tocopherol. In the current study, CD20
showed significant increase in immunohistochemical
positivity with Cd and this distinguishes cell injury,
degeneration, inflammatory conditions and death of
components of the testes [31]. A researcher noticed
that crucial aetiological element responsible for male
infertility is infection and inflammation of the epithelial
tissue and suggested that T-lymphocytes participate
in the regulation of male infertility pathogenesis [15].
A previous research suggests that fenugreek
can be regarded a potential therapeutic nutrient to
guard against male reproductive toxicity caused by
Cd through its antioxidant, anti-inflammatory and
antifibrotic actions [5].
A study using a-tocopherol and Cd showed the
seminiferous tubules and the Leydig cells almost re-
tained their normal structure with normally arranged
the seminiferous epithelium and a healthy intersti-
tium [41]. Most of the mid-pieces of the sperm had
a normal structure except a few of them showed
distorted and swollen mitochondrial sheaths [41] and
this agreed with the current findings.
Alpha-tocopherol has been found to minimise
infections [8] and activate the adaptive immune sys-
tem which was detected by the expression of B-
lymphocytes and T-lymphocytic surface antigens [36].
The antioxidant a-tocopherol has also been found to
be capable of diminishing apoptosis in degenerating
cells of the testicular tissues [42]. Therefore, the role
of a-tocopherol can be defined as a defensive role
for the testicular epithelium and lymphocytic system.
Our findings revealed that combined fenugreek
and a-tocopherol with CdCl2 decreases earlier struc-
tural alterations and reduces the injury and this co-
incides with the findings of a prior research [58].
The ameliorating effects of combined fenugreek and
a-tocopherol in the current work were slight and
these results are consistent with the progression of
alterations in the rat testes as a result of Cd intoxi-
cation explained by Sen Gupta et al. [47]. Alpha-
tocopherol supplement has demonstrated fruitful
effect in enhancing sperm quality [3]. This agreed
with our suggestion that supplements of combined
fenugreek and a-tocopherol alleviates the oxidative
stress. This regimen could re-establish the activity
of antioxidant enzymes to allow regular germ cell
differentiation and guard sperm count and protect
testicular morphology throughout Cd exposure.
Limitation of the study
The limitations of the study are due to the small
number of animals so, we advise to apply this study
on a larger number of animals and different spices to
facilitate the statistical analysis and to broaden the
feedback. Also, we advise to apply different doses
of the combination of fenugreek and a-tocopherol.
Quantitative measures as stereology are recommend-
ed to support the hypothesis of the study.

CONCLUSIONS
The results obtained from the current study re-
veal that fenugreek and a-tocopherol ameliorate
histopathological variations of the testicular tissues
created by Cd to some extent. These combined sub-
stances might be used as a dietary consumption of
humans who are exposed to environmental toxins
especially Cd.
